Iqbal ChoudharyAbstract: In the present study it is demonstrated that H 1- blockers such as triprolidine–HCl can bedetermined by a very simple, sensitive and accurate spectrophotometric procedure. The methodconsisted <strong>of</strong> interaction <strong>of</strong> triprolidine-HCl with dichloronitrobenzene in alkaline medium. Absorbance<strong>of</strong> resulting orange colour was measured at 440 nm .The reaction turned out to be selective for triprolidine–HCl with 0.005 mgml -1 as the visual limit <strong>of</strong> identification and provided a basis for a newspectrophotometric determination. The reaction obeyed Beer’s law from 0.05 mg to 0.15 mgml -1 fortriprolidine–HCl and the relative standard deviation was 0.60 %. The quantitative assessment <strong>of</strong> otherdrugs was also studied.Keywords: Triprolidine–HCl determination, dichloronitrobenzene, colour stability, analytical pharmacyIntroductionTriprolidine hydrochloride (Fig.1) is a pyridinederivative with the properties <strong>of</strong> antihistamine. It isa potent histamine H 1-receptor antagonist (H 1-blocker), with a rapid onset and long duration action,almost up to 12 hours. It is probably effective forthe symptomatic treatment <strong>of</strong> seasonal and perennialallergic rhinitis, vasomotor rhinitis, allergicconjunctivitis due to allergens, foods and prevention<strong>of</strong> allergic reactions to blood or plasma [1]. Themost common side effects are sedation, dizziness,incoordination, gastrointestinal disturbances, nausea,vomiting and diarrhea. It may also produce blurredvision, dryness <strong>of</strong> mouth, tightness <strong>of</strong> chest, blooddisorders including agranulocytosis andhaemolyticanaemia [2].In view <strong>of</strong> the importance <strong>of</strong> antihistamineconsiderable analytical work has been carried out.*Author for correspondenceIn the reversed phase HPLC [3,4] and HPLCphotodiode procedures [5], 12.5 cm Nucleosil 100t – 5C 18bonded phase column with a mobile phaseconsisting <strong>of</strong> methanol acetonitrile was used. A0.01M potassium dihydrogen phosphate solutionwas employed to adjust the pH to 6.8 [3] and UVdetection was carried out at 254 and 280 nm [4]. Inanother HPLC method different UV absorptioncharacteristics <strong>of</strong> triprolidine hydrochloride have beenused to facilitate its determination in a mixture [6]. Awavelength switching programme has to beemployed in this procedure. In the micro extractioncapillary GC procedure [7] headspace solid-phaseextraction was carried out followed by capillary gaschromatography with flame ionization detection. Therecoveries in blood extraction were 4 - 51 foldslower than those in urine extraction. In the capillaryelectrophoresis procedures [8,9] the quality <strong>of</strong>separation was dependent upon the sample diluentused [8] and only basic drugs are screened fromblood [9] while most <strong>of</strong> the spectrophotometric
