ABSTRACT<br />

Citati<strong>on</strong> |<br />

In April 2014, hundreds <str<strong>on</strong>g>of</str<strong>on</strong>g> ready-to-market c<strong>in</strong>eraria [ Senecio cruentus (Mass<strong>on</strong> ex L’Her.)<br />

DC., Asteraceae] plants, grown <strong>in</strong> pots for 4 m<strong>on</strong>ths <strong>in</strong> plastic greenhouses <strong>in</strong> Gangneung<br />

City, were severely damaged (∼30% <strong>in</strong>cidence) <str<strong>on</strong>g>by</str<strong>on</strong>g> a disease previously undocumented <strong>in</strong><br />

<strong>Korea</strong>. Diseased plants had small to large, brown or dark brown lesi<strong>on</strong>s <strong>on</strong> leaves and<br />

flowers. <str<strong>on</strong>g>Botrytis</str<strong>on</strong>g> sp. was c<strong>on</strong>sistently isolated from affected tissues. The pathogen appeared<br />

to <strong>in</strong>fect leaves al<strong>on</strong>g the marg<strong>in</strong>s and occasi<strong>on</strong>ally <strong>on</strong> other parts <str<strong>on</strong>g>of</str<strong>on</strong>g> the leaf blade and<br />

flower petals. The pathogen produced pr<str<strong>on</strong>g>of</str<strong>on</strong>g>use c<strong>on</strong>idia <strong>on</strong> the surface <str<strong>on</strong>g>of</str<strong>on</strong>g> dead and dy<strong>in</strong>g<br />

leaves and flowers, result<strong>in</strong>g <strong>in</strong> a moldy gray appearance and greatly reduc<strong>in</strong>g the aesthetic<br />

and commercial value <str<strong>on</strong>g>of</str<strong>on</strong>g> the plants. C<strong>on</strong>idia (n = 30) were <strong>on</strong>e-celled, ellipsoid or ovoid, 5.9<br />

to 10.8 × 5.2 to 8.6 μm <strong>on</strong> naturally <strong>in</strong>fected leaves, and 6.1 to 10.6 × 5.1 to 7.8 μm <strong>on</strong><br />

potato dextrose agar (PDA). M<strong>on</strong>oc<strong>on</strong>idial isolates were obta<strong>in</strong>ed <str<strong>on</strong>g>by</str<strong>on</strong>g> plat<strong>in</strong>g c<strong>on</strong>idial<br />

suspensi<strong>on</strong>s <strong>on</strong> PDA and select<strong>in</strong>g s<strong>in</strong>gle col<strong>on</strong>ies. Incubati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> isolates at 20 ± 2°C for 21<br />

days produced blackish, spherical to irregular microsclerotia <str<strong>on</strong>g>of</str<strong>on</strong>g> 1.2 to 4.0 × 1.2 to 3.0 mm (n<br />

= 20). A representative isolate (GM007) was deposited <strong>in</strong> Gangneung-W<strong>on</strong>ju Nati<strong>on</strong>al<br />

University and used for further studies. Morphological characters were c<strong>on</strong>sistent with those<br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>Botrytis</str<strong>on</strong>g> <str<strong>on</strong>g>c<strong>in</strong>erea</str<strong>on</strong>g> Pers.: Fr. (Ellis 1971). To c<strong>on</strong>firm identificati<strong>on</strong>, the <strong>in</strong>ternal transcribed<br />

spacer (ITS) regi<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> rDNA <str<strong>on</strong>g>of</str<strong>on</strong>g> the isolate was amplified with primers ITS1/ITS4 and<br />

sequenced. BLAST analysis <str<strong>on</strong>g>of</str<strong>on</strong>g> the result<strong>in</strong>g 518-bp nucleotide segment (GenBank Accessi<strong>on</strong><br />

No. KM840848) showed 100% identity with the sequence <str<strong>on</strong>g>of</str<strong>on</strong>g> Botryot<strong>in</strong>ia fuckeliana<br />

(teleomorph <str<strong>on</strong>g>of</str<strong>on</strong>g> B. <str<strong>on</strong>g>c<strong>in</strong>erea</str<strong>on</strong>g>). For further c<strong>on</strong>firmati<strong>on</strong>, nuclear prote<strong>in</strong>-cod<strong>in</strong>g genes (G3PDH<br />

and HSP60) were sequenced (Staats et al. 2005). The result<strong>in</strong>g sequences (DDBJ Accessi<strong>on</strong><br />

Nos. LC009697 and LC009698, respectively) showed 100% identity with those <str<strong>on</strong>g>of</str<strong>on</strong>g> B.<br />

fuckeliana (KP120866, KJ937067). To c<strong>on</strong>duct the pathogenicity test, <strong>in</strong>oculum was prepared<br />

<str<strong>on</strong>g>by</str<strong>on</strong>g> harvest<strong>in</strong>g c<strong>on</strong>idia from 2-week-old cultures <strong>on</strong> PDA. A c<strong>on</strong>idial suspensi<strong>on</strong> (2 × 10 4<br />

c<strong>on</strong>idia/ml) was sprayed <strong>on</strong>to healthy leaves <str<strong>on</strong>g>of</str<strong>on</strong>g> three 3-m<strong>on</strong>th-old potted c<strong>in</strong>eraria plants.<br />

Three plants were sprayed with sterilized water, serv<strong>in</strong>g as c<strong>on</strong>trols. Plants were covered with<br />

plastic bags for 3 days after <strong>in</strong>oculati<strong>on</strong> to ma<strong>in</strong>ta<strong>in</strong> high relative humidity and were placed <strong>in</strong><br />

a growth chamber at 20 ± 2°C. The first foliar lesi<strong>on</strong>s developed <strong>on</strong> leaves 4 days after<br />

<strong>in</strong>oculati<strong>on</strong>, whereas c<strong>on</strong>trol plants rema<strong>in</strong>ed symptomless. The pathogenicity test was<br />

carried out twice with similar results. The pathogen was successfully reisolated from<br />

<strong>in</strong>oculated leaves, fulfill<strong>in</strong>g Koch’s postulates. Associati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> c<strong>in</strong>eraria with B. <str<strong>on</strong>g>c<strong>in</strong>erea</str<strong>on</strong>g> has<br />

<strong>in</strong>oculated leaves, fulfill<strong>in</strong>g Koch’s postulates. Associati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> c<strong>in</strong>eraria with B. <str<strong>on</strong>g>c<strong>in</strong>erea</str<strong>on</strong>g> has<br />

been reported <strong>in</strong> the United States and the United K<strong>in</strong>gdom (Farr & Rossman 2015). To our<br />

knowledge, this is the first report <str<strong>on</strong>g>of</str<strong>on</strong>g> B. <str<strong>on</strong>g>c<strong>in</strong>erea</str<strong>on</strong>g> caus<strong>in</strong>g <str<strong>on</strong>g>Botrytis</str<strong>on</strong>g> blight <strong>on</strong> c<strong>in</strong>eraria <strong>in</strong> <strong>Korea</strong>.<br />

Accord<strong>in</strong>g to our field observati<strong>on</strong>s, leaf lesi<strong>on</strong>s expanded rapidly under cool, humid<br />

c<strong>on</strong>diti<strong>on</strong>s <str<strong>on</strong>g>of</str<strong>on</strong>g> n<strong>on</strong>heated greenhouses. Under dry c<strong>on</strong>diti<strong>on</strong>s, however, the disease developed<br />

slowly or even became quiescent. We suppose that ventilati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> the greenhouses may<br />

effectively reduce severity <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>Botrytis</str<strong>on</strong>g> blight <strong>on</strong> c<strong>in</strong>eraria.<br />
