KMUTT Annual Research Abstracts 2005 insoluble polysaccharides in plant cell walls. The anaerobic, facultative, mesophilic, xylanolytic and cellulolytic bacterium, Bacillus circulans B-6, produces the xylanolytic complexes containing a variety of xylanolytic and cellulolytic enzymes when grown on xylan or avicel (microcrystalline cellulose) under the aerobic condition. When grown on xylan, the crude enzyme consisted of 33 proteins with 16 types of xylanases and 9 types of CMCases while the crude enzyme from avicel culture contained 20 proteins with 16 types of CMCases and 12 types of xylanases. Although the subunit compositions of the crude enzymes from xylan culture and avicel culture were different, depending on the different carbon sources, the bacterial cells and their crude enzymes from both cultures were able to bind to insoluble xylan and avicel and hydrolyze the insoluble substrates. We isolated the xylanosomes from the cell surface which had multi-xylanases and - CMCases, ß-xylosidase, arabinofuranosidase, acetyl esterase, carboxymethyl cellulases and avicelase and also xylan-binding and cellulosebinding abilities. They could efficiently hydrolyze insoluble xylan, avicel and insoluble polysaccharides in plant cell walls completely to xylose and glucose as the end products. Moreover, these xylanosomes and the crude enzyme had an advantage of having their activities and stabilities at the broad pH profile between pH 4.0 and 9.0. Thus, they are the great potential for the application in degradation of insoluble polysaccharides in plant cell walls. IC-289 COMPARISON OF DIFFERENT EXTRACTION BUFFERS FOR EXTRACTION OF CYANIDE-RELEASING ENZYME FROM CASSAVA ROOT Somphit Sornyotha, Khanok Ratanakhanokchai, Khin Lay Kyu The 1 st International Conference on Fermentation Technology for Value added Agricultural Products, March 22-25, 2005, Sofitel Raja Orchid Hotel, Khon Kaen, Thailand Cyanide-releasing enzyme or linamarase (EC.3.2.1.21) in cassava root was extracted from root parenchymal tissue and root peel with phosphate buffer containing 1% polyvinylpyrrolidone K30 (buffer A), phosphate buffer containing 1% polyvinylpyrrolidone K30 and 1% Triton X-100 (buffer B), phosphate buffer containing 1% polyvinylpyrrolidone K30 177 and 1% Tween 20 (buffer C) and phosphate buffer containing 1% polyvinylpyrrolidone K30 and 1% Tween 80 (buffer D). The specific linamarase activity was found to be the highest in both extracts of tissues when extracted with buffer C, followed by extraction with buffer D, buffer B and buffer A. It was also found that when hydrophobicity of extraction buffer increased, the linamarase activity of the extract also increased. SDS-PAGE and native-PAGE analysis showed that the different protein patterns were observed depending on the extraction buffers used. The amount of proteins were found to be the highest when extracted both tissues with buffer D. The root peel extract contained at least 14 proteins on SDS-PAGE, whereas the extract of root parenchyma contained at least 19 proteins. However, the same major proteins were found in all extracts with different extraction buffers, indicating that 65 kDa was the major protein in extract of the root peel while 40 and 45 kDa were major proteins in the extract of the root parenchyma. IC-290 MULTIPLE CELLULASES AND XYLANASES FROM Bacillus circulans B6 DURING GROWTH ON AVICEL UNDER AN AEROBIC CONDITION Rattiya Waeonukul, Khin Lay Kyu, Khanok Ratanakhanokchai The 1 st International Conference on Fermentation Technology for Value added Agricultural Products, March 22-25, 2005, Sofitel Raja Orchid Hotel, Khon Kaen, Thailand When the facultative anaerobic bacterium, Bacillus circulans B6 was cultivated in mineral salt medium containing avicel as a sole source of carbon under an aerobic condition, the bacterium produced extracellular cellulolytic and xylanolytic enzymes such as avicelase, carboxymethylcellulase (CMCase), cellobiohydrolase, ß-glucosidase, xylanase and ß-xylosidase. The bacterium produced multiple cellulases and xylanases, which showed at least 12 types of CMCase and 11 types of xylanase on zymogram analysis. The bacterial cells were found to be capable of adhesion to avicel and insoluble xylan. Moreover the CMCase and xylanase enzymes could also bind to avicel at 93% and 85% and insoluble xylan at 41% and 60%, respectively. 178 IC-291 PURIFICATION AND BINDING PROPERTIES OF XYLAN-BINDING ENDOXYLANASE (48 KDA) FROM Bacillus circulans B6 Patthra Pason, Khanok Ratanakhanokchai, Khin Lay Kyu The 1 st International Conference on Fermentation Technology for Value added Agricultural Products, March 22-25, 2005, Sofitel Raja Orchid Hotel, Khon Kaen, Thailand When Bacillus circulans B6 was grown in a medium containing xylan as a carbon source, the bacterium produced multienzyme complex. The crude enzyme showed many protein bands on SDS-PAGE and at least 16 xylanases and 9 carboxymethylcellulases on zymograms. Purification of xylan-binding endoxylanase was conducted by an affinity adsorption-desorption on insoluble xylan. The molecular mass of purified xylan-binding endoxylanase was estimated at 48,000 dalton and it acted only on xylan. The enzyme was stable at pH 6.0-7.0 and temperature up to 50°C. The pH and temperature optima were 6.0 and 60°C. The enzymatic hydrolysis products of xylan were a series of short-chain xylooligosaccharides, indicating that the enzyme was an endoxylanase. The xylan-binding endoxylanase could be able to effectively hydrolyze insoluble xylans of birch wood, larch wood and oat spelt. Adsorption of enzyme to insoluble xylan and avicel were broad specific but it was affected by pH, sugar and metal ion. High pH and sugar decreased the adsorption of the purified enzyme to insoluble xylan and avicel. However, MgSO 4 , CaSO 4 , FeSO 4 and CuSO 4 increased the adsorption of the enzyme to insoluble xylan, while FeSO 4 and CuSO 4 increased the adsorption of the enzyme to avicel. The purified enzyme can be applied in hydrolysis of insoluble xylan present in agricultural wastes. Many e- Government information systems fail during the development due to their unclear requirements. This paper addresses the aspects of requirement elicitation from government agencies and related stakeholders. e-Government can be claimed as a strategic tool for supporting better government administration and public services.
