INTERFERENCE STUDIES<br />
Vaginal infections, urine, semen, and small blood admixtures do<br />
not interfere with the results of <strong>PartoSure</strong>. These substances and<br />
conditions have been evaluated in nonclinical studies and were found to<br />
contain PAMG-1 in concentrations only below the detection threshold<br />
of <strong>PartoSure</strong>.<br />

<strong>PartoSure</strong> TTD (Time to Delivery) Test<br />
INTENDED USE<br />
The <strong>PartoSure</strong> TTD (Time to Delivery) Test is a rapid, non-instrumented,<br />
qualitative immunochromatographic test for the in vitro detection<br />
of placental alpha microglobulin-1 (PAMG-1) in vaginal secretions of<br />
pregnant women. The test is for use by healthcare professionals to<br />
aid in the prediction of time to delivery when patients report signs,<br />
symptoms or complaints suggestive of labor between 20 and 37 weeks<br />
of gestation with clinically intact membranes.<br />
SUMMARY AND EXPLANATION OF THE TEST<br />
An accurate prediction of time to delivery is clinically important among<br />
pregnancies at risk for preterm delivery. This is particularly true with<br />
respect to both the administration of corticosteroids, which have an<br />
optimal bene t within 7 days of administration, 2 as well as the transfer<br />
of patients to a tertiary care center capable of caring for the birth of a<br />
premature infant.<br />
Clinical evaluation alone, including the measurement of cervical length<br />
and dilatation, is not su ciently predictive of imminent delivery. 3<br />
Similarly, currently available biomarker tests, such as that based on the<br />
detection of fetal bronectin, have extremely poor predictive values for<br />
imminent delivery. 4 Thus, the accurate prediction of time to delivery in<br />
patients with threatened preterm labor remains an important clinical<br />
concern. The <strong>PartoSure</strong> TTD Test is a rapid, non-invasive strip test<br />
with an unprecedented predictive value for delivery within 7 days in<br />
patients presenting with signs and symptoms of preterm labor. The<br />
high negative and positive predictive values of the <strong>PartoSure</strong> TTD<br />
Test may allow for the timely identi cation of patients that would<br />
bene t from available interventions such as antenatal corticosteroids.<br />
Additionally, a more accurate identi cation of these patients will likely<br />
reduce the costs associated with unnecessary admissions caused by<br />
the poor positive predictive values of existing methods. The test kit is<br />
a self-contained system that provides rapid, qualitative results without<br />
outside instrumentation or an invasive speculum examination.<br />
PRINCIPLE OF THE TEST<br />
<strong>PartoSure</strong> is a lateral ow, immunochromatographic assay designed<br />
to identify the presence of human placental-alpha-microglobulin-1<br />
(PAMG-1), which is detectable in cervicovaginal exudates when delivery<br />
is imminent. The test employs monoclonal antibodies su ciently<br />
sensitive to detect 4 pg/μl of PAMG-1 in cervicovaginal discharge.<br />
PAMG-1, a protein released from decidual cells into the amniotic cavity<br />
throughout pregnancy, is present in cervicovaginal discharge when<br />
labor and delivery are imminent. This is likely due to the transudation<br />
of the protein through preexisting pores in the chorioamniotic<br />
membranes during uterine contractions and, potentially, degradation<br />
of the extracellular matrix of fetal membranes due to an in ammatory<br />
process of labor or infection. Its presence in cervicovaginal secretions<br />
indicates that delivery is likely to occur within 7 days and may be<br />
associated with a signi cantly increased risk of intra-amniotic infection<br />
and/or in ammation. 1 Its absence indicates that delivery is not likely to<br />
occur within 7 days or 14 days.<br />
For the analysis, a sample of cervicovaginal discharge collected by<br />
vaginal swab is extracted into a solvent. The presence of PAMG-1<br />
antigen is then detected by inserting a lateral ow test strip into the vial.<br />
The sample ows from an absorbent pad to a nitrocellulose membrane,<br />
passing through a reactive area containing monoclonal anti-PAMG-1<br />
antibodies conjugated to a gold particle. The antigen-antibody complex<br />
ows to the test region where it is immobilized by a second anti-PAMG-1<br />
antibody. This event leads to the appearance of the test line. Unbound<br />
antigen-antibody complexes continue to ow along the test strip and<br />
are immobilized by a second antibody. This leads to the appearance of<br />
the internal control line.<br />
REAGENTS AND COMPONENTS<br />
The <strong>PartoSure</strong> test kit includes the following components: <strong>PartoSure</strong><br />
test strip in foil pouch with desiccant, sterile ocked vaginal swab, and<br />
plastic vial with solvent solution (0.9% NaCl, 0.05% NaN3, 0.01% Triton<br />
X100).<br />
PRECAUTIONS AND WARNINGS<br />
<strong>PartoSure</strong> is for in vitro diagnostic use only and no component of<br />
the test should be taken internally • The instructions for use must<br />
be followed exactly; failure to do so may lead to inaccurate results •<br />
Safety precautions should be observed when collecting, handling, and<br />
disposing of test samples • Use of any swab other than the sterile ocked<br />
swab provided with the test kit will alter the performance of the test and<br />
is therefore strictly prohibited • Do not use damaged components of<br />
the test kit • Used test kits are biohazardous; dispose of all components<br />
using necessary precautions • Do not use the test after the expiration<br />
date, which is printed on the product pacakaging • Test kit components<br />
are for single-use only • Do not bend or fold the test strip or the foil<br />
pouch with the test strip in it; doing so may damage the strip and lead<br />
to inaccurate results<br />
STORAGE AND STABILITY<br />
Store the <strong>PartoSure</strong> test kit in a dry place at 4 to 24°C (40 to 75°F).<br />
The test should not be frozen. When stored in the foil pouch at the<br />
recommended temperature, the test is stable until the expiration date<br />
printed on pouch. The <strong>PartoSure</strong> test strip should be used within six (6)<br />
hours after removing the test strip from the foil pouch.<br />
QUALITY CONTROL<br />
The <strong>PartoSure</strong> test strip contains an internal procedural control<br />
mechanism that ensures analytical functionality. The appearance of one<br />
or two lines in the results region of the test strip veri es the integrity of<br />
the test procedure and components.<br />
LIMITATIONS OF THE TEST<br />
<strong>PartoSure</strong> test results are qualitative and not quantitative. No<br />
quantitative interpretation should be made based on the strength of<br />
the test or control lines. In cases of only trace amounts of blood on<br />
the swab, the test will function properly. When there is a signi cant<br />
presence of blood on the swab, the test can malfunction and its use<br />
is not recommended. <strong>PartoSure</strong> should not be used earlier than 6<br />
hours after the removal of any disinfectant solutions or medicines from<br />
the vagina. Results should be used in conjunction with other clinical<br />
information. Test performance in patients without signs or symptoms of<br />
labor is unknown. Placenta previa, and performing digital exams prior to<br />
sample collection can lead to inaccurate test results. The performance of<br />
<strong>PartoSure</strong> has not been established in the presence of the following<br />
contaminants: meconium, anti-fungal creams, suppositories, lubricants,<br />
moisturizers, talcum powders, or baby oil.<br />
EXPECTED VALUES<br />
<strong>PartoSure</strong> detects trace amounts of human PAMG-1. PAMG-1 present<br />
in cervicovaginal discharge in amounts greater than or equal to 4pg/<br />
μl indicates an elevated risk of delivering within the next 7 days.<br />
The sensitivity threshold of 4pg/μl was validated in an international<br />
multicenter clinical trial evaluating the association of PAMG-1 present in<br />
cervicovaginal secretions and the test-to-delivery time interval.<br />
PERFORMANCE CHARACTERISTICS<br />
The clinical performance of the <strong>PartoSure</strong> TTD test was determined<br />
in an international multi-site study. Patients who (i) reported signs,<br />
symptoms, or complaints suggestive of preterm labor, (ii) were between<br />
20 and 36 6⁄7 weeks of gestation, and (iii) had clinically intact membranes<br />
were invited to participate in the trial. In total, 101 patients were<br />
included in the nal analysis. The following performance estimates with<br />
corresponding 95% con dence intervals were calculated:<br />

CROSS REACTIVITY<br />
The following proteins were tested for cross reactivity with the<br />
monoclonal antibodies used in <strong>PartoSure</strong>: alpha-2- microglobulin<br />
of fertility, human chorionic gonadotropin, trophoblastic beta-<br />
1-glycoprotein, human placental lactogen, alpha-fetoprotein,<br />
human serum albumin, and several IGFBP proteins. The assayed<br />
monoclonal antibodies used in <strong>PartoSure</strong> were not cross-reactive<br />
with these proteins. The antibody used in the test line was found to<br />
be cross-reactive to the IGFBP-3 protein using the ELISA method. The<br />
concentration of IGFBP-3 in vaginal discharge of pregnant women,<br />
however, does not impact the accuracy of <strong>PartoSure</strong>.<br />

DIRECTIONS FOR IN VITRO DIAGNOSTIC USE<br />
The intensity of the lines may vary; the test result is valid even if the lines are faint or uneven. Do not interpret the test result based on<br />
the intensity of the lines. Do not read or interpret the results after 10 minutes have passed since inserting the test strip into the vial.<br />
TEST PROCEDURE<br />
1. Take the solvent vial by its cap and ensure all liquid in the vial has dropped to the bottom. Open the solvent<br />
vial and place it in a vertical position.<br />
2 To collect a sample from the vagina, use only the sterile ocked swab provided with the <strong>PartoSure</strong> test kit.<br />
Remove the swab from its package following the instructions on the packaging. The tip of the swab should not<br />
touch anything prior to insertion into the vagina. Hold the swab by the middle of its shaft and, while the patient<br />
is lying at on her back, carefully insert the tip of the swab into the vagina until the ngers contact the skin (no<br />
more than 5-7 cm deep). Withdraw the swab from the vagina after 30 seconds.<br />
3 After the swab has been removed from the vagina, immediately place the tip into the provided solvent vial<br />
and rinse by rotating for 30 seconds.<br />
4. Remove the swab from the vial and dispose of it .<br />
5. Tear open the foil pouch at the tear notches and remove the <strong>PartoSure</strong> test strip.<br />
6 Insert the white end of the test strip (marked with arrows facing downward) into the vial with solvent.<br />
7 Remove the test strip from the vial if two lines are clearly visible in the test region or after 5 minutes<br />
sharp. Read the results by placing the test strip on a clean, dry, at surface. A positive result is indicated by two<br />
lines in the test region, while a negative result is indicated by a single line in the test region. Do not read or<br />
interpret the results after 10 minutes have passed since inserting the test strip into the vial.<br />
PROCEDIMIENTO DEL TEST<br />

Sie ihn vorsichtig nur soweit in die Vagina ein, bis die Finger die Haut berühren (nicht tiefer als 5-7 cm). Warten<br />
Sie 30 Sekunden und ziehen Sie anschließend den Tupfer aus der Vagina heraus.<br />
3 Überführen Sie den Tupfer nach dem Herausziehen aus der Vagina umgehend in das Fläschchen mit<br />
Lösungsmittel und eluieren Sie die Probe, indem Sie den Tupfer durch Drehen in der Flüssigkeit für 30<br />
Sekunden spülen<br />
4. Entnehmen Sie anschließend den Tupfer aus dem Fläschchen und entsorgen Sie ihn.<br />
5. Reißen Sie den Folienbeutel entlang der Perforation auf und entnehmen Sie den <strong>PartoSure</strong> Teststreifen.<br />
6 Bringen Sie das weiß markierte Ende des Teststreifens (markiert mit nach unten weisenden Pfeilen) in das<br />
Fläschchen mit dem Lösungsmittel ein.<br />
7 Entnehmen Sie den Teststreifen aus dem Fläschchen, wenn zwei Banden deutlich sichtbar werden<br />
oder nach exakt 5 Minuten. Legen Sie den Teststreifen auf eine saubere, trockene und gerade Ober äche und<br />
lesen Sie das Testergebnis ab. Erscheinen in der Testregion zwei Banden, dann ist der Test positiv. Erscheint in<br />
der Testregion eine Bande, dann ist der Test negativ. Bewerten oder interpretieren Sie die Ergebnisse nicht nach<br />
Ablauf von 10 Minuten nach dem Einbringen des Teststreifens in das Fläschchen.<br />
<strong>PartoSure</strong> Schnelltest zur Prädiktion einer Frühgeburt<br />
BESTIMMUNGSGEMÄSSE VERWENDUNG<br />
Der <strong>PartoSure</strong> TTD (Time to Delivery) Test ist ein qualitativer<br />
immunchromatographischer Schnelltest für den in-vitro Nachweis des<br />
plazentalen alpha-Mikroglobulin-1 (PAMG-1) in vaginalen Sekreten<br />
schwangerer Frauen. Die Verwendung des Tests ist medizinischem<br />
Fachpersonal vorbehalten. <strong>PartoSure</strong> unterstützt die Vorhersage<br />
der Zeit bis zur Geburt bei Patientinnen zwischen der 20. und 37.<br />

ZUSAMMENFASSUNG UND ERLÄUTERUNG DES TESTS<br />
Bei Schwangerschaften mit dem Risiko einer Frühgeburt ist eine genaue<br />
Vorhersage der Zeit bis zur Geburt von klinischer Bedeutung.Dies gilt<br />
insbesondere in Hinblick auf eine Verabreichung von Kortikosteroiden,<br />
die ihre optimale Wirkung innerhalb von 7 Tagen nach Verabreichung<br />
erzielen, 2 sowie der Verlegung von Patientinnen in ein Perinatalzentrum.<br />
Eine klinische Bewertung alleine (einschließlich der Messung von<br />
Zervixlänge und -weite) ist für eine bevorstehende Geburt nicht genügend<br />
aussagekräftig. 3 Ähnlich unzureichende prädiktive Werte in Bezug auf<br />
eine bevorstehende Geburt haben auch die verfügbaren Biomarker-<br />
Tests, wie zum Beispiel der zum Nachweis des fetalen Fibronektins. 4 Eine<br />
genaue Vorhersage der Zeit bis zur Geburt ist demnach für Patientinnen<br />
mit einem Risiko für vorzeitige Wehen klinisch weiterhin von Belang. Der<br />
<strong>PartoSure</strong> TTD Test ist ein schneller, nichtinvasiver Streifentest. Er zeichnet<br />
sich durch einen bislang nicht erreichten prädiktiven Wert für eine Geburt<br />
innerhalb von 7 Tagen bei Patientinnen mit Anzeichen und Symptomen<br />
von vorzeitigen Wehen aus. Der hohe negative und positive prädiktive<br />
Wert für den <strong>PartoSure</strong> TTD Test kann die rechtzeitige Identi zierung<br />
solcher Patientinnen ermöglichen, die von verfügbaren Interventionen,<br />
wie antenatalen Kortikosteroiden. Darüber hinaus führt dieser Test aller<br />
Wahrscheinlichkeit nach zur Einsparung von Kosten, die mit unnötigen<br />
Krankenhauseinweisungen, verursacht durch die schlechten positiven<br />
prädiktiven Werte vorhandener Methoden, verbundenen sind. Das Test-<br />
Set beinhaltet alle Komponenten, um schnell qualitative Ergebnisse<br />
liefern zu können. Zusätzliche Instrumente oder eine invasive Spekulum<br />
Untersuchung sind nicht erforderlich.<br />
TESTPRINZIP<br />
<strong>PartoSure</strong> ist ein immunchromatographischer Test (‚lateral ow Assay‘)<br />
zum Nachweis des humanen plazentalen alpha-Mikroglobulin-1 (PAMG-<br />
1), das bei bevorstehender Geburt in zervikovaginalen Absonderungen<br />
nachweisbar wird. Die für den Test verwendeten monoklonalen Antikörper<br />
sind ausreichend emp ndlich, um 4 pg/μl PAMG-1 in zervikovaginalem<br />
Aus uss nachzuweisen. Wenn die Wehen und Geburt bevorstehen, kann<br />
das während der gesamten Schwangerschaft von den Zellen der Dezidua<br />
gebildete und freigesetzte Protein PAMG-1 im zervikovaginalen Aus uss<br />
nachgewiesen werden. Dies beruht wahrscheinlich auf der Transsudation<br />
des Proteins durch die vorhandenen Poren der chorioamniotischen<br />
Membranen während der Kontraktionen des Uterus und möglicherweise<br />
auf dem Abbau der extrazellulären Matrix der fetalen Membranen<br />
aufgrund eines entzündlichen Prozesses durch die Wehen oder aufgrund<br />
einer Infektion. Ein Nachweis von PAMG-1 im zervikovaginalen Sekret zeigt<br />
an, dass es wahrscheinlich innerhalb von 7 Tagen zur Geburt kommen<br />
wird und dass möglicherweise mit einem signi kant erhöhten Risiko einer<br />
intraamniotischen Infektion und/oder einer Entzündung zu rechnen ist . 1<br />
Die Abwesenheit dieses Proteins zeigt an, dass die Geburt wahrscheinlich<br />
nicht innerhalb der nächsten 7 bis 14 Tage erfolgen wird.<br />
Für die Analyse wird eine durch vaginalen Abstrich genommene Probe des<br />
zervikovaginalen Aus usses in ein Lösungsmittel extrahiert.Anschließend<br />
wird die Anwesenheit des PAMG-1 Antigens durch das Einbringen des<br />
Teststreifens in das Fläschchen mit dem Lösungsmittel nachgewiesen.<br />
Die Probe ießt von einem Saugvlies hin zu einer Nitrozellulose-Membran<br />
und durchläuft dabei einen reaktiven Bereich mit monoklonalen, gold-<br />
konjugierten Anti-PAMG-1 Antikörpern. Der Komplex aus Antigen-<br />
Antikörper wandert weiter zur Testregion und wird dort mittels eines<br />
zweiten Anti-PAMG-1 Antikörpers immbolilisiert. Dadurch kommt es zur<br />
Ausbildung der Testbande. Ungebundene Antigen-Antikörper-Komplexe<br />
ießen weiter entlang des Teststreifens und werden anschließend mittels<br />
eines zweiten Antikörpers immobilisiert. Dies führt zur Ausbildung einer<br />
internen Kontrollbande.<br />
REAGENZIEN UND KOMPONENTEN<br />
Das <strong>PartoSure</strong> Test-Set besteht aus den folgenden Komponenten: dem<br />
<strong>PartoSure</strong> Teststreifen in einem Folienbeutel mit einem Trockenmittel,<br />
einem sterilen, be ockten vaginalen Tupfer sowie einem Plastik äschchen<br />
mit Lösungsmittel (0,9 % NaCl, 0,05 % NaN3, 0,01 % Triton X100).<br />
VORSICHTSMASSNAHMEN UND WARNHINWEISE<br />
<strong>PartoSure</strong> ist ausschließlich für den Gebrauch in der in-vitro Diagnostik<br />
bestimmt. Testkomponenten dürfen nicht verschluckt werden. •<br />
Befolgen Sie exakt die Gebrauchsanweisungen. Eine Nichtbeachtung<br />
der Anweisungen kann zu ungenauen Testergebnissen führen. • Bei der<br />
Entnahme so wie Handhabung und Entsorgung von Probenmaterial sollten<br />
Sicherheitsmaßnahmen beachtet werden • Die Verwendung eines anderen<br />

Beachtung notwendiger Vorsichtsmaßnahmen. • Verwenden Sie den Test<br />
nicht nach Ablauf des auf der Verpackung aufgedruckten Verfalldatums •<br />
Die Komponenten des Test-Sets sind für den Einmalgebrauch bestimmt •<br />
Biegen oder falten Sie weder den Teststreifen noch den Folienbeutel mit<br />
dem darin enthaltenen Teststreifen, da dieser beschädigt werden und<br />
ungenaue Ergebnisse liefern könnte.<br />
AUFBEWAHRUNG UND STABILITÄT<br />
Bewahren Sie das <strong>PartoSure</strong> Test-Set an einem trockenen Ort bei 4-24 °C<br />
(40-75 °F) auf. Der Test darf nicht eingefroren werden. Bei Aufbewahrung<br />
im Folienbeutel bei der empfohlenen Temperatur ist der Test bis zu dem<br />
auf dem Folienbeutel aufgedruckten Verfalldatum stabil. Der <strong>PartoSure</strong><br />
Teststreifen sollte innerhalb von sechs (6) Stunden nach seiner Entnahme<br />
aus dem Folienbeutel verwendet werden.<br />
QUALITÄTSKONTROLLE<br />
Der <strong>PartoSure</strong> Teststreifen enthält eine interne Kontrolle, um die<br />
analytische Funktionalität des Tests sicherzustellen. Das Erscheinen einer<br />
oder zweier Banden in der Testregion des Teststreifens bestätigt, dass das<br />
Testverfahren funktioniert und die Testkomponenten in Ordnung sind.<br />
EINSCHRÄNKUNGEN DES TESTS<br />
<strong>PartoSure</strong> Testergebnisse sind qualitativer und nicht quantitativer Natur.<br />
Von der Stärke der Test- oder Kontrollbanden sollten keine quantitativen<br />
Rückschlüsse gezogen werden. Geringe Blutmengen auf dem Tupfer haben<br />
keinen Ein uss auf die Funktionalität des Tests. Bei größeren Mengen von<br />
Blut auf dem Tupfer kann es sein, dass der Test nicht ordnungsgemäß<br />
funktioniert. Unter diesen Umständen wird von der Verwendung des Tests<br />
abgeraten. <strong>PartoSure</strong> sollte frühestens 6 Stunden nach der Entfernung<br />
von Desinfektionslösungen oder Medikamenten aus der Vagina<br />
angewendet werden. Die Ergebnisse des Tests sollten stets in Verbindung<br />
mit anderen klinischen Daten verwendet werden. Die Aussagekraft des<br />
Tests bei Patientinnen ohne Wehen ist nicht bekannt. Placenta praevia<br />
und eine Durchführung digitaler Untersuchungen vor der Probenahme<br />
kann zu falschen Testergebnissen führen. Die Leistungsfähigkeit des<br />
<strong>PartoSure</strong> Tests in Anwesenheit der folgenden Substanzen wurde nicht<br />
ermittelt: Meconium, antimykotische Cremes, Zäpfchen, Gleitmittel,<br />
Feuchtigkeitsmittel, Talkumpuder oder Babyöl.<br />
ERWARTETE WERTE<br />
<strong>PartoSure</strong> weist kleinste Spuren des humanen PAMG-1 nach. Die<br />
Anwesenheit von PAMG-1 in zervikovaginalem Aus uss in Mengen von<br />
mindestens 4 pg/μl weist auf ein erhöhtes Risiko für eine Geburt innerhalb<br />
der nächsten 7 Tage hin. Die Sensitivitätsgrenze von 4 pg/μl wurde in<br />
einer internationalen, multizentrischen klinischen Studie ermittelt, in<br />
welcher der Zusammenhangs zwischen dem Nachweis von PAMG-1 in<br />
zervikovaginalen Sekreten und der Zeitspanne zwischen Test und Geburt<br />
untersucht wurde.<br />
LEISTUNGSMERKMALE<br />
Die klinischen Eigenschaften des <strong>PartoSure</strong> TTD Tests wurden in einer<br />
internationales Multizenterstudie ermittelt. Patientinnen, (i) mit Anzeichen<br />
und Symptomen, die auf vorzeitige Wehen hindeuten, (ii) zwischen der 20.<br />
und der 36 6/7 SSW und (iii) die klinisch intakte Membranen aufwiesen,<br />
wurden eingeladen an der Studie teilzunehmen. Insgesamt wurden 101<br />
Patientinnen in die endgültige AUswertung eingeschlossen. Folgende<br />
Qualitätsparameter, mit 95% Kon denzintervallen wurden errechnet:<br />

KREUZREAKTIVITÄT<br />
Die folgenden Proteine wurden auf eine Kreuzreaktivität mit den im<br />
<strong>PartoSure</strong> Test verwendeten monoklonalen Antikörpern getestet:<br />
alpha 2-Mikroglobulin (Fertilität), humanes Choriongonadotropin,<br />
trophoblastisches beta-1-Glykoprotein, humanes Plazentalaktogen, alpha-<br />
Fetoprotein, humanes Serumalbumin und verschiedene IGFB-Proteine.<br />
Es kam zu keiner Kreuzreaktivität zwischen diesen Proteinen und den<br />
monoklonalen Antikörpern des <strong>PartoSure</strong> Tests. Bei Verwendung der<br />
ELISA-Methode zeigten die für die Testbande verwendeten Antikörper eine<br />
Kreuzreaktivität mit dem IGFBP-3 Protein.Die Konzentration des IGFBP-3<br />
in vaginalem Aus uss schwangerer Frauen beein usst jedoch nicht die<br />
Genauigkeit des <strong>PartoSure</strong> Tests.<br />
INTERFERENZSTUDIEN<br />
Vaginale Infektionen, Urin, Sperma und kleine Beimischungen von Blut<br />
beein ussen das Testergebniss des <strong>PartoSure</strong> Tests nicht. Diese in<br />
nichtklinischen Studien untersuchten Substanzen und Erkrankungen<br />
führen zu PAMG-1 Konzentrationen ausschließlich unterhalb der<br />
Nachweisgrenze des <strong>PartoSure</strong> Tests.<br />

ANLEITUNG ZUR VERWENDUNG FÜR DIE IN-VITRO -DIAGNOSTIK<br />
<strong>PartoSure</strong> Test rápido para el diagnóstico del parto pretérmino<br />
USO PREVISTO<br />
El test <strong>PartoSure</strong> (test rápido para el diagnóstico del parto pretérmino)<br />
es un test cualitativo inmunocromatográ co rápido y no instrumentado<br />
para la detección in vitro de α-1 microgobulina placentaria (PAMG-1)<br />
en las secreciones vaginales de las mujeres embarazadas. El test debe<br />
ser realizado por profesionales sanitarios y sirve para ayudar en la<br />
predicción del tiempo hasta el parto, cuando las pacientes muestran<br />
signos, síntomas o molestias que sugieran el parto entre las 20 y 37<br />
semanas de gestación con membranas clínicamente intactas.<br />
RESUMEN Y EXPLICACIÓN DEL TEST<br />
Una predicción precisa del tiempo para el parto tiene importancia clínica<br />
en los embarazos con riesgo de parto prematuro. Esto es especialmente<br />
relevante en lo que respecta tanto a la administración de corticoides,<br />
que producen un efecto óptimo a los 7 días de ser administrados, 2 como<br />
a la derivación de pacientes a un centro de atención especializada,<br />
capaz de atender el nacimiento de un bebé prematuro.<br />
Por sí sola, la evaluación clínica, incluyendo la medida de la longitud<br />
y la dilatación cervical, no es su ciente para predecir un parto<br />
inminente. 3 De forma similar, los test de biomarcadores actuales,<br />
como los basados en la detección de bronectina fetal, tienen un valor<br />
predictivo extremadamente bajo para el parto inminente. 4 Por eso,<br />
la predicción precisa del tiempo que falta para el parto en pacientes<br />
con riesgo de parto prematuro sigue siendo una gran preocupación<br />
clínica. El test <strong>PartoSure</strong> es un test rápido y no invasivo con un valor<br />
predictivo sin precedentes para el parto en un plazo de 7 días, para<br />
pacientes que presentan signos y síntomas de parto prematuro. Los<br />
altos valores de predicción, negativa y positiva, del test <strong>PartoSure</strong><br />
pueden permitir la identi cación oportuna de pacientes que se<br />
bene ciarían de las intervenciones disponibles, como los corticoides<br />
prenatales. Además, una identi cación más precisa de estos pacientes<br />
reducirá probablemente los costes asociados a las hospitalizaciones<br />
innecesarias, causadas por los precarios valores de predicción positiva<br />
de los métodos existentes. El kit del test es un sistema completo que<br />
proporciona resultados rápidos y cualitativos sin instrumentación<br />
externa ni examen invasivo con espéculo.<br />
PRINCIPIOS DEL TEST<br />
El test <strong>PartoSure</strong> es un análisis inmunocromatográ co de ujo<br />
lateral diseñado para identi car la presencia de α-1 microglobulina<br />
placentaria (PAMG-1) humana, que se puede detectar en exudados<br />
cervicovaginales cuando el parto es inminente. El test utiliza anticuerpos<br />
monoclonales con la sensibilidad su ciente para detectar 4 pg/μl de<br />
PAMG-1 en el ujo cervicovaginal. PAMG-1, una proteína liberada por<br />
las células deciduales en la cavidad amniótica durante el embarazo,<br />
está presente en el ujo cervicovaginal cuando el parto es inminente.<br />
Esto se debe probablemente a la trasudación de la proteína a través<br />
de poros preexistentes en las membranas corioamnióticas durante las<br />
contracciones uterinas y, potencialmente, a la degradación de la matriz<br />
extracelular de las membranas fetales debido a un proceso in amatorio<br />
del parto o de una infección. Su presencia en las secreciones<br />
cervicovaginales indica que el parto ocurrirá probablemente en un<br />
plazo de 7 días y que puede conllevar un riesgo considerablemente<br />
mayor de infección y/o in amación intramniótica. 1 Su ausencia indica<br />
que el parto seguramente no ocurrirá en un plazo de 7 o 14 días.<br />
Para el análisis, se extrae una muestra de ujo cervicovaginal mediante<br />
una torunda vaginal que se introduce en un disolvente. La presencia<br />
del antígeno PAMG-1 se detecta insertando una tira reactiva de ujo<br />
lateral en el vial. La muestra uye de una compresa absorbente a una<br />
membrana de nitrocelulosa, pasando a través de un área reactiva que<br />
contiene anticuerpos monoclonales anti-PAMG-1, combinados con una<br />
partícula de oro. El complejo antígeno-anticuerpo uye hacia la región<br />
de análisis donde queda inmovilizado por un segundo anticuerpo -anti-<br />
PAMG-1. Esto hace que aparezca la línea de reacción. Los complejos<br />
antígeno-anticuerpo sueltos siguen uyendo por la tira reactiva y son<br />
inmovilizados por un segundo anticuerpo. Esto hace que aparezca la<br />
línea de control interno.<br />
REACTIVOS Y COMPONENTES<br />
El kit del test <strong>PartoSure</strong> incluye los siguientes componentes: Tira<br />
reactiva <strong>PartoSure</strong> en bolsa de aluminio con desecante, torunda<br />
vaginal estéril y vial de plástico con solución disolvente (0,9% NaCl,<br />
0,05% NaN3, 0,01% Triton X100).<br />
PRECAUCIONES Y ADVERTENCIAS<br />
El test <strong>PartoSure</strong> está pensado únicamente para el diagnóstico in vitro<br />
y todos sus componentes son de uso externo • Se deben seguir las<br />
instrucciones de uso de forma exacta; de no ser así pueden obtenerse<br />
resultados poco precisos • Siga las precauciones de seguridad a la hora<br />
de obtener, manipular y desechar las muestras del análisis • El uso de<br />
cualquier torunda que no sea la torunda estéril incluida con el kit del test<br />
alterará el resultado y, por lo tanto, está estrictamente prohibido. • No<br />
utilice componentes del kit que estén dañados • Los kits utilizados son<br />
residuos biológicos. Deshágase de todos los componentes adoptando<br />
las precauciones necesarias. • No utilice el test después de la fecha de<br />
caducidad, impresa en el envase del producto • Los componentes del<br />
kit son para un solo uso • No doble o pliegue la tira reactiva o la bolsa<br />
de aluminio que la contiene, si lo hace puede dañar la tira y obtener<br />
resultados inexactos<br />
ALMACENAMIENTO Y ESTABILIDAD<br />
Guarde el kit del test <strong>PartoSure</strong> en un lugar seco de 4 a 24 ºC. El test<br />
no se debe congelar. Mientras se conserve en la bolsa de aluminio<br />
a la temperatura recomendada, el test es estable hasta la fecha de<br />
caducidad impresa en la bolsa. El test <strong>PartoSure</strong> debe usarse en un<br />
plazo de seis (6) horas después de sacar la tira reactiva de la bolsa de<br />
aluminio.<br />
CONTROL DE CALIDAD<br />
El test <strong>PartoSure</strong> contiene un mecanismo de control interno que<br />
asegura la funcionalidad analítica. La aparición de una o dos líneas en la<br />
zona de resultados de la tira reactiva veri ca la integridad del proceso y<br />
los componentes del test.<br />
LIMITACIONES DEL TEST<br />
Los resultados del test <strong>PartoSure</strong> son cualitativos, no cuantitativos.<br />
No se debe hacer ninguna interpretación cuantitativa basada en la<br />
fuerza del test o de las líneas de control. En casos en los que solo haya<br />
una pequeña cantidad de sangre en el bastoncillo, el test funcionará<br />
correctamente. Cuando haya una cantidad considerable de sangre<br />
en el bastoncillo, el test puede no funcionar correctamente y su uso<br />
no está recomendado. El test <strong>PartoSure</strong> no debe usarse antes de 6<br />
horas después de eliminar cualquier solución o medicina desinfectante<br />
de la vagina. Los resultados deben utilizarse combinados con más<br />
información clínica. Se desconoce el rendimiento del test en pacientes<br />
sin signos o síntomas de parto. La placenta previa y la realización de<br />
exámenes digitales antes de la extracción de muestras pueden dar<br />
lugar a resultados inexactos. El rendimiento del test <strong>PartoSure</strong> no se<br />
ha establecido en presencia de los siguientes contaminantes: meconio,<br />
cremas fungicidas, supositorios, lubricantes, cremas hidratantes, polvos<br />
de talco o aceite para bebés.<br />
VALORES PREVISTOS<br />
El test <strong>PartoSure</strong> detecta pequeñas cantidades de PAMG-1 humana.<br />
La PAMG-1 presente en el ujo cervicovaginal en cantidades iguales<br />
o superiores a 4 pg/μl indica un elevado riesgo de parto en los 7 días<br />
siguientes. El umbral de sensibilidad de 4 pg/μl fue validado en un<br />
ensayo clínico internacional entre varios centros que evaluaron la<br />
relación entre la PAMG-1 presente en las secreciones cervicovaginales<br />
y el intervalo de tiempo entre el test y el parto.<br />
CARACTERÍSTICAS DE RENDIMIENTO<br />
El rendimiento clínico del test <strong>PartoSure</strong> ha sido determinado en<br />
un estudio multicéntrico internacional. Aquellas pacientes que (i)<br />
presentan signos, síntomas o molestias que puedan sugerir trabajo<br />
de parto pretérmino, (ii) se encontraban entre la semana 20 y 366⁄7<br />
de gestación y (iii) con membranas clínicamente intactas, han sido<br />
invitadas a participar en el estudio. En total, han sido incluidas en el<br />
análisis 101 pacientes. Las siguientes estimaciones de rendimiento,<br />
con sus correspondientes intervalos de con anza del 95%, han sido<br />
calculadas:<br />

REACTIVIDAD CRUZADA<br />
Las siguientes proteínas fueron analizadas para la reactividad cruzada<br />
con los anticuerpos monoclonales utilizados en el test <strong>PartoSure</strong>:<br />
α-2- microglobulina de fertilidad, gonadotropina coriónica humana,<br />
β-1-glicoproteína trofoblástica, lactógeno placentario humano,<br />
α-fetoproteína, seroalbúmina humana y varias proteínas transportadoras<br />
del factor de crecimiento insulínico (IGFBP). Los anticuerpos<br />
monoclonales analizados que se utilizan en el test <strong>PartoSure</strong> no<br />
tuvieron reactividad cruzada con estas proteínas. El anticuerpo utilizado<br />
en la línea de reacción presentó reactividad cruzada con la proteína<br />
IGFBP-3 al utilizarse el método ELISA. La concentración de IGFBP-3 en la<br />
secreción vaginal de mujeres embarazadas, sin embargo, no afecta a la<br />
precisión del test <strong>PartoSure</strong>.<br />
ESTUDIO DE INTERFERENCIAS<br />
Las infecciones vaginales, orina, semen y pequeñas mezclas de sangre<br />
no inter eren con los resultados del test <strong>PartoSure</strong>. Estas sustancias y<br />
condiciones han sido evaluadas en estudios no clínicos y se descubrió<br />
que presentaban concentraciones de PAMG-1 inferiores al umbral de<br />
detección del test <strong>PartoSure</strong>.<br />

USO PREVISTO<br />
Il test di disgnostico del parto prematuro (test per il tempo al parto)<br />
<strong>PartoSure</strong> è un test rapido immunocromatogra co qualitativo e<br />
non strumentale per la rilevazione in vitro dell’alfa-1 microglobulina<br />
placentaria (PAMG-1) nelle secrezioni vaginali delle donne in gravidanza.<br />
Il test è destinato ai professionisti che operano nel settore sanitario<br />
come aiuto nella previsione di un parto imminente nelle pazienti che<br />
presentano segnali, sintomi o disturbi che suggeriscono un travaglio tra<br />
la 20ª e la 37ª settimana di gestazione, con membrane intatte dal punto<br />
di vista clinico.<br />
INTRODUZIONE E SPIEGAZIONE DEL TEST<br />
Dal punto di vista clinico, prevedere in modo accurato il parto<br />
prematuro è importante soprattutto nelle gravidanze che presentano<br />
questo rischio. Questo è particolarmente vero per ció che riguarda la<br />
somministrazione di corticosteroidi, che danno ottimi bene ci entro 7<br />
giorni dalla somministrazione, 2 sia al trasferimento delle pazienti presso<br />
un centro di cura terziario in grado di fornire assistenza alla nascita di un<br />
neonato prematuro.<br />
La sola valutazione clinica, che comprende la misurazione della<br />
lunghezza e della dilatazione della cervice, non è su ciente per<br />
prevedere l’imminenza del parto. 3 Analogamente i test a base di<br />
biomarcatori attualmente disponibili, come quello basato sulla<br />
rilevazione della bronectina fetale, forniscono valori predittivi molto<br />
scarsi di un parto imminente. 4 Per questo motivo, prevedere in modo<br />
preciso l’imminenza del parto nelle pazienti a rischio di travaglio<br />
pretermine rimane un importante problema clinico. Il test <strong>PartoSure</strong><br />
è un test a striscia rapido e non invasivo, con un valore predittivo del<br />
parto senza precedenti entro 7 giorni, in pazienti con segnali e sintomi<br />
indicativi di un travaglio pretermine. Gli alti valori predittivi, positivo<br />
e negativo, del test <strong>PartoSure</strong> possono consentire l’identi cazione<br />
tempestivadelle pazienti che trarrebbero bene ci dagli interventi<br />
disponibili, come la somministrazione prenatale di corticosteroidi.<br />
Inoltre, un’identi cazione più accurata di tali pazienti ridurrebbe anche<br />
i costi associati a ricoveri non necessari dovuti al basso valore predittivo<br />
positivo dei metodi esistenti. Il kit di test è un sistema autonomo che<br />
fornisce rapidamente risultati qualitativi, senza strumentazione esterna<br />
o un esame invasivo con lo speculum ginecologico.<br />
PRINCIPIO DEL TEST<br />
<strong>PartoSure</strong> è un test immunocromatogra co a usso laterale,<br />
progettato per identi care la presenza dell’alfa-1 microglobulina<br />
placentaria (PAMG-1) umana, rilevabile negli essudati cervico-vaginali<br />
quando il parto è imminente. Il test utilizza anticorpi monoclonali<br />
su cientemente sensibili a rilevare 4 pg/μl di PAMG-1 nelle secrezioni<br />
cervico-vaginali. La PAMG-1, una proteina rilasciata dalle cellule<br />
deciduali nella cavitá amniotica durante l’intera gravidanza, è presente<br />
nelle secrezioni cervico-vaginali quando il travaglio e il parto sono<br />
imminenti. Probabilmente questo è dovuto alla trasudazione della<br />
proteina attraverso i pori preesistenti nelle membrane corio-amniotiche<br />
durante le contrazioni uterine e, potenzialmente, alla degradazione<br />
della matrice extracellulare delle membrane fetali dovuta a un processo<br />
in ammatorio di travaglio o infezione. La sua presenza nelle secrezioni<br />
cervico-vaginali indica che il parto probabilmente avra luogo entro 7<br />
giorni e puo essere associata a un altissimo rischio di infezione e/o<br />
in ammazione intra-amniotica. 1 La sua assenza indica che il parto<br />
di cilmente avra luogo entro 7 o 14 giorni.<br />
Per l’analisi, il campione di secrezione cervico-vaginale viene estratto e<br />
raccolto mediante un tampone vaginale e si immerge in un solvente. La<br />
presenza dell’antigene PAMG-1 viene quindi rilevata inserendo nel<br />
solvente una striscia per il test a usso laterale. Il campione uisce dal<br />
tampone assorbente a una membrana di nitrocellulosa, passando<br />
attraverso l’area reattiva contenente gli anticorpi monoclonali anti-<br />
PAMG-1 combinati con una particella d’oro. Il complesso antigene-<br />
anticorpo uisce verso la zona del test dove viene immobilizzato da un<br />
secondo anticorpo anti-PAMG-1. Questo evento porta alla comparsa<br />
della linea del test. I complessi antigene-anticorpo sciolti continuano a<br />
uire lungo la striscia per il test e vengono immobilizzati da un secondo<br />
anticorpo. Questo porta alla comparsa della linea del controllo interno.<br />
REAGENTI E COMPONENTI<br />
Il kit di test <strong>PartoSure</strong> comprende i seguenti componenti: striscia per<br />
il test <strong>PartoSure</strong> in un sacchetto in alluminio con essiccante, tampone<br />
vaginale in cotone sterile e ala in plastica con soluzione solvente (0,9%<br />
NaCl, 0,05% NaN3, 0,01% Triton X100).<br />
PRECAUZIONI ED AVVERTENZE<br />
<strong>PartoSure</strong> è destinato esclusivamente all’uso diagnostico in vitro e<br />
nessun componente del test deve essere usato separatamente • Seguire<br />
esattamente le istruzioni per l’uso; la mancata osservanza potrebbe<br />
portare a risultati imprecisi • Rispettare le precauzioni di sicurezza<br />
durante la raccolta, la manipolazione e lo smaltimento dei campioni<br />
per il test • L’uso di tamponi diversi da quello in cotone sterile fornito<br />
con il kit del test altera le prestazioni del test ed è quindi severamente<br />
proibito • Non utilizzare componenti del kit di test danneggiati<br />
• I kit usati sono considerati rifi uti biologici; smaltire tutti i<br />
componenti usando le dovute precauzioni • Non usare il test dopo la<br />
data di scadenza stampata sulla confezione del prodotto • I componenti<br />
del kit sono monouso • Non curvare o piegare la striscia per il<br />
test o il sacchetto in alluminio con la striscia per il test all’interno; cosí<br />
facendo, si puó danneggiare la striscia ed ottenere risultati imprecisi<br />
CONSERVAZIONE E VALIDITÀ<br />
Conservare il test <strong>PartoSure</strong> in un luogo asciutto, ad una temperatura<br />
compresa tra 4 e 24°C (tra 40 e 75°F). Il test non deve essere congelato.<br />
Se conservato nel sacchetto in alluminio ed alla temperatura<br />
raccomandata, il test è valido no alla data di scadenza stampata sul<br />
sacchetto. La striscia per il test <strong>PartoSure</strong> deve essere utilizzata entro<br />
sei (6) ore dalla sua rimozione dal sacchetto in alluminio.<br />
CONTROLLO QUALITÀ<br />
La striscia per il test <strong>PartoSure</strong> contiene un meccanismo interno<br />
di controllo della procedura che ne assicura la funzionalitá analitica.<br />
La comparsa di una o due linee nella zona dei risultati della striscia<br />
dimostra l’integritá della procedura e dei componenti del test.<br />
LIMITAZIONI DEL TEST<br />
I risultati del test <strong>PartoSure</strong> sono qualitativi e non quantitativi. Non si<br />
devono fare interpretazioni quantitative sulla base dell’intensitá delle<br />
linee del test o del controllo. Il test funziona correttamente anche in<br />
caso di piccole tracce di sangue sul tampone. Se sul tampone si nota<br />
una quantitá notevole di sangue, il test potrebbe non funzionare<br />
correttamente e se ne sconsiglia l’uso. <strong>PartoSure</strong> non deve essere<br />
utilizzato prima che siano trascorse 6 ore dalla rimozione di qualsiasi<br />
soluzione disinfettante o di qualsiasi medicinale dalla vagina. I risultati<br />
devono essere utilizzati in combinazione con altre informazioni cliniche.<br />
Le prestazioni del test su pazienti che non presentano segnali o<br />
sintomi di travaglio sono sconosciute. La placenta previa e l’esecuzione<br />
di esami digitali prima della raccolta del campione possono portare a<br />
risultati imprecisi del test. Le prestazioni di <strong>PartoSure</strong> non sono state<br />
stabilite in presenza dei seguenti agenti contaminanti: meconio, creme<br />
antimicotiche, supposte, lubri canti, creme idratanti, talco in polvere o<br />
olio per neonati.<br />
VALORI PREVISTI<br />
<strong>PartoSure</strong> rileva piccole tracce di PAMG-1 umana. La PAMG-1 presente<br />
nelle secrezioni cervico-vaginali in quantita superiori o pari a 4pg/μl<br />
indica un elevato rischio di parto entro i successivi 7 giorni. La soglia di<br />
sensibilita di 4pg/μl è stata convalidata da una sperimentazione clinica<br />
multicentrica internazionale che ha valutato l’associazione tra la PAMG-<br />
1 presente nelle secrezioni cervico-vaginali e l’intervallo di tempo tra<br />
test e parto.<br />
CARATTERISTICHE DELLE PRESTAZIONI<br />
Il rendimento clinico del test <strong>PartoSure</strong> è stato determinato da uno<br />
studio multicentrico internazionale. Le pazienti che (i) presentavano<br />
segni, sintomi o lamentavano la possibilitá di travaglio pretermine, (ii)<br />
si trovavano tra la settimana 20 e la 366⁄7 di gestazione, e (iii) avevano<br />
le membrane clinicamente intatte, sono state invitate a partecipare nel<br />
trial. In totale, 101 pazienti sono state incluse nell’analisi nale. Sono<br />
state calcolate, con i corrispondenti intervalli di con denza (CI) al 95%,<br />
le seguenti stime di rendimento:<br />

CROSS REATTIVITÁ<br />
Le seguenti proteine sono state testate per la reattivitá incrociata con<br />
gli anticorpi monoclonali usati in <strong>PartoSure</strong>: alfa-2 microglobulina<br />
per la fertilitá, gonadotropina corionica umana, beta 1 glicoproteina<br />
trofoblastica, lattogeno placentare umano, alfa-fetoproteina, albumina<br />
sierica umana e varie proteine IGFBP. Gli anticorpi monoclonali<br />
esaminati usati in <strong>PartoSure</strong> non presentavano una reattivita incrociata<br />
con queste proteine. E’ stata rilevata la reattivitá incrociata dell’anticorpo<br />
usato nella linea del test con la proteina IGFBP-3, usando il metodo<br />
ELISA. Tuttavia la concentrazione della IGFBP-3 nelle secrezioni vaginali<br />
delle donne in gravidanza non in uisce sulla precisione di <strong>PartoSure</strong>.<br />
STUDI SULLE INTERFERENZE<br />
Infezioni vaginali, urina, seme e piccole quantitá di sangue non<br />
interferiscono con i risultati di <strong>PartoSure</strong>. Queste sostanze e<br />
condizioni sono state valutate in studi non clinici ed è stato rilevato che<br />
contengono PAMG-1 in concentrazioni inferiori alla soglia di rilevazione<br />
di <strong>PartoSure</strong>.<br />

<strong>PartoSure</strong> TTD (Doğum Zamanı) Test<br />
KULLANIM AMACI<br />
<strong>PartoSure</strong> TTD (Doğum Zamanı) Test, gebe kadınların vajinal<br />
sekresyonlarında plasental alfa mikroglobulin-1’in (PAMG-1) in<br />
vitro tespitine yönelik, hızlı, enstrüman gerektirmeyen, kalitatif bir<br />
immünokromatogra k testtir. Test, klinik olarak intakt membranların<br />
olduğu gebeliğin 20. ile 37. haftaları arasında, hastalar doğumu<br />
düşündüren bulgu, belirtiler veya şikayetleri bildirdiğinde, doğum<br />
zamanının tahmin edilmesine yardımcı olmak üzere sağlık uzmanları<br />
tarafından kullanım içindir.<br />
TEST ÖZETİ VE AÇIKLAMASI<br />
Doğum zamanına dair doğru bir tahmin, erken doğum açısından<br />
risk altında olan gebeliklerde klinik olarak önemlidir. Bu özellikle,<br />
uygulamadan itibaren 7 gün içinde optimal faydası olan<br />
kortikosteroidlerin uygulanması 2 ve bunun yanı sıra hastaların<br />
prematüre bebek doğumu için bakım verebilen üçüncü basamak bir<br />
tedavi merkezine transferi açısından geçerlidir.<br />
Serviks boyu ve dilatasyonu dahil tek başına klinik değerlendirme,<br />
yaklaşmakta olan doğumu yeterli şekilde tahmin ettirmez. 3 Benzer<br />
şekilde, fetal bronektinin saptanmasını temel alanlar gibi halihazırda<br />
kullanılmakta olan biyomarker testleri, yaklaşmakta olan doğum<br />
açısından son derece düşük prediktif değerlere sahiptir. 4 Bu nedenle,<br />
erken doğum tehlikesi olan hastalarda doğum zamanının doğru<br />
tahmini, önemli bir klinik sorun olmaya devam etmektedir. <strong>PartoSure</strong><br />
TTD Test, erken doğum sancısı bulguları ve belirtileri gösteren<br />
hastalarda 7 gün içindeki doğum için emsalsiz prediktif değeri olan<br />
hızlı, invazif olmayan bir strip testidir. <strong>PartoSure</strong> TTD Test’in yüksek<br />
negatif ve pozitif prediktif değerleri, antenatal kortikosteroidler ve<br />
gibi mevcut müdahalelerden fayda görecek olan hastaların zamanında<br />
tespit edilmesine olanak verebilir. Ayrıca, bu hastaların daha doğru<br />
biçimde tespit edilmesi, mevcut yöntemlerin düşük pozitif prediktif<br />
değerlerinden kaynaklanan gereksiz uygulamalarla ilişkili maliyetleri<br />
muhtemelen azaltacaktır. Test kiti, harici enstrümantasyon veya invazif<br />
spekulum incelemesi olmadan, hızlı, kalitatif sonuçlar veren bağımsız<br />
bir sistemdir.<br />
TEST PRENSİBİ<br />
<strong>PartoSure</strong>, doğum yaklaştığında servikovajinal eksüda sıvılarından<br />
saptanabilen insan plasental-alfa-mikroglobulin-1’in (PAMG-1) varlığını<br />
belirlemek üzere tasarlanmış bir lateral akış immünokromatogra k<br />
analizdir. Test, servikovajinal akıntıda 4 pg/μl PAMG-1’i saptamak<br />
için yeterince duyarlı olan monoklonal antikorları kullanır. Gebelik<br />
süresince amniyotik kavitedeki desidüal hücrelerden salınan bir protein<br />
olan PAMG-1, doğum sancısı ve doğum yaklaştığında servikovajinal<br />
akıntıda bulunur. Bu durum muhtemelen, uterin kasılmalar sırasında<br />
koriyoamniyotik membranlarda önceden var olan porlar aracılığıyla<br />
proteinin transüdasyonu ve potansiyel olarak, in amatuar doğum<br />
sancısı veya enfeksiyon süreci nedeniyle fetal membranların<br />
ekstraselüler matriksinin degradasyonundan ötürü oluşur. Proteinin<br />
servikovajinal sekresyonlardaki varlığı, doğumun olasılıkla 7 gün içinde<br />
meydana geleceğini gösterir ve anlamlı olarak artmış intraamniyonik<br />
enfeksiyon ve/veya en amasyon riski ile ilişkili olabilir. 1 Olmaması,<br />
doğumun muhtemelen 7 veya 14 gün içinde meydana gelmeyeceğini<br />
gösterir.<br />
Analiz için, vajinal çubuk ile alınan servikovajinal akıntı numunesi<br />
çözücü içine aktarılır. Ardından PAMG-1 antijeninin varlığı, şişe içine<br />
bir lateral akış test stripi sokularak belirlenir. Numune, altın parçacığına<br />
konjüge edilmiş monoklonal anti-PAMG-1 antikorlarını içeren reaktif<br />
alan üzerinden geçerek, emici pedden bir nitroselüloz membrana akar.<br />
Antijen-antikor kompleksi, ikinci bir anti-PAMG-1 antikoru ile immobilize<br />
edildiği test alanına akar. Bu olay, test çizgisinin görünmesine yol açar.<br />
Bağlanmayan antijen-antikor kompleksleri, test stripi boyunca akmaya<br />
devam eder ve ikinci bir antikor ile immobilize edilir. Bu durum, internal<br />
kontrol çizgisinin görünmesine yol açar.<br />
REAKTİFLER VE BİLEŞENLER<br />
<strong>PartoSure</strong> test kiti şu bileşenleri içerir: Nem çekici, steril pamuklu<br />
vajinal çubuğun bulunduğu folyo poşet içindeki <strong>PartoSure</strong> test stripi<br />

SAKLAMA VE STABİLİTE<br />
<strong>PartoSure</strong> test kitini, 4 ila 24 °C’de (40 ila 75 °F) kuru bir yerde<br />
saklayın. Test dondurulmamalıdır. Önerilen sıcaklıkta folyo poşet içinde<br />
saklandığında, test, poşet üzerinde basılı olan son kullanma tarihine<br />

<strong>PartoSure</strong> test stripi, analitik işlevsellik sağlayan internal prosedürel bir<br />
kontrol mekanizması içerir. Test stripinin sonuçlar alanındaki bir veya iki<br />
çizginin görünümü, test prosedürünün ve bileşenlerinin bütünlüğünü<br />
doğrular.<br />
TEST KISITLAMALARI<br />
<strong>PartoSure</strong> test sonuçları kantitatif değil, kalitatiftir. Test veya kontrol<br />
çizgilerinin etkinliğine dayalı olarak kantitatif yorumlama yapılmamalıdır.<br />
Çubuk üzerinde sadece eser miktarlarda kanın olduğu olgularda, test<br />
doğru olarak çalışacaktır. Çubuk üzerinde belirgin miktarda kan varsa,<br />
test hatalı olarak çalışabilir ve kullanımı önerilmez. <strong>PartoSure</strong>, vajinadan<br />
dezenfektan solüsyonlar veya ilaçların giderilmesinin ardından 6 saatten<br />
daha erken bir sürede kullanılmamalıdır. Sonuçlar, diğer klinik bilgilerle<br />
beraber kullanılmalıdır. Doğum sancısı bulgu veya belirtileri olmayan<br />
hastalarda test performansı bilinmemektedir. Plasenta previa ve<br />
numune alma öncesi dijital muayenelerin gerçekleştirilmesi, yanlış test<br />
sonuçlarına neden olabilir. <strong>PartoSure</strong> performansı, şu kontaminantların<br />
varlığında belirlenmemiştir: mekonyum, anti-fungal merhemler, tiller,<br />
kayganlaştırıcılar, nemlendiriciler, talk pudraları veya bebe yağı.<br />
BEKLENEN DEĞERLER<br />
<strong>PartoSure</strong>, eser miktarlardaki insan PAMG-1’ini belirler. Servikovajinal<br />
akıntıda 4pg/μl’dan daha yüksek veya ona eşit miktarlarda PAMG-1<br />
varlığı, sonraki 7 gün içinde doğum yapma riskinin artmış olduğunu<br />
gösterir. 4pg/μl’lik duyarlılık eşik değeri, servikovajinal sekresyonlarda<br />
bulunan PAMG-1’in ilişkisini ve testin gerçekleştirilmesinden doğuma<br />
kadar geçen süreyi değerlendiren uluslararası çok merkezli bir klinik<br />
çalışmada onaylanmıştır.<br />
PERFORMANS ÖZELLİKLERİ<br />
<strong>PartoSure</strong> TTD testinin klinik performansı ,uluslararası çoklu saha<br />
çalışmaları ile belirlendi. Preterm doğum düşündüren belirtiler,<br />
bulgular yada yakınmalar ile rapor edilen; gebeliğin 20. ve 36. 6/7<br />
haftaları arasındaki ve klinik olarak intakt membrana sahip olan hastalar<br />
araştırmaya iştirakları için davet edildi. Son analizlere, toplam 101 hasta<br />
dahil edildi. %95 oranındaki güvenilirlik aralığı ile sonraki performans<br />
değerlendirmeleri hesaplandı:<br />

ÇAPRAZ REAKTİFLİK<br />
Şu proteinler <strong>PartoSure</strong> test stripinde kullanılan monoklonal<br />
antikorlar ile çapraz reakti ik açısından test edilmiştir: fertiliteyle<br />
ilgili alfa-2-mikroglobulin, insan koryonik gonadotropini, trofoblastik<br />
beta-1-glikoprotein, insan plasental laktojeni, alfa-fetoprotein, insan<br />
serum albümini ve çeşitli IGFBP proteinleri. <strong>PartoSure</strong> test stripinde<br />
kullanılan, test edilen monoklonal antikorlar, bu proteinlerle çapraz<br />
reaksiyona neden olmamıştır. Test çizgisinde kullanılan antikorun, ELISA<br />
yönteminin kullanılması ile IGFBP-3 proteiniyle çapraz reaksiyona neden<br />
olduğu belirlenmiştir. Ancak, gebe kadınların vajinal akıntısındaki<br />
IGFBP-3 konsantrasyonu, <strong>PartoSure</strong> test stripinin doğruluğunu<br />
etkilemez.<br />
KARŞILIKLI ETKİLEŞİM ÇALIŞMALARI<br />
Vajinal enfeksiyonlar, idrar, semen ve küçük miktardaki kan katkı<br />
maddeleri, <strong>PartoSure</strong> test stripi sonuçları ile etkileşime girmez. Bu<br />
maddeler ve koşullar klinik olmayan çalışmalarda değerlendirilmiştir<br />
ve bunların yalnızca <strong>PartoSure</strong> test stripinin saptama eşik değerinin<br />

<strong>PartoSure</strong> Test rapide du délai d’accouchement en situation de prématurité<br />
USAGE PRÉVU<br />
<strong>PartoSure</strong>, test rapide du délai d’accouchement en situation de<br />
prématurité, est un test immuno-chromatographique rapide et qualitatif<br />
qui ne nécessite aucune instrumentalisation particulière. Il est destiné<br />
à la détection in vivo de l’alpha-1 microglobuline placentaire (PAMG-1)<br />
contenue dans les sécrétions vaginales des femmes enceintes. Ce test<br />
est destiné à être utilisé par des professionnels de santé a n de les aider<br />
à prévoir le délai d’accouchement lorsque des patientes présentant des<br />
membranes cliniquement intactes leur font part de signes, symptômes<br />
ou douleurs, laissant présager le début du travail entre la vingtième et la<br />
trente-septième semaine de grossesse.<br />
RÉSUMÉ ET EXPLICATION DU TEST<br />
Une prévision la plus précise possible de la date d’accouchement est<br />
importante d’un point de vue clinique pour les grossesses présentant un<br />
risque d’accouchement prématuré. Ceci est d’autant plus vrai lorsqu’il<br />
s’agit d’administrer des corticoïdes, qui donnent un résultat optimal dans<br />
un délai de sept jours après administration, 2 ou lorsqu’il s’agit de transférer<br />
des patientes vers une maternité de niveau supérieur capable de prendre<br />
en charge la naissance d’un enfant prématuré. 3<br />
En outre, les tests actuellement disponibles utilisant des biomarqueurs, tels<br />
que celui basé sur la détection de la bronectine fœtale, ont une valeur<br />
prédictive extrêmement limitée pour identi er le délai d’accouchement. 4<br />
Par conséquent, la précision des prévisions concernant la date<br />
d’accouchement des patientes atteintes d’une menace d’accouchement<br />
prématuré demeure une préoccupation clinique importante. Le test<br />
<strong>PartoSure</strong> est un test rapide, non invasif, sous forme de bandelette, o rant<br />
une valeur prédictive sans précédent, car il peut prédire un accouchement<br />
jusqu’a sept jours avant pour les patientes présentant des signes et<br />
symptômes d’une menace d’accouchement prématuré. Les fortes valeurs<br />
prédictives positives et négatives du test <strong>PartoSure</strong> peuvent permettre<br />
d’identi er à temps<br />
des patientes susceptibles de nécessiter des interventions comme<br />
l’administration de corticoïdes. En outre, une identi cation plus précise<br />
de ces patientes est susceptible de réduire les coûts liés à des admissions<br />
inutiles dues aux faibles valeurs prédictives positives des méthodes<br />
existantes. Ce kit est un système autonome fournissant rapidement des<br />
résultats qualitatifs, sans instrumentation extérieure ni examen invasif a<br />
l’aide d’un speculum.<br />
PRINCIPE DU TEST<br />
<strong>PartoSure</strong> est un test immuno-chromatographique à ux latéraux conçu<br />
pour détecter la présence de l’alpha-1 microglobuline placentaire (PAMG-<br />
1) se trouvant dans les exsudats cervico-vaginaux lorsque l’accouchement<br />
est imminent. Le test utilise des anticorps monoclonaux su samment<br />
sensibles pour détecter 4 ng/μl de PAMG-<br />
1 dans les secrétions vaginales. La PAMG-1 est une protéine libérée par<br />
des cellules déciduales, dans la cavité amniotique et ceci tout au long de la<br />
grossesse. Elle est présente dans les secrétions vaginales lorsque le travail<br />
et l’accouchement sont imminents. Cela est probablement causé par la<br />
transsudation de la protéine a travers les pores déjà existants dans<br />
les membranes chorioamniotiques pendant les contractions utérines<br />
et, potentiellement, par la dégradation de la matrice extracellulaire des<br />
membranes fœtales en raison d’un processus in ammatoire lors du travail<br />
ou d’une infection. La présence de cette protéine dans les secrétions<br />
cervico-vaginales indique que l’accouchement est susceptible de survenir<br />
dans un délai de sept jours et peut être associée<br />
à un risque considérablement accru d’infection intra-amniotique et/ou<br />
d’in ammation. 1 L’absence de cette protéine indique que l’accouchement<br />
n’aura probablement pas lieu avant sept à quatorze jours.<br />
Pour e ectuer l’analyse, un échantillon de sécrétion cervico-vaginale est<br />
prélevé à l’aide d’un écouvillon vaginal, puis placé dans un solvant. La<br />
présence de l’antigène de PAMG-1 est ensuite détectée en introduisant<br />
la bandelette du test dans le acon. L’échantillon migre depuis le tampon<br />
absorbant vers une membrane de nitrate de cellulose, en traversant une<br />
surface réactive contenant des anticorps monoclonaux anti-PAMG-1<br />
associés à une particule d’or. Le complexe antigène-anticorps migre vers<br />
la zone de test où il est immobilisé par un second anticorps anti-PAMG-1.<br />
Cet événement conduit à l’apparition de la bande de test. Les complexes<br />
antigène-anticorps qui n’ont pas été immobilisés continuent de migrer le<br />
long de la bandelette de test et sont immobilisés par un second anticorps.<br />
Cela conduit à l’apparition de la bande de contrôle interne.<br />
RÉACTIFS ET COMPOSANTS<br />
Le kit <strong>PartoSure</strong> contient les composants suivants : une bandelette de test<br />
<strong>PartoSure</strong> dans un sachet aluminium avec déshydratant, un écouvillon<br />
vaginal stérile oqué et un acon en plastique contenant une solution<br />
préparée à l’aide d’un solvant (0,9 % NaCl, 0,05 % NaN3, 0,01 % Triton X100).<br />
PRÉCAUTIONS ET MISES EN GARDE<br />
L’analyse <strong>PartoSure</strong> est réservée au diagnostic in vitro et aucun<br />
élément du test ne doit être utilisé de manière interne • Les instructions<br />
d’utilisation doivent être suivies rigoureusement ; la non-observance de<br />
ces instructions est susceptible de fausser les résultats • Des mesures de<br />
sécurité doivent être observées lors du prélèvement, de la manipulation<br />
et de l’élimination des échantillons réalisés avec le kit. L’utilisation de tout<br />
écouvillon autre que l’écouvillon stérile oqué fourni avec le kit<br />
altérera les résultats du test et par conséquent est strictement interdite.<br />
• N’utilisez pas des composants du kit s’ils sont endommagés • Les kits sont<br />
biologiquement dangereux ; éliminez tous les composants en prenant<br />
toutes les précautions nécessaires • N’utilisez pas le test au-delà de la<br />
date d’expiration notée sur l’emballage du produit • Les composants du<br />
kit sont à usage unique • Ne tordez pas et ne pliez pas la bandelette du<br />
test ni le sachet d’aluminium avec la bandelette à l’intérieur, cela pourrait<br />
endommager la bandelette et fausser les résultats.<br />
CONSERVATION ET STABILITÉ<br />
Conservez le kit <strong>PartoSure</strong> dans un endroit sec à une température<br />
comprise entre 4 et 24 °C (entre 40 et 75 °F). Ne congelez pas le test.<br />
Conservé dans son sachet d’aluminium à la température recommandée,<br />
le test reste stable jusqu’à la date d’expiration imprimée sur le sachet. La<br />
bandelette du test <strong>PartoSure</strong> doit être utilisée dans les six (6) heures une<br />
fois retirée de son sachet d’aluminium.<br />
CONTRÔLE QUALITÉ<br />
Les bandelettes du test <strong>PartoSure</strong> contiennent un mécanisme de<br />
contrôle procédural interne permettant de garantir leur e cacité<br />
analytique. L’apparition d’une ou deux bandes dans la zone de résultat<br />
de la bandelette du test con rme l’intégrité de la procédure du test et<br />
de ses composants.<br />
LIMITES DU TEST<br />
Les résultats des tests <strong>PartoSure</strong> sont qualitatifs et non quantitatifs.<br />
Aucune analyse quantitative ne devrait être faite en fonction de<br />
l’intensité du test ou des bandes de contrôle. Le test fonctionnera<br />
correctement même en présence de quantités in mes de sang sur<br />
l’écouvillon. En cas de quantités de sang importantes sur l’écouvillon,<br />
le test risque de ne pas fonctionner correctement et son usage n’est pas<br />
recommandé. <strong>PartoSure</strong> ne doit pas être utilisé dans les six heures qui<br />
suivent la prise d’un ovule ou d’une solution désinfectante vaginale. Les<br />
résultats doivent être utilisés conjointement avec d’autres informations<br />
cliniques. L’e cacité du test sur des patientes ne démontrant aucun signe<br />
ou symptôme de travail est inconnue. La présence de placenta prévia ainsi<br />
que la réalisation d’examens gynécologiques avant le prélèvement d’un<br />
échantillon peuvent fausser les résultats. L’e cacité du test <strong>PartoSure</strong> n’a<br />
pas été établie en cas de présence des contaminants suivants : méconium,<br />
crèmes antifongiques, suppositoires, lubri ants, hydratants, talc, ou huile<br />
pour bébés.<br />
VALEURS DE REFERENCE<br />
<strong>PartoSure</strong> détecte les traces de PAMG-1 humaine. Une quantité de<br />
PAMG-1 supérieure ou égale à 4 ng/μl présente dans les secrétions cervico-<br />
vaginales indique un risque élevé d’accouchement dans les sept<br />
prochains jours. Le seuil de sensibilité de 4 ng/μl a été validé lors d’une<br />
étude multicentrique internationale portant sur la corrélation entre la<br />
quantité de PAMG-1 présente dans les secrétions cervico-vaginales et<br />
l’intervalle entre le test et l’accouchement.<br />
DONNÉES SUR LES PERFORMANCES<br />
La performance clinique du test <strong>PartoSure</strong> a été déterminée dans une<br />
étude multicentrique internationale. Les patientes qui (i) présentaient des<br />
signes ou symptômes évoquant un travail prématuré, (ii) entre 20 et 36 6/7<br />
semaines d’aménorrhée, (iii) à membranes intactes d’après la clinique ont<br />
été invitées à participer à l’étude. Au total, 101 patientes ont été inclues<br />
dans l’analyse nale. Les estimations des performances suivantes ont été<br />
calculées avec un intervalle de con ance à 95%:<br />

RÉACTIVITÉ CROISÉE<br />
Les protéines suivantes ont subi un test de réactivité croisée avec<br />
les anticorps monoclonaux utilisés pour le test <strong>PartoSure</strong>:<br />
α2-microglobuline de fertilité, gonadotrophine chorionique humaine,<br />
s-1-glycoprotéine trophoblastique, lactogène placentaire humain,<br />
foetoproteine α, sérum albumine humaine et plusieurs protéines de<br />
liaison aux IGF. Les anticorps monoclonaux testés servant au test<br />
<strong>PartoSure</strong> n’ont pas présentés de réactivité croisée avec ces protéines.<br />
L’anticorps utilisé pour la bande du test a présenté une réactivité<br />
croisée à la protéine 3 de liaison aux IGF selon la méthode ELISA. La<br />
concentration d’IGFBP-3 dans les secrétions vaginales des femmes<br />
enceintes n’a ecte cependant pas la précision du test <strong>PartoSure</strong>.<br />
ÉTUDES SUR L’INTERFÉRENCE<br />
Les infections vaginales, l’urine, le sperme ainsi que les petits mélanges<br />
de sang n’interfèrent pas avec les résultats du test <strong>PartoSure</strong>. Ces<br />
substances et pathologies ont été évaluées lors d’études non cliniques<br />
et se sont révélées contenir de la PAMG-1 uniquement en concentrations<br />
inferieures au seuil de détection de <strong>PartoSure</strong>.<br />

INSTRUCTIONS POUR UN USAGE DIAGNOSTIQUE IN VITRO<br />
MÉTHODE DE TEST<br />
1. Prendre le acon de solvant au niveau du bouchon et s’assurer que tout le liquide descende au fond du acon.<br />
Ouvrir le acon de solvant et le maintenir en position verticale.<br />
2 A n de prélever un échantillon à partir du vagin, n’utiliser que l’écouvillon stérile oqué fourni avec le kit<br />
<strong>PartoSure</strong>. Retirer l’écouvillon de son emballage en suivant les instructions gurant sur ce dernier.<br />
L’extrémité de l’écouvillon ne doit être mise en contact avec aucune autre surface avant son insertion dans le<br />
vagin. Tenir l’écouvillon au milieu de la tige et introduire avec précaution l’extrémité de l’écouvillon dans le vagin<br />
lorsque la patiente est en position allongée jusqu’à ce que les doigts entrent en contact avec la peau (pas plus<br />
de 5 a 7 cm de profondeur). Retirer l’écouvillon du vagin après 30 secondes.<br />
3 Une fois l’écouvillon retiré, placer immédiatement l’extrémité dans le acon de solvant fourni a cet e et,<br />
puisrincer l’écouvillon en répétant plusieurs rotations pendant 30 secondes.<br />
4. Retirer l’écouvillon du acon et le jeter.<br />
5. Ouvrir le sachet d’aluminium au niveau des encoches et retirer la bandelette du test <strong>PartoSure</strong>.<br />
6 Plonger la partie blanche de la bandelette (indiquée par des èches pointées vers le bas) dans le<br />
acon de solvant.<br />
7 Retirer la bandelette dès que deux bandes sont clairement visibles dans la zone de test ou après cinq<br />
minutes. Lire les résultats en plaçant la bandelette du test sur une surface propre, sèche et plane.<br />
Deux bandes dans la zone de test indiquent un résultat positif tandis qu’une ligne dans la zone de test indique<br />
un résultat négatif. Ne pas lire ou interpréter les résultats au-delà de 10 minutes d’immersion de la bandelette du<br />
test dans le acon.<br />
PROCEDURA DEL TEST<br />
1. Prendere la ala del solvente per il cappuccio e assicurarsi che tutto il liquido nella ala sia caduto sul fondo.<br />
Aprire la ala del solvente e metterla in posizione verticale.<br />
2 Per raccogliere un campione dalla vagina, utilizzare esclusivamente il tampone in cotone sterile fornito con<br />
il kit di test <strong>PartoSure</strong>. Rimuovere il tampone dal suo contenitore seguendo le istruzioni sulla confezione. La<br />
punta del tampone non deve toccare nulla prima di essere inserita nella vagina. Tenere il tampone al centro del<br />
suo bastoncino; con la paziente in posizione supina, inserire con cautela la punta del tampone nella vagina no<br />

secondi.<br />
3 Dopo aver rimosso il tampone dalla vagina, immergere immediatamente la punta nella ala del solvente<br />
fornita e sciacquarlo ruotandolo per 30 secondi.<br />
4. Rimuovere il tampone dalla ala e smaltirlo.<br />
5. Aprire il sacchetto in alluminio strappando lungo la dentellatura ed estrarre la striscia per il test <strong>PartoSure</strong>.<br />
6 Immergere l’estremitá bianca della striscia (contrassegnata da frecce rivolte verso il basso) nella ala con il<br />
solvente.<br />
7 Rimuovere la striscia dalla ala se nella zona del test sono chiaramente visibili due linee o dopo 5<br />
minuti esatti. Leggere i risultati posizionando la striscia per il test su una super cie piatta, pulita e asciutta. Il<br />
risultato positivo è indicato da due linee nella zona del test, mentre il risultato negativo è indicato da una sola<br />
linea nella zona del test. Non leggere o interpretare i risultati dopo che sono trascorsi 10 minuti dall’immersione<br />
della striscia nella ala.<br />
TEST PROSEDÜRÜ<br />
1. Çözücü şişesini kapağından tutarak alın ve şişedeki tüm sıvının dip tarafta olduğundan emin olun. Çözücü<br />
şişesini açın ve dikey konumda yerleştirin.<br />
2 Vajinadan numune almak için, yalnızca <strong>PartoSure</strong> test kiti ile verilen steril pamuklu çubuğu kullanın.<br />
Ambalajındaki talimatları izleyerek çubuğu paketinden çıkarın. Çubuğun ucu, vajina içine sokulmadan önce<br />
herhangi bir şeyle temas etmemelidir. Hasta sırtüstü uzanırken, çubuğu sapının ortasından tutun ve parmaklar<br />
ciltle temas edene kadar çubuğun ucunu dikkatli bir şekilde vajina içine sokun (5-7 cm’den derine olmayacak<br />
şekilde). 30 saniye sonra çubuğu vajinadan çıkarın.<br />
3 Çubuk vajinadan çıkarıldıktan sonra, ucunu hemen verilen çözücü içine yerleştirin ve 30 saniye süreyle<br />
döndürerek ha fçe yıkayın.<br />
4. Çubuğu şişeden çıkarın ve atın.<br />
5. Folyo poşetini yırtma çentiklerinden yırtarak açın ve <strong>PartoSure</strong> test stripini çıkarın.<br />
6 Test stripinin beyaz ucunu (aşağıyı gösteren oklarla işaretli) çözücünün olduğu şişe içine sokun.<br />
7 Test alanında açıkça görülebilir iki çizgi oluştuğunda veya tam 5 dakika sonra test stripini çıkarın. Test<br />
stripini temiz, kuru, düz bir yüzeye yerleştirerek sonuçları okuyun. Negatif sonuç test alanında tek bir çizgi ile<br />
gösterilirken, pozitif sonuç test alanında iki çizgi ile gösterilir. Test stripinin şişe içine sokulmasının üzerinden 10<br />
dakika geçmesinden sonraya kadar sonuçları okumayın veya yorumlamayın.<br />
