a. Acetólisis <strong>de</strong> Erdtman (2).<br />

Se colocaron las anteras completas<br />

fijadas previam<strong>en</strong>te <strong>de</strong>ntro <strong>de</strong> un<br />

tubo <strong>de</strong> c<strong>en</strong>trifugación con ácido acético<br />

glacial puro y se c<strong>en</strong>trífugo tres<br />

veces a 2000 rpm durante 3 minutos.<br />

Se preparó la mezcla acetolítica utilizando<br />

para ello ácido acético anhidro<br />

y ácido sulfúrico (9:1). Gradualm<strong>en</strong>te<br />

se fueron uni<strong>en</strong>do ambos solutos por<br />

las pare<strong>de</strong>s <strong><strong>de</strong>l</strong> recipi<strong>en</strong>te. Se incluyó<br />

el material vegetal <strong>en</strong> la mezcla<br />

acetolítica recién preparada y se colocó<br />

<strong>en</strong> baño <strong>de</strong> María, parti<strong>en</strong>do <strong>de</strong><br />

frío hasta ebullición por 5 a 10 minutos.<br />

Se <strong>de</strong>jó reposar hasta <strong>en</strong>friar y se<br />

c<strong>en</strong>trífugo a 2000 rpm por 3 minutos.<br />

Se <strong>de</strong>cantó el sobr<strong>en</strong>adante y el residuo,<br />

<strong>en</strong> el fondo <strong><strong>de</strong>l</strong> tubo, se lavó con<br />

ácido acético glacial puro y se c<strong>en</strong>trífugo<br />

por tres veces a 2000 rpm durante<br />

3 minutos. Se lavó dos veces con<br />

agua <strong>de</strong>stilada y se <strong>de</strong>cantó. El material<br />

se sometió a un proceso <strong>de</strong> <strong>de</strong>shidratación<br />

<strong>en</strong> una batería <strong>de</strong> alcohol<br />

etílico, parti<strong>en</strong>do <strong>de</strong> 70 hasta 100%,<br />

cada paso con una duración <strong>de</strong> 3 minutos<br />

y <strong>de</strong>cantando los alcoholes con<br />

precaución. Se agregó b<strong>en</strong>c<strong>en</strong>o y se<br />

<strong>de</strong>jó reposar por 5 minutos, se c<strong>en</strong>trífugo<br />

a 2000 rpm y nuevam<strong>en</strong>te se <strong>de</strong>jó<br />

reposar. Se preparó una mezcla<br />

b<strong>en</strong>c<strong>en</strong>o: aceite <strong>de</strong> silicón (1:1); se colocó<br />

el material vegetal <strong>en</strong> <strong>en</strong>vases<br />

pequeños y se le añadió la mezcla. Se<br />

<strong>de</strong>jó reposar por 24 horas, <strong>de</strong>stapado<br />

a fin <strong>de</strong> facilitar la evaporación <strong><strong>de</strong>l</strong><br />

b<strong>en</strong>c<strong>en</strong>o. Para las observaciones al<br />

microscopio óptico, se colocó una gota<br />

<strong><strong>de</strong>l</strong> preparado <strong>en</strong> un portaobjetos, se<br />

cubrió con una cubreobjeto y se esperó<br />

el tiempo necesario para que se<br />

ext<strong>en</strong>diera la muestra <strong>de</strong> <strong>grano</strong>s <strong>de</strong><br />

<strong>pol<strong>en</strong></strong>.<br />

<strong>en</strong>d of tube was cleaned with pure glacial<br />

acetic acid and it was c<strong>en</strong>trifuged<br />

three times at 2000 rpm during 3<br />

minutes. It was cleaned twice with<br />

distilled water and <strong>de</strong>canted. Material<br />

was submitted to a <strong>de</strong>hydration<br />

process in a ethylic alcohol battery,<br />

from 70 to 100%, each path with 3<br />

minutes of duration and by <strong>de</strong>canting<br />

alcohols with a lot of prev<strong>en</strong>ting.<br />

B<strong>en</strong>z<strong>en</strong>e was ad<strong>de</strong>d and left at rest<br />

during 5 minutes. It was c<strong>en</strong>trifuged<br />

at 2000 rpm and left at rest again. A<br />

b<strong>en</strong>z<strong>en</strong>e mix was prepared: silicon oil<br />

(1:1); vegetable material was located<br />

at small recipi<strong>en</strong>ts and mix was<br />

ad<strong>de</strong>d. It left at rest during 24 hours,<br />

without tape for making easier the<br />

b<strong>en</strong>z<strong>en</strong>e evaporation. For<br />

observations to optical microscope, a<br />

drop of preparation was placed on<br />

microscope sli<strong>de</strong>s and covered with<br />

other one and time required for grain<br />

sample being ext<strong>en</strong><strong>de</strong>d<br />

b. Potassium hydroxi<strong>de</strong><br />

treatm<strong>en</strong>t (KOH) (2).<br />

Anther was inclu<strong>de</strong>d in KOH at<br />

10% and placed in Water Bath.<br />

Preparation was left at rest and it was<br />

cleaned with distilled water.<br />

Vegetable material was <strong>de</strong>hydrated<br />

by passing through an ethylic alcohol<br />

battery (70, 95 and 100%), each path<br />

of 5 minutes. It was c<strong>en</strong>trifuged at<br />

2000 rpm three times and during 3<br />

minutes each time and being <strong>de</strong>canted<br />

carefully in each path. Vegetable material<br />

was placed in b<strong>en</strong>z<strong>en</strong>e during 5<br />

minutes and it was <strong>de</strong>canted. It was<br />

covered with a mix of b<strong>en</strong>z<strong>en</strong>e:silicon<br />

in proportion of 1:1. It was left at rest<br />

for 24 hours or until b<strong>en</strong>z<strong>en</strong>e was<br />

evaporated. For optical microscope<br />

(OM) observation, drops of<br />
