For all standard stirred cells<br />

<strong>Generon</strong>’s ultrafiltration membranes are a series of anisotropic<br />

membranes specially selected following research into the<br />

performances of existing membrane discs in the market.<br />

The membrane has been proprietary-treated to ensure low non-specific<br />

protein binding at higher speed.<br />

The membrane discs are available in a complete range of diameters for<br />

stirred cells and for other filtration products which require ultrafiltration membranes.<br />

The hydrophilic regenerated cellulose membrane discs offer superior<br />

chemical resistance and are preferred for highest protein<br />

recovery when handling dilute protein solutions.<br />

The polyethersulphone membrane discs have a more open structure which provides fast separation<br />

flux. Use the polyethersulphone membranes for fast protein separation over a broad pH range.<br />

Two-Compartment Bioreactor for Efficient Cell Cultivation<br />

Description<br />

The CELLine bioreactor is a disposable,<br />

two-compartment cultivation device suitable<br />

for many cell culture applications, e. g. the<br />

production of monoclonal antibodies on a<br />

laboratory scale. Two different sizes of the<br />

CELLine are available: CELLine CL350 and<br />

CL 1000. Details are listed below.<br />

Introduction<br />

Efficient cell cultivation is dependent on<br />

an optimal supply of oxygen and nutrients,<br />

as well as an efficient removal of inhibiting<br />

metabolic waste products. In static cell<br />

culture the optimal balance of these factors<br />

is not given. These limitations lead to a<br />

reduced cell growth and are unfavorable for<br />

achieving high levels of protein expression.<br />

Features & Benefits<br />

The two-compartment bioreactor CELLine<br />

is designed to overcome these limitations<br />

by dividing the bioreactor into a medium<br />

compartment and a cell compartment. A<br />

semi-permeable membrane (10 kDa MWCO)<br />

between the compartments allows small<br />

molecules to diffuse from one compartment<br />

to the other. Higher molecular weight molecules<br />

secreted by the proliferating cells are<br />

retained within the cell compartment. This<br />

results in a continuous flow of nutrients<br />

into the cell compartment and a concurrent<br />

removal of any inhibitory waste products.<br />

The cell compartment and the medium compartment<br />

are individually accessible which<br />

enables the cells to be supplied with fresh<br />

medium according to their individual needs.<br />

In addition, the bottom of the cell compartment<br />

is made of a gas permeable membrane<br />

which ensures an optimal oxygen supply<br />

to and carbon dioxide exchange from the<br />

cell culture. All together, this bioreactor<br />

construction allows optimal maintenance<br />

of the cells at quasi in vivo cultivation<br />

conditions. These conditions enable cells<br />

to proliferate to very high densities within<br />

the cell compartment.<br />

Application<br />

The CELLine is perfectly suited for a wide<br />

range of applications involving suspension<br />

cell culture, like monoclonal antibody<br />

production or long-term continuous culture<br />

maintenance. The unit is optimised for<br />

cultivation of hybridoma, CHO (Chinese<br />

hamster ovary) cells, NSO (mouse myeloma)<br />

cells, SF (Spodoptera frugiperda, insect cells)<br />

cells and many more.<br />

High Cell Density Cultivation<br />

With the two-compartment technology of<br />

CELLine, deficiencies in nutrition or oxygenation<br />

are problems of the past. Therefore,<br />

cell densities inside the cell compartment are<br />

usually between 10 7 to 10 8 cells per ml which<br />

is about two magnitudes higher than with<br />

conventional static cell culture techniques.<br />

High product concentrations<br />

Benefiting from the high cell densities,<br />

product concentrations achieved in CELLine<br />

are in the range of 1 to 5 mg per ml, making<br />

the bioreactor most suitable for antibody<br />

expression in hybridomas, protein expression<br />

in transfected cell lines, or virus production.<br />

Product concentrations in the cell compartment<br />

of CELLine are in most cases 50–100<br />

times above those found in static cell culture<br />

disposables, such as flasks and roller bottles.<br />

Long-term, continuous culture<br />

maintentance<br />

Separate access to the medium compartment<br />

of CELLine allows for continuous delivery<br />

and efficient oxygenation of the cells, thus<br />

enabling the user to perform continuous<br />

studies on a specific cell culture, or to benefit<br />

from a consistent protein production over<br />

several weeks.

Proteus ‘1-step batch’ Midi Spin Column Pack<br />

Any Resin<br />

1. Remove the clear TOP spin cap and load the required amount of resin. Add 10 ml<br />

binding buffer, replace the clear TOP spin cap and spin at 200 g for 2 min to equlibrate<br />

the resin matrix.<br />

2. Carefully remove the spin column barrel and push the beige BOTTOM cap on to the end<br />

of the tapered part of the spin column. Place the spin column into a fresh 50 ml<br />

centrifuge tube.<br />

3. Remove the clear TOP spin cap, load the sample, either filtered or unfiltered, into the<br />

upper reservoir (containing the resin) and screw the yellow TOP cap tightly on to the<br />

centrifuge tube.<br />

4. Place the column on a standard tube roller and mix for a required time. Most<br />

applications require only a 30 min incubation.<br />

5. After batch incubation, replace the yellow TOP cap with the clear TOP spin cap.<br />

Carefully remove the spin column barrel from the centrifuge tube and the beige<br />

BOTTOM cap from the tapered end of the spin column. Place the spin column barrel<br />

into a 50 ml centrifuge tube and spin at 200g for 2 min. Remove the filtrate.<br />

6. Add 15 ml wash buffer. Wash off the unbound protein at 200 g for 2 min. Remove the<br />

filtrate. Repeat this step.<br />

7. Place the spin column barrel into a fresh tube. Add up to 10 ml elution buffer to the<br />

upper reservoir and elute the purified protein at 200 g for 2 min.<br />

<strong>Generon</strong>’s Proteus ‘1-step-batch’ spin columns<br />

allow you to batch mix your filtered or<br />

unclarified sample with the resin of your choice<br />

and then wash and elute your purified and<br />

concentrated protein in a small volume. These<br />

columns allow you to process multiple samples<br />

in parallel. The spin column barrels fit all 50 ml<br />

conical centrifuge tubes and can be used in<br />

standard bench centrifuges containing a swing<br />

bucket rotor.<br />

There is no resin carry-over to a subsequent downstream analyses, no messy transfer of<br />

samples/resin slurries and negligible hold-up volume. As the device is contained, it is ideal for<br />

handling biohazardous samples and samples from infectious sources.<br />

(High Capacity) Spin Column Kits: The Gold Standard<br />

Residual endotoxin contamination in advanced biotherapy products is an expensive and<br />

often difficult contaminant to control.<br />

Many commercially-available protocols are unable to remove endotoxins effectively<br />

and are based on non-affinity chromatography methods e.g. Ion exchange<br />

chromatography, phase separation using Triton X-114 or require time consuming and<br />

expensive affinity steps. These costly resins are often supplied as loose resin or packed<br />

in slow gravity columns.<br />

The Proteus NoEndo TM spin column kits offer a standardised method for high grade<br />

clearance of endotoxin from recombinant proteins, antibodies and viral vectors. These<br />

agents are increasingly being designed for therapeutic applications, hence moving them<br />

forward efficiently through in vivo studies requires pure preparations of the samples.<br />

LPS molecule<br />

Next generation Proteus kits combine the quality separation you expect from gravity flow columns with the speed and<br />

ease-of-use of spin columns. Both column formats reveal a high degree of innovation! The Standard and High Capacity<br />

columns incorporate pre-packed resin cartridges utilizing our FlowGo TM technology. The Mini columns are empty<br />

columns that incorporate our proprietary SelfSeal TM membrane technology. This ensures that there is no passage of<br />

the sample through the membrane during the batch incubation at 4 0 C or at room temperature.<br />

NoEndoS and NoEndoHC Columns:<br />

The proprietary FlowGo TM technology regulates sample movement through the technologically-advanced affinity resin<br />

cartridge, increasing both endotoxin removal and protein recovery. Uniquely, we offer flow rate control for endotoxin<br />

removal in a centrifuge.<br />

NoEndoM Columns:<br />

The NoEndo TM Mini columns incorporate a SelfSeal TM membrane technology which retains the NoEndo TM resin and<br />

sample in the batch incubation chamber. When the column spun in a low speed centrifuge, the pores of the<br />

membrane dilate and the filtered eluate is collected in the bottom of the centrifuge tube.<br />

When to use Proteus NoEndo TM Mini, NoEndo TM Standard or NoEndo TM High Capacity spin columns?<br />

The Proteus NoEndo TM columns have different endotoxin binding capacities. For samples with endotoxin loads less<br />

than 3,000 EU, Proteus NoEndo TM M columns are ideal. For samples with endotoxin loads less than 30,000 EU, Proteus<br />

The Peristaltic Pump Collar<br />

(Optional Accessories)<br />

Large volumes of dilute and particle-laden samples can be loaded on to Proteus spin columns using the peristaltic pump<br />

collar. The peristaltic pump collar (Fig. 1) locks the Proteus sealing cap securely to the spin column barrel to ensure that<br />

high sample loading flow rates can be used (Fig. 2).<br />

Materials Included:<br />

Peristaltic Pump Collar: Black Acetal<br />

Peristaltic Pump Tubing: Nalgene 180 tubing (L/S 16, 4 ft). Tubing is configured for 0.8 ml/rev. For example<br />

when taking into account the back pressure exerted by the resin column, 10-20 rpm<br />

would provide a flow rate of approximately 5-8 ml/min.<br />

Required equipment:<br />

Peristaltic pump: Variable speed peristaltic pump capable of speeds less than 20 rpm e.g. Masterflex L/S<br />

pump.<br />

Peristaltic pump head: Pump head designed for L/S 16 tubing E.g. Masterflex L/S Easy-Load pump head for<br />

precision tubing. Pump head can accommodate L/S 13, L/S 14, L/S 16, L/S 25, L/S 17,<br />

and L/S 18 precision tubing.<br />

Operation:<br />

Load the correct size tubing in to the peristaltic pump head. (For optimum tubing life, keep the tubing straight where it<br />

exits and enters the pump)<br />

Insert the white fittings on to the end of the peristaltic pump tubing. Connect one end of the tubing tightly to the barb<br />

connector on the sealing cap. Place the other end of the tubing into the filtered sample reservoir.<br />

Fill the column barrel with 18 ml filtered sample. Push the sealing cap fully into the top of the column barrel and then<br />

slide the peristaltic pump collar on to the sealing cap. Turn the collar (a near ¼ turn) until one of its legs slides under the<br />

hinge of the column barrel lid (Fig. 2). Ensure that the collar is positioned over the top rim of the column barrel.<br />

Run the peristaltic pump at the desired flow rates noting that 10-20 rpm (using the Type 16 tubing) is equivalent to flow<br />

rates of approximately 5-8 ml/min.<br />

The Proteus spin column can be run in peristaltic mode only. All you need to purchase is the peristaltic pump collar,<br />

sample loading cap and peristaltic pump tubing configured in cat no. PC-PPCSC (Fig. 2).

The gel matrix of Centri•Pure is<br />

Zetadex�25, a beaded composite<br />

material developed by emp BIOTECH<br />

and composed partially of polymerized<br />

dextran. It exhibits high selectivity, high<br />

resolution and chemical stability.<br />

Molecules purified with Zetadex�25 are<br />

separated according to size. Smaller<br />

molecules pass significantly slower<br />

through the column than larger<br />

molecules. Buffer and pH effects on resolution are minimal.<br />

The size exclusion cut�off for Zetadex�25 is 10 kD for proteins. Proteins larger than<br />

10 kD in a sample volume of up to 2.5 mL can be purified with an elution volume of<br />

Four Step Protocol<br />

1. Column Preparation<br />

Remove the cap from the top and then the<br />

bottom cap of the Centri•Pure P25 Column.<br />

Allow excess column fluid to drain (via gravity)<br />

into a suitable waste reservoir.<br />

3. Sample Application<br />

Transfer 2.5 mL sample to the Centri•Pure P25<br />

Column. Allow the sample to enter the gel bed<br />

completely.<br />

2. Column Equilibration<br />

Equilibrate the column with approximately<br />

25 mL of the required buffer (use the same<br />

buffer for both equilibration and elution).<br />

Allow the equilibration buffer to enter the<br />

4. Elution<br />

Place a tube for sample collection under the<br />

Centri•Pure P25 Column. Transfer 3.5 mL of<br />

elution buffer to the column and elute the<br />

NuSep nUView Tris-Glycine (TG) precast gels offer improved resolution and<br />

increased mass range<br />

NuSep’s nUView Tris-Glycine (TG) gels are based on<br />

a reformulation of our popular iGel. nUView TG gels<br />

offer improved resolution, broader mass range,<br />

decreased run time and increased stability.<br />

Separations using nUView TG gels can be carried out<br />

under native or denaturing conditions using a broad<br />

array of popular buffer systems. In this study we<br />

compare the resolution and migration patterns of<br />

nUView TG gels with the Bio-Rad Mini-PROTEAN®<br />

TGX TM and Invitrogen NuPAGE precast gels.<br />

Comparison of migration patterns<br />

nUView TG gels come in a variety of well<br />

configurations and acrylamide formulations (Figure 1).<br />

One of the most popular formulations is the 4-20%<br />

because it offers a broad molecular weight range and<br />

separation characteristics suitable for a wide range of<br />

proteins. We first compared the migration patterns<br />

between the nUView Tris-Glycine, Bio-Rad Mini-<br />

PROTEAN TGX and Invitrogen NuPage precast gels<br />

using the Invitrogen Mark 12 protein standards (Figure<br />

2). Excellent separation performance is achieved with<br />

the nUView TG gels when compared to the TGX and<br />

NuPAGE gels. At the lower MW region (4-2kDA)<br />

nUView TG gels achieve higher resolution than both<br />

the TGX and NuPAGE gels with both insulin chains<br />

completely resolved. Furthermore separation with<br />

nUView TG is achieved across the entire length of the<br />

gel providing a broader MW range from 200kDA to<br />

2kDa.<br />

Comparison of resolution<br />

Gel selection is dependent not only on MW range but on<br />

achievable resolution for the sample of interest. To evaluate<br />

the resolution of nUView TG gels a variety of protein<br />

samples were subjected to SDS page (Figure 4).<br />

Resolution was then compared between the nUView TG,<br />

Bio-Rad TGX, and Invitrogen NuPAGE using the SDS page<br />

analysis of Pisum sativum extract. Comparison of the<br />

banding pattern indicated an increased resolution for the<br />

nUView TG gels, evident by the increase in protein band<br />

separation and number (Figure 5).<br />

Simple 1-step Protocol<br />

Remove the gel from the cassette and place<br />

gel into container.<br />

Cover gel with 25 ml QC stain and leave for<br />

minimum 15 minutes or until all weak<br />

protein bands are fully developed.<br />

Store the gel in the QC stain overnight.<br />

Then, either leave in QC stain or transfer to<br />

DI water for gel storage. (N.B. A minimum 1<br />

hour full stain is recommended before<br />

storing the gel in water.)<br />

Lysed bacterial cell pellet in PBS, 1% Triton X-100 and protease inhibitors (1:500). Samples sonicated at 20% amplitude for<br />

GENERON Detergents for Protein Purifications, Reconstitutions and Crystallizations<br />

GENERON has teamed up with Glycon, who have developed new procedures for the preparation of ultra pure<br />

alkyl-β-glycoside-detergents. All α-isomers are completely removed by chromatography. All of our detergents are<br />

crystallized to greater than 99.5% purity. The re-crystallization procedure removes all side products and<br />

lipid alcohols eg dodecanol and the final detergents are presented in a lyophilized form. There is no α−isomer<br />

present in the Glycon detergents.<br />

The anomeric purity of the detergent is critical for the isolation and crystallization of membrane proteins<br />

DETERGENT ANALYSES<br />

All of our detergents are crystallised to be greater than 99.5% pure as measured by TLC, HPLC,<br />

double melting point and by the following analytical procedures.<br />

Measurement of purity (HPLC, TLC)<br />

Each lot is first analysed by TLC and HPLC. All detergents are greater than 99.5% pure. TLC was<br />

performed on silica gel (Merck 60 F (254), dichloromethane/methanol 7:1) and detection was effected<br />

by spraying the TLC plate with a solution of ethanol-sulfuric acid (9:1) followed by heating. HPLC was<br />

performed on a standard C18 column with an eluant of methanol/water. The ration of methanol to<br />

water may vary from 65/35 to 85/15 depending on the hydrophobicity of the detergent. A 50% solution<br />

of detergent in MeOH is injected into the HPLC system. [P. Fromme, H. T. Witt, Improved isolation and<br />

Alcohol and α-isomer contamination<br />

β-n-Alkylglycoside detergents are prepared by glycosylation of the hydrophobic alcohol with activated<br />

glucose or maltose acetates. Nearly all products are purified chromatographically and by<br />

crystallisation. Alcohols and α-isomers are completely removed during the manufacturing process.<br />

Trace amounts of this alcohol in the detergent lot can cause cloudiness in the detergent solution. The<br />

β-anomeric purity of the detergent is advantageous for the crystallization of membrane proteins.<br />

Measurement of purity (Double melting point)<br />

The double melting properties of β-n-alkylglycosides have long been known (E. Fischer, B. Helferich;<br />

Liebigs Ann. Chem. 383, 68 (1911). Alkylglycosides have liquid crystal properties and they can form<br />

mesophases in the melt (a state of order between crystals and liquids). In the smectic A mesophase<br />

the amphiphilic rod-like molecules form layered structures - a microscopic separation by a sequence<br />

of hydrophilic and lipophilic layers. In the isotropic phase, the molecules are packed as a liquid in<br />

a random way. An optical polarised microscope equipped with a hotstage and a central processor<br />

is used to identify transition temperatures from the smectic A to the isotropic phase. Measurement of<br />

transition temperatures (double melting temperatures) gives a very sensitive test for the contamination<br />

of the crystalline detergent with hydrophobic alcohols, α-glycosides, solvents and others organic/<br />

inorganic impurities.<br />

Absorbance<br />

Alkyl-glucosides and -maltosides have low absorbance throughout the UV region. MEGA detergents,<br />

containing amido group, have high absorbance at 230 nm. The absorbance of the 1% w/v detergent<br />

solution in water is measured in the UV region.<br />

Fluorescence<br />

A very low level of aromatic impurity can still result in a large fluorescence background. The<br />

fluorescence of the detergent solution is compared to a standard 0.1% BSA solution. The excitation<br />

wavelength is 280 nm and the emission is measured at 345 nm.<br />

Conductance<br />

Measurement of conductance gives a very sensitive test for the presence of ionic impurities. A solution<br />

of non-ionic detergent should have conductance nearly the same as de-ionised water.<br />

Measurement of pH<br />

The pH of the detergent solution at appropriate concentration is measured. The pH value of the<br />

solution should be neutral.<br />

Solubility in water<br />

Some of the impurities (e.g. hydrophobic alcohol) in detergent preparations are not soluble in water.<br />

Therefore cloudiness of a detergent solution at a concentration where it is known to be soluble<br />

indicates the presence of an insoluble impurity.<br />

bleed, 4 immunizations and 3 production bleeds for<br />

For Chickens, the 56 day protocol includes 1 pre-immune<br />

bleed, 4 immunizations, 3 test bleeds and egg collection<br />

(approximately 14 eggs). IgY purification from egg yolks is also<br />
