Identification and Characterization of MajorProteins Carrying <strong>ABO</strong> Blood Group Antigens inthe Human Kidney.Localization and immunoblotting analysis of <strong>ABO</strong>antigens in the cortex of human kidney. (A) Thelocalization of <strong>ABO</strong> antigens was examined in the cortexof normal human kidney by immunohistochemistry usingmouse monoclonal anti-A antibody. Most of vascularendothelial cells of arteries, veins, glomerulus, andperitubular capillaries were stained (a-c), and intensestaining was observed in epithelial cells of convoluteddistal tubules and collecting ducts. (B) Immunoreactivitywas compared between sections without (a and c) orwith preexposure to ethanol (b and d). (C) Localization ofimmunoreactivity was further examined byimmunoelectron microscopy using rabbit polyclonal anti-A. (D and E) Immunoblotting analysis with anti-bloodtype B antibody was conducted using lysate from thecortex of normal human kidney. The lysate wasseparated on a 5% to 15% linear gradient SDS-PAGE gel.The immunoreactive proteins were clearly observed bylong exposure of immunoblot to X-ray film and could bedetected as protein bands with approximate molecularmasses of 270, 220, 180, 150, 130, 116, 100, and 60 kDa(D). (E) The lysate without (control) and with digestion byglycopeptidase F were separated on a 5% to 15% lineargradient SDS-PAGE gel and processed forimmunoblotting with anti-B antibody.
