www.pelobiotech.com 2. Stem Cell Culture Media Induced pluripotent stem cells enable researchers to push the boundaries of in vitro models opening opportunities for clinical applications. The designation "iPS" (induced pluripotent stem) is applied to cells that mirror the morphology and growth properties of embryonic stem (ES) cells while expressing essential ES cell marker genes, as elucidated by Takahashi & Yamanaka in 2006. iPSCs can be cultured in various types of media ranging from classic DMEM with specific supplements and serum added to fully chemically defined media. Furthermore, defined media makes the process reliable and without batch effects. We offer media in our own Cellovations® brand and from different suppliers for ES/iPSCs and iPS-derived cells. For iPSCs, feeder cells are often utilized to help maintain their growth while they remain being pluripotent; however, iPSCs can be cultured in either feeder-layer or feeder-free conditions. 2.1 Feeder-dependent Culture Growth factors are needed to foster the growth and proliferation of stem cells. One way of providing these growth factors is through the direct addition of factors such as bFGF into the media. An alternative is with the application of feeder cells which are able to provide growth factors, cytokines, and extracellular matrix components all of which facilitates cell attachment and overall increases cell viability. Interestingly enough, feeder cells have also been shown to preserve the stem cell telomere integrity better ensuring that there is more reliable capabilities of self-renewal (as shown in the Guo et al, 2018 paper published in Nature Communications).
