CytoSeeing - Reversible cytoplasm blue fluorescent dye

CytoSeeing is a newly developed fluorescent dye, which can stain cytoplasm promptly by just

adding to culture medium. After observation, fluorescent dye can be easily removed by replacing

medium which does not contain CytoSeeing. It is well known that morpholgy change of nuclear

and cytoplasm are related to differentiation, function and signal response of cells. Conventional

cytoplasm staining dyes are used for cell tracking. Therefore, once dyes are incorporated into

cells, the dyes remain in cytoplasm even after medium change.

The dyes are diluted in the course of cell division, and disappear after 3 to 6 population doubling.

If dyes remained in the cells, it is difficult to observe the cell with other probes after live cell

imaging. CytoSeeing overcomes this conventional problem. 4

AcroleinRED - Cell-based Acrolein Detection Reagent

Product Background

Acrolein, the simplest α, ß-unsaturated aldehyde (chemical name; 2-propenal), is highly toxic metabolite for

cells. Acrolein is found in a common dietary and environmental pollutant such as tobacco smoke. Acrolein is

also endogenously generated in cells, especially under the oxidative stress condition. Enzymatic oxidation of

threonineand polyamine species, reactive oxygen species (ROS)-mediated oxidation of highly unsaturated lipids

are majorsource of acrolein in cells. Recent evidences indicate acrolein is more toxic to cells than ROS such as

hydrogenperoxide (H2O2) or hydroxyl radicals. Because of its highly reactivity, acrolein in the cell immediately

reacts with various biomolecules such as DNA/RNA and proteins, and subsequently impairs their functions.

Establishment of the detection methods of cellular acrolein is an important issue to understand physiological

relevance of acrolein. AcroleinRED is the world first cell-based acrolein-detection reagent under the live

cellcondition without any pre-treatment and cell lysis. This reagent is based on phenylazide-acrolein click

chemistry discovered by Drs. Katsunori Tanaka and Ambara Pradipta (Ref.1). AcroleinRED specifically react with

either extracellular acrolein released from cell surface lipids or intracellular acrolein generated via enzymatic

pathway and label acrolein with TAMRA fluorophore. In the case of extracellular labeling, TAMRA-labeled

acrolein are immediately incorporated into cells through the endocytosis pathway. The TAMRA-labeled acrolein

react with biomolecules such as a protein and stay in the cells (Figure 1).

Ap3, SHG Imaging Dye

Ap3 is a newly synthesized novel molecule for second harmonic generation

(SHG) imaging. SHG is a nonlinear optical process, the interaction of two

photons with a nonlinear material generates photons with twice the energy.

SHG imaging is a powerful tool to visualize cell and tissue structure and

function. However, most of dyes for SHG imaging emit strong fluorescence

signals in addition to SHG signals. The fluorescence emission from dyes

disturbs multimodal imagining as well as SHG signal imaging. Ap3 is the first

novel dye designed specifically for SHG imaging with virtually no fluorescence

signals, improved photostability and less phototoxicity. Ap3 enables us to

detect true SHG signals and realizes multimodal imaging.

LiveReceptor AMPAR - Endogenous AMPAR Labeling Reagent

Neurotransmitter receptors including glutamate receptors and GABA

receptors etc. located on post-synapse in neuronal cells play various

roles in brain functions. To understand physiological roles of

neurotransmitter receptors, live cell imaging is one of the powerful

approaches. Conventional imaging methods on live cellsrelay on a

genetically engineered proteins fused with fluorescent proteins such

as GFP. However, one serious problem is that the functions and

movement of over-expressed neurotransmitter receptors with non-physiological tags are not precisely

correlated with endogenous native receptors. The labeling methods for endogenous receptors are desirable to

observe physiological functions of receptors. LiveReceptor is the world first reagent series for target-specific

receptor labeling. The principle of LiveReceptor is based on ligand-directed acyl imidazole (LDAI) chemistry

(ref.1,2). LDAI-based chemical labelling is driven by selective ligand-protein recognition, which facilitates an

acyl substitution reaction of labeling reagents on nucleophilic amino acid residues including Lys, Ser and Tyr

located near ligand-binding domain. After wash out, the labelled receptors which have free ligand-binding

pockets are observed on live cells.

PolyamineRED - Intracellular Polyamine Detection Reagent

PolyamineRED is a reagent for imaging

intracellular polyamines specifically without any

pretreatment or cell lysis. This can be used for

live cells, therefore it is suitable for detection or

semi-quantitative analysis of intracellular

polyamines.

The polyamine species, including putrescine,

spermidine and spermine etc. and its acetyl

derivatives, are one of the essential class of

metabolites which have liner alkyl structure

with two or more amines. Polyamines are found

in all living organisms with high concentration,

from sub-millimolar to millimolar, in the cells.

Polyamines have polycationic properties and

shows an enormous number of biological functions. For example, polyamines interact with DNA/RNA in the

nuclear and regulate gene expression. Polyamines also interact with negatively charged proteins and control its

function. The major source of polyamines is an amino acid ornithine. In the case of mammalian, ornithine is

converted to putrescine by ornithine decarboxylase (ODC), followed by synthesizing spermindine and

spermine. Because ODC is highly expressed in cancer cells, polyamines are considered as one of the cancer

marker.
