Lightning-Link <strong>kits</strong><br />
The Lightning-Link kit allows you to label an antibody with minimal hands-on time - less than 30<br />
seconds. Simply add your antibody to a tube of Lightning-Link mix and the label becomes covalently<br />
attached to your antibody in a gentle controlled process.<br />
As Lightning-Link is a one-step antibody <strong>labeling</strong> procedure (i.e. is not interrupted by desalting or<br />
dialysis steps), your antibody can be prepared with only microgram quantities of antibody, with recoveries<br />
approaching 100%. Furthermore, the technology is easily scalable (μg to mg) ensuring quality<br />
and consistency. Thus Lightning-Link provides a simple antibody <strong>labeling</strong> method which saves both<br />

<strong>Antibody</strong><br />
Does the antibody have to be purified?<br />
Yes – purified antibodies are perfect for Lightning-Link. Please note<br />
common additives present in buffer formulations such as BSA (up to<br />
1%) and gelatin (up to 0.5%) are tolerated - please see our website for<br />
further technical information. Antibodies which are present in ascites<br />
fluid, serum or hybridoma culture media need to be purified.<br />
What is the ideal starting concentration of the antibody?<br />
<strong>Antibody</strong> concentration should be in the range of 0.5-2.5mg/ml.<br />
Is the Lightning-Link antibody <strong>labeling</strong> chemistry isotype or species<br />
specific? No.<br />
Methodology<br />
Is the hands-on time really only 30 seconds?<br />
Yes – naturally you will need to familiarize yourself with the<br />
instructions before using Lightning-Link products for the first time,<br />
but after that your hands-on time should be no more than 30 seconds.<br />
How scalable is the technology?<br />
Scalability is one of the major advantages of Lightning-Link. Rather<br />
than risk valuable antibody you can label a small quantity and be<br />
confident that the bulk quantity made later will have essentially<br />
identical properties.<br />

<strong>Antibody</strong>-Label Optimization (Micro-optimizer )<br />
Optimization of labeled antibodies at the microgram scale. Use our<br />
proprietary technologies to enhance the performance of your labeled<br />
antibodies. By creating an array of trial conjugates (5 formulations per<br />
label) at low microgram scale (20μg per formulation) combined with<br />
selection by you in the assay of interest, the best possible conjugate<br />

What functional groups do I need on my antibody to use<br />
Lightning-Link?<br />
Lightning-Link requires amine groups on the molecule to be labeled.<br />
Most proteins or peptides have lysine and/or alpha-amino groups. All<br />
antibodies have multiple amine functions.<br />
Can proteins other than antibodies be labeled?<br />
Yes – while <strong>labeling</strong> of primary antibodies is a major application, the<br />
only requirement is that the protein or molecule to be labeled has<br />
amine functionality.<br />
Is the label attached to my protein by a covalent bond?<br />
Yes – conjugates formed with Lightning-Link are perfectly stable.<br />
Will my antibody couple to itself or form high molecular weight<br />
aggregates?<br />
No – the antibody <strong>labeling</strong> chemistry is unidirectional and therefore<br />
<strong>labeling</strong> will only occur between the antibody and the label.<br />
<strong>Antibody</strong> Buffers<br />
What buffer additives can be used and what should be avoided?<br />
Additives such as salts (e.g NaCl), sugars (e.g. sucrose) and<br />
chelators (e.g. EDTA) have no effect on the antibody <strong>labeling</strong><br />
reaction. The main additives to avoid are amino acids (e.g. glycine),<br />
blockers (e.g. ethanolamine), tris buffer above 20mM and thiols<br />
(DTT, mercaptoethanol).<br />
