Aneks 1 – Pembuatan Produk SterilPENGAWASAN MUTU155. Uji sterilitas yang dilakukan terhadapproduk jadi hendaklah dianggap hanyasebagai bagian akhir dari rangkaiantindakan pengendalian untukmemastikan sterilitas dari produk. Ujisterilitas ini hendaklah divalidasi untukproduk yang berkaitan.156. Sampel yang diambil untuk pengujiansterilitas hendaklah mewakilikeseluruhan bets, tetapi secara khusushendaklah mencakup sampel yangdiambil dari bagian bets yang dianggappaling berisiko terhadap kontaminasi,misal:a) untuk produk yang diisi secaraaseptis, sampel hendaklahmencakup wadah yang diisi padaawal dan akhir proses pengisianbets serta setelah intervensi yangsignifikan; danb) untuk produk yang disterilisasicara panas dalam wadah akhir,sampel hendaklah diambil daribagian muatan dengan suhuterendah.157. Kepastian sterilitas dari produk jadidiperoleh melalui validasi siklussterilisasi untuk produk yang disterilisasiakhir, dan melalui “media fill” untukproduk yang diproses secara aseptis.Catatan pengolahan bets dan, dalam halproses aseptis, catatan mutulingkungan, hendaklah diperiksa sejalandengan hasil uji sterilitas. Prosedurpengujian sterilitas hendaklah divalidasiuntuk produk yang berkaitan. Metodefarmakope harus digunakan untukvalidasi dan kinerja pengujian sterilitas.Untuk produk injeksi, Air untuk Injeksi,produk antara dan produk jadi hendaklahdipantau terhadap endotoksin denganmenggunakan metode farmakope yangdiakui dan tervalidasi untuk tiap jenis produk.Untuk larutan infus-volume-besar,pemantauan air atau produk antarahendaklah selalu dilakukan sebagaipengujian tambahan terhadap pengujianyang dipersyaratkan dalam monografiQUALITY CONTROLAnnex 1 – Manufacture of SterilePharmaceutical Products155. The sterility test applied to the finishedproduct should only be regarded as thelast in a series of control measures bywhich sterility is assured. The testshould be validated for the product(s)concerned.156. Samples taken for sterility testing shouldbe representative of the whole of thebatch, but should in particular includesamples taken from parts of the batchconsidered to be most at risk ofcontamination, e.g.:a) for products which have been filledaseptically, samples should includecontainers filled at the beginningand end of the batch and after anysignificant intervention; andb) for products which have been heatsterilized in their final containers,consideration should be given totaking samples from the potentiallycoolest part of the load.157. The sterility of the finished product isensured by validation of sterilizationcycle in the case of terminally sterilizedproducts, and “media-fills” runs foraseptically processed products. Batchprocessing records and, in the case ofaseptic processing, environmentalquality records, should be examined inconjunction with the results the sterilitytests. The sterility test procedure shouldbe validated for a given product.Pharmacopoeial methods must be usedfor the validation and performance of thesterility test.For injectable products, the Water forInjection and the intermediate and finishedproducts should be monitored for endotoxins,using an established pharmacopoeialmethod that has been validated for each typeof product. For large-volume infusionsolutions, monitoring of water orintermediates should always be done, inaddition to any tests required by an approvedmonograph for the finished product.
