Looking for the causes of ageingNew research has thrown doubt onthe role of proteins called sirtuins inthe mechanism by which someorganisms’ lifespans have beenextended in the lab. The claimedtarget of some anti-ageing creams,sirtuins have been linked to ageingand longevity in yeast, nematodeworms and fruit flies. When theseorganisms’ genes were manipulatedto overproduce sirtuin proteins, theylived longer – up to 50 per cent longerin the case of nematodes.A paper published in Nature inSeptember 2011 describedexperiments led by Dr David Gems atthe Wellcome Trust-funded Instituteof Healthy Ageing, University CollegeLondon. This showed – surprisingly– that sirtuin genes were not thecause of the longer lifespans seenpreviously. Dr Gems suggestsscientific efforts should be redirectedtowards other genes that seem to playmore of a role in ageing.Blood pressure genesTwo studies published in Nature andNature Genetics in September 2011have identified several gene regionsassociated with blood pressure andother measurements that can predictcardiovascular disease. The findingsidentify potential targets for newtherapies to prevent heart diseaseand stroke.Both studies were from theInternational Consortium for BloodPressure Genome-Wide AssociationStudies and were funded by theWellcome Trust, the MedicalResearch Council, the British HeartFoundation and the NationalInstitute for Health Research. Theresearch involved hundreds ofscientists from 24 countries aroundthe world.More than one billion people havehigh blood pressure; even smallchanges in blood pressure canincrease risk of heart disease andstroke, the biggest cause of deathworldwide.Larynx transplantProfessor Martin Birchall at theRoyal National Throat, Nose andEar Hospital in London hasdeveloped pioneering techniquesthat are transforming transplantsof the larynx.In October 2010, he joined aninternational team of surgeons at theUniversity of California, Davis torestore the voice of a woman whoselarynx had been damaged duringsurgery. She had not spoken for 11years and could not breathe, eat ordrink unaided. The 18-hour operationwas first time that the larynx, tracheaand thyroid had been transplantedtogether. After two months ofrehabilitation, during which thenerves regenerated, the patientlearned to speak again and she cannow breathe, eat, drink and talkunaided.The surgical techniques ProfessorBirchall used in the operation weredeveloped over more than ten years ofresearch funded principally by theWellcome Trust.From left:The Children of the 90s study is now looking at severalgenerations of families.
