b. Specimens with high protein content such as pleural, peritoneal or pericardial fluids<br />

can be preserved <strong>for</strong> 24 to 48 hours with refrigeration. The protein-rich fluid in which<br />

the cells are bathed acts as a tissue culture medium in preserving cellular morphology.<br />

c. Specimens with low mucus or protein content such as urine or CSF will be<br />

preserved <strong>for</strong> only 1-2 hours even if refrigerated. The fluid medium in which these<br />

cells are bathed contains enzymatic agents capable of causing cell destruction.<br />

Refrigeration may inhibit bacterial growth but does not protect the cells.<br />

d. Specimens with low pH, such as gastric material, must be collected on ice and be<br />

processed within minutes of collection to prevent cellular destruction by HCl.<br />
