GİRİŞ INTRODUCTION<br />
İstanbul Üniversitesi Genetik Kulübü (İÜGEN), İstanbul<br />
Üniversitesi Fen Fakültesi bünyesinde 2003 yılından<br />
itibaren aktif faaliyet gösteren bir öğrenci kulübüdür.<br />
Kurulduğu günden itibaren bilimsel bilgi birikiminin<br />
paylaşımını sağlayarak öğrenciler arasında sosyalleşme<br />
ağı oluşturmaktadır. Çalışmalarını, insanlara ve bilime<br />
karşı hissettiği sorumluluk bilinciyle<br />
gerçekleştirmektedir.<br />
İÜGEN kulübünün kuruluşuyla birlikte başlayan Moleküler<br />
Biyoloji ve Genetik Kış Okulu etkinliği, önce ulusal çapta,<br />
2008 yılından itibaren yurt dışından bilimin en güncel<br />
konuları ile uğraşan bilim insanlarının davet edilmesi ile<br />
etkinliklerini uluslararası çapta gerçekleştirmeye<br />
başlamıştır. İÜGEN, faaliyetlerine başladığı 2003 yılından<br />
itibaren düzenlediği etkinliklerle 7000’e yakın öğrenciyi<br />
ve 150’den fazla öğretim üyesini buluşturmuştur.<br />
Moleküler Biyoloji Genetik Kış Okulu etkinliğine<br />
çoğunlukla Moleküler Biyoloji Genetik ve Biyoloji<br />
bölümlerinden katılım olsa da, Tıp, mühendislik,<br />
matematik, fizik ve astronomi gibi bölümlerden<br />
gerçekleşen katılımlar, konuya ilgisi olan herkesin<br />
katılabileceği bir etkinlik olduğunu göstermektedir.<br />
Moleküler Biyoloji Genetik Kış Okulu etkinliği diğer tüm<br />
kulüp etkinlikleri gibi öğrenciler tarafından<br />
düzenlenmekte, etkinliğe katılan öğrencilerin ve davet<br />
edilen öğretim üyelerinin taleplerine en iyi şekilde karşılık<br />
verebilmek için çalışmalarını ciddiyetle<br />
gerçekleştirmektedir.<br />
Kulübün gerçekleştirdiği gerek etkinlikler, gerek sosyal<br />
çalışmalar, gerekse kongreler; düzenli ve hedefe yönelik<br />
çalışmaların ürünüdür. Bu bağlamda kulübümüz,<br />
bünyesinde bulunan üyelerin disiplinli ve organize<br />
çalışmaları sayesinde tüm İÜGEN organizasyonları<br />
başarıyla gerçekleştirilmiş ve gerçekleştirilmeye devam<br />
edilmektedir.<br />
Tüm çalışmalarımızla, bilime ve bilginin aktarılmasına<br />
yardımcı olabilmeyi hedeflemekteyiz. Eğer siz de bilime<br />
meraklıysanız, sitemiz ve/veya sosyal medya<br />
hesaplarımız aracılığıyla bizi takip edebilir, güncel<br />
etkinliklerimizden haberdar olabilirsiniz.<br />
İstanbul University Genetics Club (İÜGEN) is an active<br />
students club & society which has been operating within<br />
the structure of İstanbul University Faculty of Science<br />
since 2003. İÜGEN has been creating socializing network<br />
by empowering scientific knowledge shared between<br />
students and academic members, since its foundation. it<br />
has been performing Works, with sense of responsibilty<br />
felt towards to science and people.<br />
Molecular Biology and Genetics Winter School event,<br />
which has started with establishment of İUGEN, at first on<br />
national scale, from 2008 onwards organized on an<br />
international scale, by inviting academic members from<br />
abroad. Since 2003, it has brought nearly 7000 students<br />
and more than 150 academic members together with<br />
events organised by İUGEN.<br />
Generally, Molecular Biology and Genetics Winter School<br />
event’s atendees are from Molecular Biology and<br />
Genetics and Biology department. However, there are<br />
other participants apart from these departments such as;<br />
medical, math, physics and astronomy which showes us<br />
that anyone who is interested in this field can attend to<br />
this event.<br />
Molecular Biology and Genetics Winter School event is<br />
organised by students, same as all other students club<br />
events and studies are conducted earnestly in order to<br />
respond to demands of participants and invited academic<br />
members in the best way.<br />
All events, social activities and conferences carried out by<br />
club, are result of targeted and regular work. In this<br />
context of our club, accomplished by all İUGEN<br />
organizations and it will continue to to be performed<br />
successfully through organized and disciplined efforts of<br />
our club members.<br />
We aim to help the science and share the knowledge with<br />
our work. İf you are insterested in science, you can follow<br />

Fatma Neşe Kök 1994 yılında Orta Doğu Teknik<br />
Üniversitesi Kimya Mühendisliğinden mezuniyetini<br />
takiben 1997 yılında M.Sc ve 2001 yılında Ph.D.<br />
derecesini aynı üniversitenin Biyoteknoloji Programından<br />
aldı. Humboldt Vakfı bursu ile gittiği Max Planck<br />
Enstitüsü Polimer Araştırmaları (Mainz, Almanya) ve Max<br />
Plank Enstitüsü Biyokimya (Mantinstred, Almanya)<br />
Merkezlerinde doktora sonrası çalışmalarını<br />
tamamladıktan sonra (Şub 2003-Tem 2005) halen<br />
bulunduğu İstanbul Teknik Üniversitesi Moleküler Biyoloji<br />
ve Genetik Bölümünde Öğretim Üyesi olarak çalışmaya<br />
başladı (2005). İlgi alanları arasında biyomoleküllerin<br />
immobilizasyonu, biyomalzemeler, doku mühendisliği<br />
yapı iskeleleri, biyosensörler, kontrollü salım sistemleri ve<br />
yapay lipid zarlar konuları bulunmaktadır.<br />
Fatma Neşe Kök took M.Sc degree in 1997 and Ph. D<br />
degree in 2001 in the Biotechnology programme of the<br />
same university her graduation from Chemistry<br />
Engineering of Midlle East Technical University in 1994.<br />
After she completed her post. Ph. D. Studies in Max<br />
Planck Institute Polimer Researches (Mainz,Germany)<br />
and Max Planck Institute Biochemistry Centers<br />
(Mantinstred, Germany),she started to serve as lecturer in<br />
the department of Molecular Biology and Genetics of<br />
Istanbul Technical University in which she still works<br />
(February 2003-July 2005). Among her intrests are<br />
immobilization of biomolecules, biomaterials, tissue<br />
engineering scaffold, biocensors, controlled emission<br />
systems and artificial lipid membranes.<br />

Some biomaterials such as polimer, ceramic, metallic or<br />
composite are used in order to compensate the function<br />
loss caused by the damaged tissues because of various<br />
reasons like illness and accidents. The interaction<br />
between the implants produced from these biomaterials<br />
and the alive tissue must be good in order to perform<br />
successfully and this interaction performs on that<br />
surface. Features of the surface needs to have different<br />
features from those of the material for a successful<br />
performance. Physical, chemical and biological surface<br />
modifications are done for lots of purposes such as for<br />
increasing the cell adhesion, for adding an antimicrobial<br />
feature for decreasing trombojenisity and for stopping the<br />
corosion as an example of these methods, increasing the<br />
osteo entegration ability of titanium implant surfaces<br />
througt arc-PVD method (accumulating physical steam)<br />
increasing anti- trombojenic features by attaching<br />
proteins to PTFE surfaces (politetrafloroetilen) on which<br />
plasm is applied and increasing the multiplation and<br />
adhesion of the cell in tissue engineering by the help of<br />
polimer mixtures can be counted.<br />

Vücutta hastalık ya da kaza gibi çeşitli sebeplerden hasar<br />
görmüş dokuların yol açtığı işlev kayıplarını giderebilmek<br />
için polimer, metal, seramik ya da kompozit çeşitli<br />
biyomalzemeler kullanılır. Bu biyomalzemelerden üretilen<br />
implantların başarılı olabilmesi için yerleştirildikleri<br />
bölgedeki canlı doku ile etkileşiminin iyi olması<br />
gerekmektedir ve bu etkileşim yüzey üzerinden<br />
gerçekleşir. Başarılı bir uygulama için çoğu zaman yüzey<br />
özelliklerinin malzemenin yığın özelliklerinden değişik<br />
niteliklere sahip olması gerekir. Kullanım alanına göre<br />
hücre tutunmasını arttırmak, antimikrobiyal özellik<br />
kazandırmak, trombojenisiteyi azaltmak, korozyonu<br />
engellemek gibi pek çok amaçla, fiziksel, kimyasal ve<br />
biyolojik yüzey modifikasyonları yapılmaktadır. Bu<br />
yöntemlere örnek olarak, arc-PVD (fiziksel buhar<br />
biriktirme) yöntemi ile titanyum implantların yüzeylerinin<br />
osteoentegrasyon kabiliyetinin arttırılmasına<br />
çalışılması1,2, plazma uygulanan PTFE<br />
(politetrafloroetilen) yüzeylere protein bağlanarak antitrombojenik<br />
özelliklerinin arttırılması3 ve polimer<br />
karışımları yardımı ile doku mühendisliğinde hücre<br />
tutunması ve çoğalmasının arttırılması4 gibi uygulamalar<br />

Karbohidrat-Peptid epitoplarının klasik MHC-aracılı<br />
immun cevaplarda rolü olduğu hakkında kanıtlar<br />
birikmekte. O-glikozillenmiş peptidlerin T-hücresince<br />
tanınması, mikroorganizmalara karşı immün korunmada,<br />
ve özellikle tümör terapilerinde aşılamada aracı olmaları<br />
yüzünden yüksek derece biyomedikal öneme sahip.<br />
Epitelyum tip 1 transmembran mucin (MUC1), meme<br />
kanserinin nüksetme ihtimalini izlemek için bir<br />
işaretleyici olarak belirlenmiştir ve aktif özgül bağışıklıkla<br />
kanser tedavisindeki immünoterapötik stratejiler için<br />
umut verici bir hedef olacaktır. Mucin'in tümörle-ilgili<br />
glikoformlarının doğal insan bağışıklık cevabı, antikorun<br />
tepkisini glikozillenmemiş peptid epitoplarından çok<br />
glikopeptidlere gösterdiğini belirtiyor. MUC1'in deneysel<br />
immünizasyondaki tümörle-ilgili glikoformlarının zayıf<br />
immünojenitesinin üstesinden gelebilmek için antijenin<br />
etkili bir şeklide işlenmesi 1 ve 2. sınıfın seçmeli<br />
glikozilasyon sitlerinin tanımlanarak moleküler<br />
gereklilikler hakkında öngörü sahibi olabilmek için bu<br />
müdahaleler yapıldı. CD4+ T-hibridoma'lar ile yapılan<br />
çalışmalar, immünoproteozomal veya endozomal işleme<br />
makinelerinin O-glikopeptid ürünleri T hücrelerine etkili<br />
bir şekilde sunulabildiğini ve bu glikanlar TCR olarak<br />
tanımlanan epitopların integral kısmını oluşturabildiğini<br />
doğrular niteliktedir. İnsan MUC1 transgenik faredeki<br />
immunizasyon stratejileri, glikoformların<br />
glikozillenmemiş epitoplarının farklı formları immun<br />
sistem tarfından yabancı olarak algılandığını ve immun<br />
toleransı etkili bir şekilde kırabileceğini göstermiştir. Bu<br />
bulgulara dayanarak,APC-hedefli ve antijen spesifik<br />
kendi tepkisini arttıran epitoplardan oluşan multi epitop<br />
yapılara sahip transgenik farelere daha etkili aşılar<br />
tasarlandı ve başarı ile verildi.<br />
Evidence has accumulated that carbohydrate-peptide<br />
epitopes do play a role in classical MHC-mediated<br />
immune responses. T-cell recognition of O-glycosylated<br />
peptides is potentially of high biomedical significance,<br />
because it can mediate the immune protection against<br />
microorganisms, and in particular the vaccination in antitumor<br />
therapies. The epithelial type 1 transmembrane<br />
mucin MUC1 is established as a marker for monitoring<br />
recurrence of breast cancer and is a promising target for<br />
immunotherapeutic strategies to treat cancer by active<br />
specific immunization. Natural human immune responses<br />
to the tumor-associated glycoforms of the mucin indicate<br />
that antibody reactivities are more directed to<br />
glycopeptide than to non-glycosylated peptide epitopes.<br />
To overcome the weak immunogenicity of the tumorassociated<br />
glycoform of MUC1 in experimental<br />
immunization, attempts were made to get insight into the<br />
molecular requirements for effective antigen processing<br />
and to identify class I and II processing permissive<br />
glycosylation sites. Evidence based on work with CD4+ T-<br />
hybridomas confirms that Oglycopeptide products of the<br />
immunoproteasomal or endosomal processing<br />
machineries can be effectively presented to T-cells and<br />
that glycans can form integral parts of the TCR defined<br />
epitopes. Immunization strategies in human MUC1<br />
transgenic mice have demonstrated that different from<br />
nonglycosylated epitopes the glycoforms are recognized<br />
as foreign by the immune system and can effectively break<br />
immunotolerance. Based on these findings superior<br />
vaccines have been designed and successfully applied in<br />
transgenic mice that have a multi-epitope composition in<br />
common by comprising self-adjuvanting, APC-targeting<br />
and antigen-specific epitopes.<br />

Myelin is a material produced by specialized glial cells of<br />
the nervous system that forms an isolating layer around<br />
axons, facilitating neuronal electrical impulse<br />
propagation and communication. Dysmyelination or<br />
demyelination (improper development or loss of myelin,<br />
respectively) occurs in many neurological disorders, such<br />
as multiple sclerosis, Pelizaeus–Merzbacher disease and<br />
other leukodystrophies, and as a consequence of spinal<br />
cord injury, leading to disruption of electrical impulse<br />
conductivity, atrophy of neurons, and permanent<br />
functional deficits. Exploring myelin biology and modeling<br />
myelination disorders in vitro using human and mouse<br />
cells should facilitate closing the gaps in the<br />
understanding of underlying mechanism by which<br />
myelination occurs and should promote the efforts<br />
towards novel therapies.<br />
Miyelin özelleşmiş glial hücreleri tarafından üretilen,<br />
aksonlar etrafında yalıtıcı bir tabaka oluşturup, nöronal<br />
elektriksel impulsun ortaya çıkışını ve iletişimi hızlandırıcı<br />
bir materyaldir. Dismiyelinizasyon ya da demiyelinizasyon<br />
(sırasıyla, miyelinin hatalı gelişimi veya kaybı), Multiple<br />
Sclerosis (MS), Pelizaeus-Merzbacher hastalığı, diğer<br />
lökodistrofiler ve omurilik yaralanmaları gibi nörolojik<br />
rahatsızlıklarda görülen durumlar olup, elektriksel impuls<br />
iletkenliğinde kesiklikler, nöronlarda atrofi ve kalıcı<br />
işlevsel eksikliklerle sonuçlanabilir. İnsan ve fare<br />
hücrelerini kullanarak in vitro da miyelin biyolojisini<br />
araştırmak ve miyelinizasyon hastalıklarını modellemek,<br />
Miyelinleşme altında yatan mekanizma hakkında bilgi<br />
eksikliğinin kapanmasını hızlandırıp yeni tedavi<br />
yöntemlerinin bulunmasına yardımcı olacaktır.<br />

Prof. Dr. Y. Murat Elçin 1996’dan bu yana Ankara<br />
Üniversitesi’nde öğretim üyesi (Biyokimya) olarak görev<br />
yapmaktadır. Aynı zamanda 2005 yılından beri A.Ü. Fen<br />
Fakültesi Kimya Bölümü Biyokimya ABD’nın başkanlığını<br />
ve 2010 yılından bu yana A.Ü. Kök Hücre Enstitüsü‘nün<br />
kurucu müdür yardımcılığını sürdürmektedir. Prof. Elçin,<br />
Biyokimya alanındaki yüksek lisans (1990) ve doktora<br />
(1993) öğreniminin ardından doktora sonrası<br />
çalışmalarını ABD’de Kaliforniya Üniversitesi (Los<br />
Angeles) Tıp Fakültesi’nde gerçekleştirmiştir (1995).<br />
Sonraki yıllarda Britanya’da Oxford Üniversitesi’nde<br />
(1997), ABD’de UCLA’de (1998) ve Almanya Giessen<br />
Üniversitesi’nde (2000, 2001) ziyaretçi araştırmacı veya<br />
konuk bilim adamı olarak araştırmalar yapmıştır. Prof.<br />
Elçin’in başlıca araştırma konuları klinik doku<br />
mühendisliği, kök hücreler, rejeneratif tıp, rejeneratif<br />
biyomalzemeler ve tıbbi cihazlardır. 2003 yılında Dr. M.<br />
Binnet ile işbirliği çerçevesinde Türkiye’deki ilk klinik<br />
doku mühendisliği araştırmasını gerçekleştirmiştir.<br />
Kurucusu ve lideri olduğu Araştırma Grubu (ElçinLab)<br />
halen on beşten fazla üyeye sahip olup, yıllar içerisinde<br />
yaklaşık 6 milyon TL’lik (ulusal ve uluslararası) araştırma<br />
kaynağı kullanmıştır. Geçmişte ve günümüzde<br />
danışmanlığını yaptığı 35’in üzerinde lisansüstü öğrencisi<br />
bulunmaktadır (15 Doktora ve 20+ YL öğrencisi). Prof.<br />
Elçin’in, yayınlanmış 70 civarında SCI kapsamında<br />
uluslararası makalesi (Web of Science’da 1025+ atıf ve<br />
h-indeksi 19) ve kitap bölümü ile 170+ konferans bildirisi<br />
bulunmaktadır.<br />
Y. Murat ELÇİN, PhD is a Professor of Biochemistry at<br />
Ankara University, Ankara, Turkey since 1996. He also<br />
serves as Chair of Biochemistry Division at Faculty of<br />
Science (since 2005) and as founding Deputy Director of<br />
Ankara University Stem Cell Institute (since 2010). He<br />
studied chemistry at Ankara University; received MSc<br />
(1990) and PhD (1993) titles in Biochemistry from AU;<br />
performed postdoctoral training at University of<br />
California-Los Angeles School of Medicine, Los Angeles,<br />
USA (1995). He was a research associate/visiting<br />
scientist at Oxford University, UK (1997), UCLA, USA<br />
(1998) and University of Giessen, Germany (2000,<br />
2001). Prof. Elcin’s major research interests are clinical<br />
tissue engineering, stem cells, regenerative medicine,<br />
regenerative biomaterials and medical devices. He has<br />
been involved in the first clinical tissue engineering study<br />
of Turkey (2003), in collaboration with Prof. Dr. M. Binnet,<br />
MD. He has established the ElcinLab and is currently<br />
leading a research group of >15 members (lab funding of<br />
>2M € from national and international sources). He is (or<br />
has been) the supervisor of >35 post-graduate studies<br />
(15 PhD, >20 MSc). He is the co-author of around 70<br />
peer-reviewed publications in scientific journals and<br />
books, and around 170 conference proceedings (with<br />
around thousand citations, h-index of 19 in SCI).<br />

Prof. Elcin is the inventor of several pending patents and<br />
the founder of a startup company. He is editor of the book<br />
“Tissue Engineering, Stem Cells and Gene Therapies”<br />
(2003 Kluwer-Plenum), and translation editor of the book<br />
“Lehninger- Principles of Biochemistry-5 th ed.” (2013<br />
Palme). Prof. Elcin is an elected associate member<br />
(2008) of the Turkish Academy of Sciences (TÜBA) and<br />
the coordinator of TÜBA Stem Cell Working Group, and<br />
recipient of several science awards. He is the founding<br />
member of the Biomaterials and Tissue Engineering<br />
Society (Turkey), and The Society for Cellular Therapy and<br />
Regenerative Medicine (Turkey). He has organized several<br />
international scientific meetings, including BIOMED<br />
2002 and BIOMED 2011. He serves (or has served) as<br />
scientific consultant or as advisory committee member<br />
for a number of national and international organizations<br />
and funding agencies. He is in the editorial board of ten<br />
scientific journals including, Stem Cell Reviews and<br />
Reports (Springer), Journal of Tissue Engineering and<br />
Regenerative Medicine (Wiley), and Genes and Diseases<br />
(Elsevier).<br />
Prof. Elçin’in çok sayıda patent başvuruları ve gelişme<br />
aşamasında bir teknokent şirketi bulunmaktadır. “Tissue<br />
Engineering, Stem Cells and Gene Therapies,” (2003<br />
Kluwer-Plenum) başlıklı kitabın editörlüğünü ve<br />
“Lehninger- Biyokimyanın Temel İlkeleri-5. Basım”<br />
kitabının (Palme 2013) çeviri editörlüğünü<br />
gerçekleştirmiştir. Prof. Elçin, 2008’den bu yana Türkiye<br />
Bilimler Akademisi’nin (TÜBA) seçilmiş asosiye üyesi<br />
olup, TÜBA Kök Hücre Çalışma Grubu’nun yürütücüsüdür.<br />
Ankara Üniversitesi 2008 yılı Bilim Ödülü ve 2002 TÜBA-<br />
GEBİP Ödülü sahibidir. Biyomalzeme ve Doku<br />
Mühendisliği Derneği ile Hücresel Tedavi ve Rejeneratif<br />
Tıp Derneği’nin kurucu üyesidir. Kariyeri boyunca BIOMED<br />
2002 ve BIOMED 2011 başta olmak üzere, uluslararası<br />
bilimsel toplantıları düzenlemiştir. Geçmişten günümüze<br />

Rejeneratif tıp (yenileyici tıp), hastalık, yaşlanma veya travma<br />
sonucu zarar gören ya da işlevlerini kaybeden doku, organ<br />
veya sistemlerin, insan hücreleri, dokuları veya organlarının<br />
değiştirilmesi, mühendisliği veya yenilenmesi süreçleriyle<br />
ilgilenen bir çevrimsel (translational) araştırma alanıdır.<br />
Hücre, kök hücre ve gen tedavileri, doku ve organ<br />
mühendisliği ve rejeneratif biyomalzemeler alanın<br />
kapsamındadır. Rejeneratif tıp yaklaşımları, vücudun kendi<br />
onarım mekanizmalarının üstesinden gelemediği<br />
hastalıklarda, doku/organ kayıplarında veya işlev<br />
bozukluklarında devreye girmektedir. Hastanın vücuduna<br />
hücrelerin, doku taslaklarının veya rejeneratif<br />
biyomalzemelerin nakledilmesi yolu ile iyileşme süreçlerine<br />
doğrudan veya dolaylı olarak katkı sağlanabilmektedir (invivo<br />
yenilenme). Doku mühendisliği, laboratuvarda (in-vitro<br />
şartlarda) ekstraselüler matriks-benzeri özel destek yapıları<br />
üzerinde çoğaltılan hücrelerin fizyolojik şartları taklit eden<br />
biyoreaktörler içinde doku taslaklarına dönüştürülmesini ve<br />
sonrasında hastanın vücuduna nakledilmesini içeren<br />
rejeneratif tıp teknolojisidir. Organ mühendisliğinin ise, tam<br />
organ inşa etmek gibi zor bir hedefi vardır. Bu nedenle<br />
allojenik (insan kaynaklı) veya zenojenik (hayvan kaynaklı)<br />
tam organların hücrelerinden uzaklaştırılıp, yerine hastanın<br />
kendi kök hücrelerinin tohumlanmasını içeren kişiye-özel<br />
organ oluşturma yaklaşımı gündeme gelmiştir.Kök hücreler,<br />
asimetrik bölünme özelliği gösteren uzmanlaşmamış<br />
(farklılaşmamış) hücrelerdir. Asimetrik bölünme özelliklerine<br />
bağlı olarak, hem kendi eşdeğerlerine (aynı özelliklere sahip<br />
yeni bir kök hücreleri), hem de geçiş (transit) hücreleri<br />
aracılığıyla özelleşmiş (uzmanlaşmış) hücrelere (karaciğer<br />
hücresi, sinir hücresi gibi) dönüşebilirler. Bu üstün özellikleri<br />
nedeniyle rejeneratif tıbbın başlıca unsurlarındandır. Kök<br />
hücrelerin hasarlı/hastalıklı dokuların yerini alarak ve/veya<br />
salgıladıkları sitokinler (parakrin etkiler) aracılığıyla etkinlik<br />
gösterdikleri kabul edilmektedir. Kök hücre tedavileri (kemik<br />
iliği hücreleri, hematopoetik kök hücreler) ilk önce 1970’li<br />
yıllarda kan kanserlerinin tedavisinde uygulanmaya<br />
başlanmıştır. Sonraları vücudun tüm dokularında bulunduğu<br />
anlaşılan mezenkimal kök hücreler gündeme gelmiştir.İnsan<br />
embriyonik kök hücrelerinin laboratuvarda çoğaltılabilmesi<br />
rejeneratif tıp için bir dönüm noktası olmuştur. Son olarak,<br />
vücut hücrelerinin yeniden programlanarak uyarılmış<br />
pluripotent kök hücrelere (embriyonik kök hücre-benzeri<br />
hücrelere) dönüştürülebilmesiyle kişiye-özel tıp (personalized<br />
medicine) kavramı daha anlamlı hale gelmiştir.<br />
Regenerative medicine (regenerating medicine) is an area<br />
of research that disease, aging, trauma caused damaged<br />
or lost function of tissue, organs or systems human cells,<br />
replacement of human cells, tissues or organs,<br />
engineering or dealing with renewal process cyclically<br />
(translational). Cells, stem cells and gene therapies,<br />
tissue and organ engineering and regenerative<br />
biomaterials are within the scope area. Regenerative<br />
medicine approaches are put in diseases that body's own<br />
repair mechanisms cannot overcome or tissue/organ<br />
dysfunction or loss. With transport the regenerative cells<br />
of tissues or biomaterials to the patient's body, it can<br />
contribute directly or indirectly to the healing process (invivo<br />
regeneration). Tissue engineering is a regenerative<br />
medicine technology that includes in the laboratory (invitro)<br />
the cells which growth in extracellular matrix-like<br />
special support structures, to turn them into tissue plan<br />
within the bioreactors that mimic physiological conditions<br />
and then transplant them to the patient’s body. But the<br />
organ engineering has a goal to build the total organs.<br />
Therefore, after removing of allogeneic (Human origin) or<br />
xenogeneic (Animal origin) cells from total organs, it has<br />
come into question that seeding the patient's own stem<br />
cells to create person-specific organs. Stem cells are<br />
undifferentiated biological cells that showing the<br />
asymmetric division. Depending on the asymmetric<br />
division characteristics, they can turn into their<br />
equivalents (a new stem cells have the same<br />
characteristics) and by transition (transit) cells they can<br />
give specialized (differentiated) cells (neurons, liver cells<br />
etc.). These are the main elements of regenerative<br />
medicine due to their outstanding properties. It is<br />
accepted that the stem cells show their efficacy by<br />
secreting the cytokines (paracrine effects) and replacing<br />
the damaged/diseased tissues. Stem cells treatments,<br />
firstly it began to be applied in the treatments of blood<br />
cancers in the 1970s. Then the presence of mesenchymal<br />
stem cells apparently came up where they are located<br />
every tissue of the body. Reproducing the human<br />
embryonic stem cells in the laboratory has been a turning<br />
point for the regenerative medicine. Finally, by<br />
reprogramming the somatic cells to the induced<br />
pluripotent stem cells, person-specific medical concept<br />
has become more meaningful.<br />

Aslı Tolun received her undergraduate degree in physics<br />
from Robert College which is now Boğaziçi University.<br />
After she received a Master's degree in biophysics from<br />
the University of Uppsala and a Scientific Microbiology<br />
doctoral degree from Pennsylvania State University, she<br />
pursued her postdoctoral work for 3 years in San Diego<br />
Department of Biology at the University of California.<br />
In 1982 she returned to Turkey and began working as a<br />
faculty member in Department of Molecular Biology and<br />
Genetics at Boğaziçi University. She received a TÜBİTAK<br />
Encouragement Award and two Boğaziçi University Senior<br />
Researcher Awards. She established the Ethics<br />
Committee on Human Research of the Boğaziçi University<br />
and chaired it for three years. She advised the Science<br />
Students Club of the University. She was a former member<br />
of the Turkish Academy of Sciences.<br />
The main interests of Dr. Tolun's team are searching for<br />
new disease genes and population genetics. The majority<br />
of her study cases are about nervous system<br />
(neurological) disease, especially neurodegenerative<br />
ones.<br />
As a result of her genetic studies about a wide range of<br />
diseases such as eye diseases and infertility in men, she<br />
discovered twelve new disease genes. In addition, she<br />
identified seven new types of disease which is caused<br />
from defects of known genes.<br />
Aslı Tolun şimdi Boğaziçi Üniversitesi olan Robert Kolej<br />
Yüksek Okulundan fizik lisans derecesi, Pennsylvania Eyalet<br />
Üniversitesinden Biyofizik yüksek lisans derecesi ve<br />
Uppsala Üniversitesinden bilimsel mikrobiyoloji doktora<br />
derecesi aldıktan sonra, üç yıl California Üniversitesi San<br />
Diego Biyoloji Bölümünde doktora sonrası çalışması yaptı.<br />
1982 yılında Türkiye’ye dönerek, Boğaziçi Üniversitesi<br />
Moleküler Biyoloji ve Genetik Bölümünde öğretim üyesi<br />
olarak çalışmaya başladı. TÜBİTAK Teşvik Ödülü ile iki kez<br />
Boğaziçi Üniversitesi Kıdemli Araştırıcı Ödülü aldı. Boğaziçi<br />
Üniversitesinin İnsan Araştırmaları Etik Kurulunu kurdu ve<br />
üç yıl başkanlığını yaptı. Üniversitenin öğrenci Bilim Kulübü<br />
danışmanlığını yaptı. Eski Türkiye Bilimler Akademisi<br />
üyesiydi.<br />
Dr. Tolun’un ekibinin araştırma alanları yeni hastalık genleri<br />
arama ve toplum genetiğidir. Çalıştığı hastalıkların büyük<br />
çoğunluğu sinir sistemi (nöroloji) hastalığı olup, önemli bir<br />
kısmı hızla ilerleyen (nörodejeneratif) türdendir. Göz<br />
hastalıkları ve erkekte kısırlık gibi geniş bir yelpazedeki<br />
hastalıklar üzerine yapılmış genetik araştırmalar<br />
sonucunda on iki yeni hastalık geni bulunmuştur. Ayrıca,<br />
bilinen genlerdeki kusurlardan kaynaklanmış yedi yeni<br />
hastalık tipi belirlenmiştir.<br />

SEARCHING FOR NEW<br />
DISEASE GENES<br />
Son yıllarda geliştirilen genetik analiz teknolojilerinin en<br />
yaygın kullanım alanı yoğun emek ve parasal kaynak<br />
gerektiren yeni hastalık genlerinin belirlenmesidir.<br />
Hastalıkların çoğunun nadir olmasından dolayı bulgular<br />
çok az sayıda aileyi ilgilendirse de, bu bulgular insan<br />
genetiğini çözmemize çok büyük katkı sağlar. Bilimsel<br />
açıdan çok önemli olmalarının nedeni, genlerimizin<br />
işlevlerinin en kesin olarak genetik hastalıkların<br />
çalışılmasıyla çözülmesidir. Aydınlatılan her yeni gen<br />
işlevi yeni bir hücresel mekanizmaya ışık tutarak genetik<br />
bilimine katkıda bulunur. Bir insan geninin işlevinin<br />
anlaşılmasının en kesin yolu ise, o geninde hata olan<br />
kişilerin (hastaların) klinik olarak incelenmesidir. Bu<br />
amaçla, nadir hastalıklardan muzdarip ailelerde<br />
hastalıktan sorumlu gen aranır. Yapılan genetik<br />
araştırmayla, “Bu ailenin hastalarında hangi gen<br />
kusurludur ki böyle bir hastalık ortaya çıkmıştır?”<br />
sorusuna yanıt aranır. Genetik araştırmada o genin önce<br />
yeri belirlenir, sonra kendinin bulunmasına çalışılır. Yeni<br />
nesil genetik dizileme teknolojileri tüm genlerimizin<br />
yapısını ortaya çıkarabilmektedir. Ama bu teknolojiler<br />
sayesinde, sağlıklı görünen kişilerde bile çok sayıda gen<br />
kusuru olduğunu anladık. Dolayısıyla, hangi gen<br />
kusurunun hastalıktan sorumlu olduğunu anlamak için<br />
çeşitli analizlerin yapılması elzemdir.<br />
Genin bulunması genellikle hastalara genellikle bir yarar<br />
sağlamasa da, ailede yeni hasta çocukların doğması<br />
önlenebilir. Hastalık geni bulmaya yönelik araştırmalar<br />
ana hatlarıyla, teknik ayrıntılara girilmeden anlatılacak ve<br />
keşfedilmiş hastalık genlerinden örnekler verilecektir.<br />
In recent years, The most common application of<br />
developed genetic analysis technologies is identification<br />
of new disease genes which is labor intensive and<br />
requires financial resources. Even though few families<br />
care about the diseases since most of them are very rare,<br />
the findings about those diseases provide a very large<br />
contribution to solving human genetics.<br />
Function of our genes is generally understood by studying<br />
genetic disease cases, and that is why studying genetic<br />
diseases have a huge significance. Every new discovered<br />
gene functions brighten to a new cellular mechanism<br />
function and contributes to genetics. The most accurate<br />
way of understanding the function of a human gene is to<br />
examine people’s gene with errors (the patient) clinically.<br />
For this purpose, the gene responsible for the disease is<br />
searched among families that are suffering from rare<br />
diseases.<br />
By the genetic research, it is searched for answers<br />
following question:<br />
"Which gene is defective in this family of the patient that<br />
has emerged such a disease?"<br />
In genetic research, firstly the position of gene is<br />

generation genetic sequencing technology can emerge<br />
the structure of all our genes. Moreover, thanks to this<br />
technology, we also understand that there is a large<br />
number of gene defects even in apparently healthy<br />
people. Therefore, it is essential make various analysis to<br />
understand which gene defect is responsible for the<br />
disease. Although generally, identification of gene does<br />
not provide any benefit to patients, the birth of diseased<br />
babies can be prevented. The main line of research to find<br />
disease gene will be explained without technical details<br />
and examples of discovered disease genes will be given.<br />

the director of Çorlu Brunch of Elçi Educational Institutes.<br />
In 2008, she established Pera. Lenguages Academy as<br />
cfounder and established Pera Cafeteria in 2012. She<br />
still has continued her activities.<br />
Hülya Perin living in Tekirdağ Çorlu, served as (2008-<br />
2010) vice president of Young Entrepreneurs Commission<br />
of Tekirdağ of TOBB. She took part in work Commission<br />
since 2010. She has been serving as the president of<br />
Young Entrepreneur Commission and since 2013, the<br />
president of work committee. She was elected as a<br />
member of Çorlu Municipality. In addition to her other<br />
works, she has continued to work actively in the<br />
municipality too.<br />

Meslek Komitesinde görev aldı. 2010 yılından bu yana iki<br />
dönemdir İl Genç Girişimciler İcra Kurulu Başkanlığı,<br />
2013’den itibaren de 14. Meslek Komitesi Başkanlığı<br />

Tim Hunt was, until his retirement in 2010, a ‘principal<br />
scientist’ (note, not THE principal scientist) at Cancer<br />
Research UK, Clare Hall Laboratories, in South Mimms,<br />
Hertfordshire. Tim was born in 1943 and grew up in<br />
Oxford, moving to Cambridge to read Natural Sciences<br />
in 1961. He obtained his Ph.D. (“The Synthesis of<br />
Haemoglobin”) from the Department of Biochemistry in<br />
Cambridge in 1968. He spent almost 30 years in<br />
Cambridge, working in the Department of Biochemistry,<br />
at first on the control of protein synthesis in red blood<br />
cells, sea urchin eggs and clam oocytes and from about<br />
1983 onwards on the control of the cell cycle. He has<br />
also worked in the USA; he was a postdoctoral Fellow at<br />
the Albert Einstein College of Medicine from 1968-70<br />
and spent summers at the Marine Biological<br />
Laboratory, Woods Hole from 1977 until 1985,<br />
teaching laboratory courses and doing research. In<br />
1982, he discovered cyclins, which turned out to be<br />
components of “Key Regulator(s) of the Cell Cycle”. This<br />
led to a share of the Nobel Prize in Physiology or<br />
Medicine in 2001, together with Lee Hartwell and Paul<br />
Nurse. Tim Hunt was chairman of the council of EMBO<br />
(European Molecular Biology Organization) from 2006 -<br />
2010. From 2011 to 2015, he was a member of the<br />
Scientific Council of the ERC (European Research<br />
Council). Tim has been helping to make up problems for<br />
Molecular Biology of the Cell by Alberts et al. since<br />
1985, and The Problems Book is now in its 6th edition.<br />

Oxford’da bilim adamı olmak isteyerek, kurşun boruları<br />
eritmek ya da tuz solüsyonlarını zehirli ve patlayıcı gazlar<br />
haline getirmek için elektrolize etmek gibi süreçlerle ve<br />
araç gereçlerle ilgilenerek büyüdüm. Neyse ki, bendeki bu<br />
hevesi kimya ve biyolojinin daha derin ve daha formal<br />
anlayışına kanalize eden harika öğretmenlerim oldu ve<br />
böylelikle Cambridge Üniversitesinde Biyokimya alanında<br />
hemoglobin sentezleri üzerine doktora çalışması yapma<br />
imkanım oldu. Buralara nasıl ulaştığımı sizlerle<br />
paylaşacağım; bu bir kazaydı ve aynı zamanda derse<br />
devam ettiğim yerdi. Hem sentezinin globin sentezi ile<br />
nasıl iş birliği yaptığını çözmek bir çok ilginç yan yol, çok<br />
fazla seyahat ile birlikte 10 yılımı aldı.<br />
Daha sonra, üzerinde çalışılacak iyi bir sorun bulmak ise<br />
bir başka 7 yılımı daha aldı fakat 22 Temmuz 1992’de<br />
Woods Hole’da Deniz Biyolojisi Laboratuvarında hem<br />
araştırma hem de öğretmenlik yaparken sonraları siklin adı<br />
verilen önemli bir proteinin, döllenmiş deniz kirpisi<br />
yumurtalarının ilk kez bölünmesinden hemen önce ortadan<br />
kaybolduklarını hayranlıkla izledim. Bu proteinin ne<br />
olduğunu, ne işe yaradığını anlamam 7 yılımı aldı ve bu<br />
süreç, protein sentezlerinin kontrolünden hücre bölünmesi<br />
kontrolüne doğru yol alan bir süreçti. Fakat şaşırtıcı olan<br />
şu ki temelde yatan mekanizmalar benzerdir ve fosfat<br />
tortularını diğer proteinlerin üzerine yapıştırarak onların<br />
davranışını değiştiren ve protein kinazlar olarak bilinen bir<br />
enzim türü de bu sürece dahildir.<br />
Son zamanlarda, hücre bölünmesini başlatan, sonlandıran<br />
ve çok önemli olduğu ortaya çıkan proteinler ve onların<br />
kontrolünden fosfatları çıkartan enzimleri çalışmak ilgim<br />
dahiline girdi. Hemen hemen her zaman,farklı şeyleri test<br />
etmek için tasarlanmış deneylerden kaynaklanan bu yolda<br />
beklenmedik keşiflere ulaşılmıştır!<br />
I grew up in Oxford wanting to be a scientist, loving<br />
gadgets and processes like melting lead pipes or<br />
electrolyzing salt solutions to make poisonous and<br />
explosive gases. Luckily, I had excellent teachers who<br />
channeled these enthusiasms into a deeper and more<br />
formal understanding of chemistry and biology (physics,<br />
alas, was beyond my grasp) so that it was possible to<br />
study at Cambridge University and carry on there with a<br />
Ph.D. in biochemistry, on the business of the control of<br />
haemoglobin synthesis. I’ll explain how I arrived at this—it<br />
was an accident—and also where I pursued the subject. It<br />
took ten years, many interesting side roads, a lot of travel<br />
and a devastating fire to solve the problem of how the<br />
synthesis of haem was coordinated with the synthesis of<br />
globin.<br />
After that, it took another 7 years or so to find a really<br />
good new problem to work on, but on July 22nd 1982 I<br />
was teaching and researching at the Marine Biological<br />
Laboratory, Woods Hole, and saw to my amazement that a<br />
prominent protein, later called cyclin, disappeared just<br />
before fertilized sea urchin eggs divided for the first (and<br />
every subsequent) time they divided. Finding out what this<br />
protein was, and what it did, took another six or seven<br />
years of very exciting work, leading away from the control<br />
of protein synthesis to the control of cell division. Yet<br />
amazingly, the underlying mechanisms were identical,<br />
involving a class of enzymes known as protein kinases,<br />
which attach phosphate residues onto other proteins,<br />
thereby modifying their behavior. I’ve always liked<br />
biological switches and finding how they work.<br />
Most recently, however, I’ve been drawn to the study of the<br />
enzymes that remove phosphates from proteins and their<br />
control, which turn out to be very important in the switches<br />
that initiate and terminate cell division. The path was<br />
marked by unexpected discoveries all along the way,<br />
almost always stemming from sensible experiments<br />
designed to test something different!<br />

Mehmet Baki Yokeş was born in 1967 in Istanbul. He<br />
received his PhD degree at Bogazici University<br />
Department of Molecular Biology and Genetics in 2005<br />
which he also completed his undergraduate and graduate<br />
programs at the same University. He has published<br />
national and international 80 scientific papers about<br />
marine biodiversity, ecology and genetics. He worked as a<br />
researcher and manager about 14 national and<br />
international projects related to marine life. As a<br />
supervisor on Two European Union Projects, M. Baki<br />
Yokeş also supervising to both national and international<br />
Non-profit Organization about marine diversity. Currently<br />
works as a lecturer at Haliç University Molecular Biology<br />
and Genetics, M. Baki Yokeş also Head of the Department<br />
and Vice-Dean of the Faculty of Arts and Sciences.<br />
Mehmet Baki Yokeş 1967 yılında İstanbul’da doğmuştur.<br />
Lisans ve Yüksek Lisans eğitimini tamamladığı Boğaziçi<br />
Üniversitesi Moleküler Biyoloji ve Genetik Bölümünden<br />

almıştır. İki Avrupa Birliği Projesinde danışman olarak yer<br />
alan M. Baki Yokeş ulusal ve uluslararası STK’lara denizel<br />
biyoçeşitlilik danışmanı olarak görev almaktadır. Halen<br />
Haliç Üniversitesi Moleküler Biyoloji ve Genetik Bölümü<br />
Öğretim Üyesi olan M. Baki Yokeş Fen Edebiyat Fakültesi<br />

BIODIVERSITY IN THE<br />
MEDITERRANEAN SEA<br />
Akdeniz jeolojik geçmişi ve doğu-batı doğrultusunda<br />
uzanan kapalı bir deniz olması nedeniyle denizel<br />
biyoçeşitlilik bakımından sıcak bir nokta oluşturmaktadır.<br />
Bugüne kadar Akdeniz’de varlığı tespit edilmiş 17.000<br />
civarında tür listelenmiş olsa da, bu listenin hala eksik<br />
olduğu kabul edilmektedir. Afrika kıtasının Avrupa<br />
kıtasına çarpmasıyla arada kapalı kalan Tetis Denizi’nin<br />
tamamına yakının kuruması sonucunda büyük tuz<br />
yataklarının oluştuğu Mesinian Krizi sonunda Cebeli Tarık<br />
Boğazı kırılıp Atlantik suları tekrar bu alana dolmuş ve<br />
bugünkü Akdeniz ortaya çıkmıştır. Atlantik’ten Akdeniz’e<br />
yayılan türler genelde Atlantik’ten daha tuzlu ve daha<br />
sıcak bir denizle karşılaşmış olmalarının yanı sıra, çok<br />
değişik çevresel koşullara sahip bölgelere de uyum<br />
sağlamak zorunda kalmışlardır. Akdeniz biyotasının<br />
kökeni Atlantik olmasına karşın çevre koşullarının<br />
çeşitliliği Akdeniz’e endemik türlerin ortaya çıkmasına yol<br />
açmıştır. Akdeniz alan olarak dünya denizlerinin %0,8’ini<br />
kapsamasına karşın denizel türlerin %8’ini içermektedir.<br />
Süveyş Kanalı’nın açılmış olması ve deniz taşımacılığı<br />
önemli sayıda yabancı türün dünya denizlerinden<br />
Akdeniz’e taşınmasına ve yerel ekosistemlerin<br />
değişmesine yol açmaktadır. Bugüne kadar Akdeniz<br />
kıyılarında 400’ün üzerinde yabancı denizel tür tespit<br />
edilmiştir. Bu türlerin bazılarının ekonomik bir değer<br />
olmasının yanında ekonomik zarara yol açan, insan<br />
sağlığını olumsuz etkileyen türler de bulunmaktadır.<br />
Akdeniz’in jeolojik koşulları ve dolayısıyla çevresel<br />
koşulları var olduğu günden bu yana sürekli bir değişim<br />
göstermekte ve biyotanın hızlı bir şekilde değişmesine yol<br />
açmaktadır.<br />
Due its geological history and enclosed sea lying on the<br />
east-west direction, the Mediterranean is an important<br />
hot spot for the marine biodiversity. Heretofore, in the<br />
Mediterranean, the presences of around 17.000 listed<br />
species has been identified, but there are still considered<br />
missing from this list. The impact between African and<br />
European continent resulted almost all of the enclosed<br />
Tethys Sea to dry and at the end of the Messinian crisis<br />
huge salt deposits formed, by the final event of breaking<br />
the strait of Gibraltar, the Atlantic Ocean has filled the<br />
gap and shaped the Mediterranean to its present<br />
appearance. The spreading species to Mediterranean<br />
from Atlantic have encountered with a sea that is more<br />
salty and warm, they have been forced to adapt a new<br />
environmental conditions. Although, the origin of the<br />
Mediterranean biota is Atlantic but due to diversity of<br />
environmental conditions have led to emergence of<br />
endemic species in Mediterranean. Mediterranean<br />
contains 8% of marine species despite that it covers 0.8%<br />
of the world's oceans. The opening of Suez Canal, a<br />
significant number of foreign species spread to<br />
Mediterranean by maritime transport from world seas and<br />
as a result it leads to a change in the local ecosystem.<br />
More than 400 foreign marine species in the<br />
Mediterranean Sea have been identified so far. Besides<br />
being some of the economic value of these species, some<br />
leading to economic losses and there are also species<br />
that affect human health. Mediterranean's geological and<br />
environmental conditions, there is a continuous change<br />
within and it also leads to change of biota quickly since<br />
the day it ever existed.<br />

TRANSLATIONAL MEDICINE<br />
Son 30 yılda akciğer kanserinin hücresel ve moleküler<br />
biyolojide genetiğini anlamak için bir çok bilimsel çalışma<br />
yapılmasına rağmen hala akciğer kanserinden dolayı ölüm<br />
oranı diğer kanserlerin oranıdan daha yüksektir. ( ve<br />
şüphesiz Türkiye’de de). Tütün kullanımından kaçınma<br />
hastalıktan korunmak için bilinen tek yol olsada hiçbir<br />
tarama methodu etki sağlamamıştır. Sınırlı sayıda hedefe<br />
yönelik terapiler başarılı sonuçlar vermiştir ve radyo ve<br />
kemoterapiye verilen tepkiler uzun ömürlü değildir ve<br />
anlaşılması zordur. Tüm bu faktörler genellikle tanıdan<br />
sonra kısa bir yaşam beklentisine yol açar.<br />
Genetik mutasyon sıralamasında büyük farklar yansıtan<br />
histopatolojik özelliklerine bağlı olarak akciğer kanseri<br />
başlıca iki gruba ayrılabilir; küçük hücreli olmayan akciğer<br />
kanseri (NSCLC) ve küçük hücreli akciğer kanseri (SCLC).<br />
Çeşitli akciğer kanseri tiplerinin başlangıç ve ilerlemesinin<br />
moleküler yönden idare edilişinin anlaşılmasıyla bilimsel<br />
gelişmeler sağlandı. Hedeflenen terapatik müdahaleler<br />
için kendi potansiyelleri vurgulanarak akciğer kanserinde<br />
bulunan başlıca genetik lezyonlar sunulacak. Akciğer<br />
kanserinin moleküler ve hücresel süreçlerini anlamamız<br />
fare modellerini (GEMMs) genetik olarak geliştirmemize<br />
olanak sağladı. Bu modeller (GEMMs) küçük hücreli ve<br />
küçük hücreli olamayan akciğer kanser tiplerinin ikisini de<br />
yakından taklit eder ve akciğer kanserinin kökeni olan<br />
muhtemel hücreleri tanımlamamıza olanak sağlar. Akciğer<br />
kanser onarımında kanser hücrelerinin kökeninin yerinin<br />
belirlenmesi ve onların olası rolleri ele alınacak. Ancak fare<br />
modelleri (GEMMs) ve alınan kültürlenmiş klonal fare<br />
tümör hücreleri insan akciğer kanseririn karmaşık tümör<br />
heterojenliğini tam yansıtamadı. Bu nedenle insan akciğer<br />
kanser dokusuna giriş ve canlı tümör dokusunu korunması<br />
için Hasta Türetilmiş ksenotransplant (pdx) modeller<br />
yaratma yolunda açık bir ihtiyaç kalır. Pdx modeller klonal<br />
ayrışmayı çalışmak aynı zamanda da birincil akciğer kanser<br />
tedavisinin yanıtını çalışmak içinde idealdir. Akciğer<br />
kanseri çalışmasındaki başka bir güncel gelişme, erken<br />
tanı ve akciğer kanseri hastalarının tedavi yanıtı ve<br />
longitudinal analizi ile sıvı biyopsi kullanımında ortaya<br />
çıktı.<br />
Although in the last 30 years much scientific progress has<br />
been made in understanding some of the genetics,<br />
molecular and cellular biology of lung cancer. This did not<br />
result in lowering the mortality rate for lung cancer which<br />
is still the highest for any cancer around the world (and<br />
certainly for Turkey). Avoidance of the use of tobacco is<br />
the only known way to prevent the disease whereas no<br />
screening method has proved effective. Limited amount of<br />
targeted therapies have been proven to be successful and<br />
responses to radio/chemotherapy are not durable and are<br />
difficult to understand. All these factors often lead to a<br />
short life-expectancy after diagnosis.<br />
Lung cancer can be divided in two main sub groups, Non-<br />
Small Cell Lung Cancer (NSCLC) and Small Cell Lung<br />
Cancer (SCLC), depending on its histopathological<br />
characteristics that reflect major differences in their<br />
respective range of genetic mutations. Scientific progress<br />
has been made in understanding the molecular pathways<br />
governing the onset and progression of the various lung<br />
cancer types. An overview of major genetic lesions found<br />
in lung cancer will be presented with an emphasis on their<br />
potential for targeted therapeutic intervention. Our better<br />
understanding of molecular and cell biological processes<br />
in lung cancer enabled us to develop geneticallyengineered<br />
mouse models (GEMMs). These GEMMs for<br />
both NSCLC and SCLC closely mimic human lung cancer<br />
and enabled us to identify the probable cells of origin for<br />
lung cancer. The nature of these lung cancer cells of origin<br />
will be addressed as well as their putative role in lung<br />
cancer maintenance.<br />
However, GEMMs and the derived cultured clonal mouse<br />
tumor cell lines cannot fully reflect the complex tumor<br />
heterogeneity of human lung cancer. A clear need remains<br />
therefore to access primary human lung cancer tissue and<br />
preserve the living tumor tissue by way of creating Patient-<br />
Derived Xenotransplant (PDX) models. The PDX models<br />
are ideally suited to study clonal divergence but also<br />
therapy response of primary human lung cancer. Another<br />
recent development in the study of lung cancer has<br />
emerged in the use of “liquid biopsies” for early diagnosis<br />
and longitudinal analyses of therapy response from lung<br />
cancer patients.<br />

Basically, patient-blood is collected to isolate free<br />
circulating tumor DNA (ctDNA) samples or exosomes<br />
and/or platelets which contain absorbed tumor RNA.<br />
Either the ctDNA or platelet or exosome RNA can be<br />
subjected to next generation sequencing methods that<br />
are sensitive enough to detect changes in the level and<br />
contents of isolated ctDNA or RNA. The latter might<br />
thus reflect the occurrence of lung cancer lesions in a<br />
patient as means for early diagnosis. In case of<br />
longitudinal sampling during therapy of a lung cancer<br />
patient, changes in gene expression and genetic<br />
lesions can be determined that govern a specific<br />
therapy response.<br />
Summarizing, the character of current lung cancer<br />
research at the IBG will be presented:<br />
1) Use of GEMMs for the study of basic lung cancer<br />
biology, testing and refining of targeted therapies<br />
against lung cancer<br />
2) Developing human lung cancer PDX models with an<br />
emphasis on SCLC for mimicking human lung cancer in<br />
a therapy model derived from each single patient<br />
3) Applying new and innovative techniques of “liquid<br />
biopsies” for the discovery of new diagnostic and<br />
prognostic markers for lung cancer therapy response.<br />
Accordingly, aim of our ongoing research is to gain<br />
insight into some of the molecular pathways and<br />
mechanisms that refer rapid resistance to radiochemotherapy<br />
of drug sensitivity of the different<br />
(N)SCLC cell populations. We will also know if intrinsic<br />
drug resistant populations are already harbored inside<br />
primary (N)SCLC or if they are de novo originated inside<br />
relapsed lesions only. Finally, our PDX models can be<br />
used for follow-up studies for (N)SCLC biology and<br />
therapy strategies, both for us and the scientific<br />
community.<br />
In conclusion, our studies should provide us with a<br />
better understanding of the phenotypic complexity of<br />
lung cancer and has implications for comprehending<br />
the underlying molecular and cellular mechanisms that<br />
govern lung cancer drug resistance. Moreover, our new<br />
PDX models will be of valuable use for extensive drug<br />
screens and should thereby lead to discover new<br />
candidate genes that control some of these pivotal<br />
molecular pathways. Our combined results should<br />
therefore improve the design of new, more efficient<br />
targeted- and chemo- therapeutic approaches against<br />
lung cancer.<br />
Serbest dolaşan tümör DNA’sını izole etmek için tümör<br />
RNA’sını absorbe etmiş olan örnekler, eksozomlar yada<br />
trombositler içeren hasta kan toplandı. Hem ctDNA<br />
hemde trombositler ya da eksozomlar, değişiklikleri<br />
belirlemek için yeterli hassasiyette olan ve izole edilmiş<br />
ctDNA ve RNA’ları içeren gelecek nesil dizileme<br />
tekniklerine tabi tutulabilir. Son olarak, belki böylece<br />
erken tanı aracı olarak bir hastada akciğer kanseri<br />
lezyonları oluşumunu yansıtıyor olabilir. Boylamsal<br />
örnekleme olması halinde, akciğer kanseri hastasının<br />
tedavisi sırasında, gen ekspresyonlarındaki değişiklik ve<br />
genetik lezyonlar belirli bir tedavi yanıtı olarak<br />
tanımlanabilir.<br />
Özetle, İBG’de yapılan akciğer kanser araştırmasının<br />
özellikleri şöyle sunulacak;<br />
1) Temel kanser biyolojisinin çalışılması için model<br />
farelerin (GEMM) kullanımı, test edilmesi ve akciğer<br />
kanserine karşı hedeflenen tedavileri arıtma<br />
2) Her bir hastadan elde edilen terapi modellerinde, insan<br />
akciğer kanserinin taklit edilmesi için SCLC üzerinde<br />
durularak pdx modelleri geliştirme<br />
3)Akciğer kanserinde yeni tanı ve prognostik belirteçlerin<br />
keşfi için sıvı biyopsinin yeni ve inovatif tekniklerini<br />
uygulama<br />
Buna bağlı olarak, çalışmamızın temel hedefi farklı<br />
(N)SCLC hücre popülasyonlarının ilaca hasas radiokemoterapi’de<br />
ortaya çıkan hızlı direncin, moleküler<br />
mekanizmalarının ve yolaklarının aydınlatılması<br />
amaçlanmaktadır. Ayrıca ilaca dirençli esas<br />
populasyonların öncül (N)SCLC’de veya eğer de novo<br />
kökenli ise nükseden bölgelerin içerisinde olduğunu<br />
biliyoruz. Son olarak, PDX modellerimiz hem bizim hemde<br />
bilim dünyasında (N)SCLC biyolojisi çalışmalarında veya<br />
terapi stratejisi geliştirmede kullanabilmektedir.<br />
Sonuç olarak, çalışmalarımız bize akciğer kanserinin<br />
fenotipik kompleksisitesi ve ilaçlara dirençli akciğer<br />
kanserinin altında yatan moleküler ve hücresel<br />
mekanizmaları yöneten yolakları anlamamıza olanak<br />
sağlamaktadır. Dahası, yeni oluşturulan PDX modelleri,<br />
kapsamlı ilaçların gösterilmesi ve bazı önemli moleküler<br />
yolaklarını kontrol eden yeni aday genlerin bulunmasında<br />
değerli bir uygulama yöntemi olabilecektir.<br />
Çalışmalarımızın sonuçları altında, akciğer kanserine<br />
karşı yeni,daha hedef etkili kemo-terapotik ilaçların<br />
geliştirilmesini sağlayacaktır.<br />

Mitokondriyal hastalıklar, her yıl 5000 doğumda 1 görülen<br />
klinik olarak heterojen bir hastalık grubudur. Mitokondriyal<br />
hastalıkların altında yatan en sık sebep Mitokondriyal<br />
Oksidatif Fosforilasyon (OXPHOS) yolağında görülen bir<br />
bozukluk olup, nöromüsküler veya kardiyovasküler<br />
semptomlara yol açar. Nüklear veya Mitokondriyal DNA da<br />
meydana gelen çoklu mutasyonlar OXPHOS sisteminin<br />
çalışmasını etkiler. Bu sebeple klinik açıdan şeffaf<br />
olmayan genetik bir hastalık fenotipini oluşturur.<br />
Metot ve Sonuçlar<br />
Şüpheli Mitokondriyal bozuklukları olan hipertrofik<br />
kardiyomiyopatisi olan hastaların (HCM) toplam boyutu<br />
16.6kb baz çiftlik mtDNA’sı iki uzun menzilli PCR(LR-PCR)<br />
fragmanları ve ek destek biyokimya verileri ile taranmıştır.<br />
Hedeflenen gen dizisinin ve alt varyant analizinin başarısı<br />
veya tanı verimi, teşhislerde kullanılan standart araçlar ile<br />
ekzom dizileme veya ek belirli kriterlerine bağlı kalınmıştır.<br />

Introduction<br />
Mitochondrial disorders are a clinically heterogeneous<br />
group of disorders that have a prevalence of 1 in 5000<br />
newborns each year. Dysfunction of the mitochondrial<br />
oxidative phosphorylation (OXPHOS) pathway, which<br />
underlies mitochondrial disease most often leads to<br />
neuromuscular or cardiovascular symptoms. Multiple<br />
mutations in the mitochondrial DNA or nuclear DNA can<br />
affect proper functioning of the OXPHOS, which results in<br />
an unclear complex genetic clinical phenotype.<br />
Methods and Results<br />
Hypertrophic cardiomyopathy patients (HCM) with<br />
suspected mitochondrial disorders and additional<br />
supportive biochemistry data are first screened for<br />
targeted mtDNA, by two Long Range PCR (LR-PCR)<br />
fragments covering the complete size of 16.6kb base<br />
pairs. The success or diagnostic yield of targeted gene<br />
sequencing and downstream variant analysis are<br />
depended on standard tools in diagnostics with<br />
additional specific criteria or strategy for exome<br />
sequencing.<br />

Exome sequencing in a consanguineous family<br />
revealed a novel homozygous splice site mutation in a<br />
recent discovered QRSL1 gene for causing infantile<br />
hypertrophic cardiomyopathy. The effect of the splicing<br />
was determined and caused a complete and partial<br />
skipping of exon 8, which resulted in a loss of function<br />
within this protein. QRSL1 or glutaminyl-mt tRNAGln<br />
amidotransferase protein is essential for human<br />
mitochondrial translation proteins. In two other nonconsanguineous<br />
families we found new compound<br />
heterozygous mutations in mitochondrial aminoacyltRNA<br />
synthetases genes AARS2 and MTO1, which were<br />
filtered in our data for mitochondrial gene panels and<br />
resulting in HCM within newborns.<br />
Conclusion<br />
We report the first consanguineous family with a causal<br />
homozygous splicing defect of QRSL1. This recent<br />
novel gene is related to bioenergetics defects involved<br />
in glycolysis, as well in mitochondrial respiration<br />
deficiencies on patient derived cell lines. Finally, we<br />
included also new additional relevant mutations for<br />
AARS2 and MTO1, both already known pathogenic<br />
HCM genes. Nevertheless, we improved this<br />
downstream NGS variant analysis by introducing<br />
zebrafish or rodent models, including integrated<br />
systems biology and metabolomics data.<br />
Akraba aile içinde yapılan ekzom sekanslaması, yeni<br />
keşfedilen ve infantil hipertrofik kardiyopiyotiye sebep<br />
olan QRSL1 geninde yeni bir homozigot eklenme<br />
bölgesinde mutasyon olduğunu ortaya çıkarmıştır. Ekleme<br />
bölgesinin etkisi, 8. eksonda gerçekleşen tam veya kısmı<br />
bir atlama ile kendini göstererek protein yapısında<br />
fonksiyon kaybına neden olduğu tanımlanmıştır. QRSL1<br />
veya glutaminil-mt tRNAGln amidotransferaz proteini,<br />
insan mitokondriyal translasyon proteinleri için gereklidir.<br />
Akraba olmayan iki ailede ise, mitokondriyal aminoaçiltRNA<br />
sentetaz genleri olan AARS2 ve MTO1 genlerinde<br />
yeni, bileşik bir mutasyon bulundu ve mitokondriyel gen<br />
panellerinde elde edilen veriler taranarak yeni doğanlarda<br />
HCM ile sonuçlandığı görüldü.<br />
Tartışma<br />
İlk akraba ailede homozigot ekleme bozukluğu QRSL1'in<br />
fonksiyon bozukluğundan kaynaklanmaktadır. Yeni açığa<br />
çıkan bu gen, hasta kaynaklı hücre soylarının glikoliz<br />
yolağındaki biyoenerjetik bozukluklar ve mitokondriyal<br />
solunum yetersizlikleri ile ilgilidir. Son olarak, patojenik<br />
olarak önceden bilinen HCM genleri olan AARS2 ve<br />
MTO1'e ek olarak yeni alakalı mutasyonlar dâhil<br />
edilmiştir. Yine de, biz zebra balığı ve kemirgen modelleri<br />
kullanarak, alt NGS (yeni nesil dizileme) varyentlerini<br />
sistem biyolojisi ve metabolomik verilerini kullanarak<br />
iyileştirmiş bulunmaktayız.<br />

‘PRECİSİON MEDİCİNE’<br />
Farmakogenetik bir hastanın ilaç tedavisine yanıtını açıklamak<br />
ya da öngörebilmek amacıyla DNA analizi yapılmasına dayanan<br />
bir uygulamadır. Günümüzde uygulanan ilaç tedavileri herkese<br />
aynı şekilde doz ayarlaması yapılmaksızın “deneme-yanılma”<br />
yolu ile gerçekleştirilmektedir. Farmakogenomik testler<br />
“genotipleme” yani ilaç metabolizmasında değişik fenotiplere<br />
(ilaçların metabolize edilme yetisi) neden olan enzimleri<br />
kodlayan genlerdeki ya da ilaçların etki ettiği hedef moleküller ve<br />
iyon kanallarını şifreleyen genlerdeki varyasyonların belirlenmesi<br />
esasına dayanır.<br />
Hastane yatışlarının %5-7’sinin advers ilaç reaksiyonlarına (ADR)<br />
bağlı olarak gerçekleştiği bilinmektedir. Tüm ölüm nedenleri<br />
arasında ADR’ları beşinci sırada yer almaktadır. Tedavi amaçlı<br />
kullanılan tüm ilaçların sadece %25-60’ının etkili tedavi<br />
sağladığı bilinmektedir. Böylece tedavi altında olan kişilerin<br />
%40-75 oranında ilaçlardan faydalanamadığı bir gerçektir.<br />
Hastaların tedaviye başlamadan önce farmakogenomik test<br />
yaptırmaları, yanlış ilaç kullanımı nedeni ile fayda<br />
sağlanamayacak ya da yan etki oluşturacak komplikasyonları<br />
%80 oranında azaltabilecektir.<br />
Pharmacogenetics is a techic that based on DNA analysis in<br />
order to describe or predict a patient’s response to a drug<br />
therapy. Nowadays the drug treatments are carried out to<br />
everyone in the same way without dose adjustment “by trial<br />
and error”. Pharmagenomics tests which<br />
means“genotyping”, is based on determination of gene<br />
variations;which encodes enzymes that couse of different<br />
phenotype on drug metabolism or drugs that have an effect<br />
on target molecules and genes that encodes ion channels.<br />
5-7 percent of hospitalization cases are known to occur due<br />
to adverse drug reactions(ADR). ADRs are the fifth most<br />
common couse of death, among all of it. All drugs used for<br />
treatment is known to provide effective treatment of only<br />
25-60%. Thus, verify the fact of %40-75 of patients that<br />
under treatment can not benefit from drugs. The<br />
phamacogenomic test before starting treatment, can<br />
reduce pass over due to usage of wrong drug or<br />
complications that can arise from side effects up to %80.<br />

80 percent of drugs used in treatment of complex<br />
diseases are metabolized by cytochrome P450<br />
enzyme(Figure 1). As seen in the Figure 1, oncology drugs<br />
used in cancer treatment and drugs that used for<br />
treatment of psychiatric disorders are metabolized by<br />
CYO3A4 gene product that correspond to %36 of all<br />
drugs. Therefore, the first application of<br />
pharmacogenomics in routine health care system has<br />
been in cancer and psychiatry. in our country and the<br />
world, excess frequency of the complex diseases such as;<br />
cardiovascular diseases, type 2 diabetes, B-<br />
blockers(CYP2D6) used in treatment of hypertension,<br />
anticoagulants(warfarin,<br />
acenacoumarol),<br />
clopidogrel(plavix) and seen at a rate of %45 of proton<br />
pump inhibitors has revealed the necessity of the usage of<br />
the pharmagenetics in complex diseases too.<br />
Antidepressanta and B-blockers can be metabolized ultra<br />
fast, medium or slow, due to the person’s CYP2D6<br />
genotype. In parallel with, after the pharmagenomics test<br />
performed, advers side effects can eleminate by<br />
personalized dosage adjustment and also it will help<br />
person to benefit from drug(Figure 2).<br />
Kompleks hastalıkların tedavisinde kullanılan ilaçların<br />
%80’i sitokrom P450 enzimleri tarafından metabolize<br />
edilmektedir (Şekil 1). Şekil 1’de görüldüğü üzere kanser<br />
tedavisinde kullanılan onkoloji ilaçları ve psikiyatrik<br />
bozuklukların tedavisinde kullanılan ilaçlar, tüm ilaçların<br />
%36’sına karşılık gelecek şekilde CYP3A4 gen ürünü<br />
tarafından metabolize edilmektedir. Bu nedenle<br />
farmakogenomiğin sağlık sisteminde rutin olarak ilk<br />
uygulamaları kanser ve psikiyatride olmuştur. Dünyada ve<br />
ülkemizde görülme sıklığı çok fazla olan kompleks<br />
hastalıklardan kardiyovasküler, tip 2 diyabet,<br />
hipertansiyon tedavisinde kullanılan β blokerler<br />
(CYP2D6), antikoagülanlar (warfarin, acenacoumarol),<br />
klopidogrel (plavix), proton pompa inhibitörlerinin ise %<br />
45 gibi bir oranda görülmesi farmakogenetiğin kompleks<br />
hastalıklarda da kullanılması gerekliliğini ortaya<br />
koymuştur. Antidepresanlar ve β blokerlerin kişinin<br />
CYP2D6 genotipine göre ultra hızlı, orta derecede veya<br />
yavaş olarak metabolize edilmesine bağlı olarak<br />
farmakogenomik test sonrası kişiye özel doz ayarının<br />
yapılarak kullanılması advers yan etkileri ortadan<br />
kaldıracağı gibi kişinin ilaçtan yararlanımını da<br />
sağlayacaktır (Şekil 2).<br />
