(author for correspondence; itiryaki@ksu.edu.tr) Received 05 March 2007; Accepted 11 December 2007 ___________________________________________________________________________________ Abstract Response of auxin resistant mutant, axr1-24, on salt stress in primary root growth was tested over a range of NaCl concentrations and results were compared with wild type. The axr1-24 was more resistant to the inhibitory effect of any salt concentrations tested than wild type, except 128 mM NaCl. Total inhibition of root elongation was 62% and 70% for axr1-24 and wild type, respectively, at 64 mM NaCl. Exogenous application of salicylic acid (SA) or 1-aminocyclopropane-1-carboxylic acid resulted in a significant increase in jasmonic acid (JA)-inducible allene oxide synthase (AOS) gene expression, while induction was severely reduced on axr1-24 seedlings treated with SA after 24 h. The results of this study provided a link between auxin and salt stress as well as SA and JA signaling through AXR1. Key Words: Hormone signaling, salt stress, auxin, jasmonic acid, axr1. Arabidosis thaliana’da oksin sinyal geni axr1in tuz stresi ve jasmonik asit ile uyarılabilen gen ekspresyonundaki rolü Özet Research Article 189 Oksin duyarsız mutant axr1-24’ün tuz stresine karşı olan tepkisi farklı konsantrasyonlarda NaCl içeren kök büyüme ortamında test edilerek sonuçlar yabani form olan kontrol bitkileri ile karşılaştırılmıştır. Sonuçlar axr1-24’ün 128 mM NaCl tuz konsantrasyonu hariç kök büyümesini engelleyen diğer NaCl konsantrasyonlarına karşı çok daha dayanıklı olduğunu göstermiştir. 64 mM NaCl içeren kök büyüme ortamında, kök büyüme uzunluğunda meydana gelen toplam engellenme oranı axr1-24 ve yabani form için sırasıyla %62 ve %70 seviyesinde olmuştur. Dışarıdan uygulanan salisilik asit (SA) ve 1-aminosiklopropan- 1-karboksilik asit, jasmonic asit (JA) ile uyarılabilen allene oksit sentez (AOS) enzimini kodlayan genin ifadesinde önemli derecede artışa neden olurken, axr1-24 fidelerinde SA muamelesinden 24 saat sonra büyük bir azalma meydana gelmiştir. Bu çalışma sonuçları oksin ile tuz stresi ve SA ile JA sinyalilizasyonları arasında AXR1 aracılığı ile meydana gelen bir ilişkinin olduğunu ortaya koymaktadır.
