providing the first unequivocal evidence for the existence<br />

of VRs (Fig. 6). This initial binding assay was,<br />

however, plagued by a very high nonspecific binding,<br />

which prevented the detection of VRs in the spinal cord<br />

or peripheral tissues.<br />

The methodological means to overcome the problems<br />

created by the high nonspecific RTX binding was furnished<br />

by an accidental observation of ours. In search of<br />

endogenous lig<strong>and</strong>s for VRs, we assayed a variety of<br />

body fluids <strong>and</strong> tissue extracts <strong>and</strong> found that the acute<br />

phase serum protein, 1-acid glycoprotein (also known<br />

as orosomucoid), binds RTX (Szallasi et al., 1992). 1-<br />

Acid glycoprotein is an important drug-binding protein<br />

(Paxton, 1983; Kremer et al., 1988; Maruyama et al.,<br />

1990). We showed that RTX binds to the well known<br />

drug-recognition domain on 1-acid glycoprotein that it<br />

shares with warfarin (Szallasi et al., 1992). Plasma<br />

binding of RTX to 1-acid glycoprotein may have clear<br />

consequences on pharmacokinetics upon systemic administration<br />

that we will discuss later. RTX binding to<br />

1-acid glycoprotein, however, differs in two very important<br />

aspects from VR binding. First, it is not markedly
