Accordingly Appendix 16 in Annex (B) is substituted for the existing Appendices 16and 17 in the Establishments Code.03. The provisions in the present <strong>Circular</strong> in regard to obtaining approval for foreigntravel of Public Officers shall take effect superseding the provisions in all other <strong>Circular</strong>sissued in this regard.Sgd./ D. 10.4 In respect of an officer proceeding abroad for such purpose as study ortraining, scholarship, seminar or workshop awarded by a sponsoring ForeignInstitution, the application perfected as in Appendix 16, should be addressedto the Secretary to the President/ Secretary to the Prime Minister/Secretaryto the appropriate Ministry/Secretary to the <strong>Provincial</strong> Governor by theSecretary to the Ministry/Chief Secretary to the <strong>Provincial</strong> <strong>Council</strong> asappropriate. An additional copy of the application should be forwarded tothe Director-General of Foreign Resources.10.5 In respect of an Officer referred to in Sub section 10.1 above, who isspecifically nominated by a Minister to proceed abroad at Governmentexpense for such purpose as participation in an international conference, anddiscussion and negotiations for any international agreement, the applicationperfected as in Appendix 16, should be forwarded by the Secretary to theMinistry/Chief Secretary of the <strong>Provincial</strong> <strong>Council</strong> as appropriate, to theSecretary concerned under 10.4 above.10.6 When a team of representatives is participating in a foreign tour, in case anysingle member is required to obtain the prior approval of His Excellency thePresident, permission of His Excellency the President should be obtained forall members of the team at one and the same time. Moreover, in the case ofsuch a team of representatives, permission should be obtained from thePrime Minister/Minister/Governor as the case may be, as a team at one andthe same time. In such a case, coordination of such procedures should bedone by the Ministry where the leader of the team in working, or by theMinistry in charge of the subject, as most appropriate. In the case ofdiscussions and agreements to be concluded for the purpose of obtainingforeign aid for the Government of Sri Lanka, the Ministry of Finance andPlanning shall be the coordinating Ministry.10.7 As soon as permission is granted by His Excellency the President/PrimeMinister/Minister/Secretary to the Ministry for a foreign tour as referred toin Sub Section 10.5 above, particulars pertaining to the officer’s foreigntour, together with a copy of the service agreement he has entered into withthe Government where appropriate,, should be furnished by the Secretary tothe Ministry /Chief Secretary of the <strong>Provincial</strong> <strong>Council</strong>, to the administrativeauthority of the Service to which the officer belongs.Example:In the case of an officer in the Sri LankaAdministrative serviceIn the case of an officer in the Sri LankaPlanning serviceIn the case of an officer in the Sri LankaEngineering service- To the Secretary of the Ministryin charge of the subject ofPublic Administration.- To the Secretary of the SriLanka Planning Service Board.- To the Director of EngineeringServices20

We wanted to test whetherprotein levels of any predicted miR-143 targets were altered inthe miR-143 ASO-treated adipocytes. However, of the miR-143targets proposed by Lewis et al. (4), only antibodies for ERK5/BMK1 were commercially available. Levels of ERK5 protein innormal adipocytes were compared with adipocytes that hadbeen treated with miR-143 ASO before induction of differentiation.After 7 or 10 days of differentiation, the level of ERK5protein, normalized to G3PDH levels, was 2-fold higher in themiR-143 ASO-treated cells (Fig. 4). The levels of ERK5 proteinwere unchanged in adipocytes treated with an ASO targetingmiR-23b, which also inhibited adipocyte differentiation in thisassay (data not shown). These data suggest that the up-regulationof ERK5 is specific to the miR-143 ASO treatment andnot a general result of inhibition of adipocyte differentiation,consistent with the hypothesis that ERK5 is a miR-143 targetgene.DISCUSSIONIn this study we have screened ASOs targeting miRNAs fortheir ability to modulate adipocyte differentiation in a culturedprimary human adipocyte model system. These data, togetherwith a global analysis of miRNA expression patterns, allowedus to identify several miRNAs involved in adipocyte differentiation.2-O-MOE ASOs were a useful tool for dissecting thebiological importance of miRNAs, and our library of miRNAASOs will be of use in a variety of in vitro and in vivo systemsto rapidly uncover the biological roles of miRNAs. In addition,ASOs targeting miRNA may be valuable as therapeutics in avariety of disease indications.One miRNA identified in the screen, miR-143, was of particularinterest. The observation that miR-143 is up-regulated asadipocytes differentiate, taken together with the inhibition ofadipocyte differentiation by miR-143 ASO, indicates that miR-143 normally promotes adipocyte differentiation. Our own efforts to find acancer cell line expressing miR-143 were also unsuccessful(data not shown).
