Our Grade 1 classification implies that we have seen no impurities when at least 100 µg
lipid has been run on a thin layer plate and subsequently charred by a technique which
reveals the presence of 1 µg lipid. Grade 2 products are made either by simpler methods
or batches which just fail to pass our tests as Grade 1.
Vivaspin Ultrafiltration Family –
sample concentration up to 20 ml
Vivaspin centrifugal concentrators offer
the optimal solution to any concentration
or buffer exchange application with
their broad range of volume capacities
and MWCOs.
Highest flow rates and recoveries are
achieved due to their particular vertical
membrane design.
Incomparably safe sample handling and
convenience is assured by the patented
dead-stop technology allowing direct
recovery of the concentrate without
further respin.
Feature Benefit
Vivaspin applications
– Concentration of biological samples
containing antigens, antibodies,
enzymes or nucleic acids
– Concentration of diluted protein
samples from chromatography eluates
– Desalting or Diafiltration |
Buffer exchange
– Sample concentration for crystallization
and NMR spectroscopy
– Removal of very small or large
contaminants
Vertical, large membrane design Fast, minimal membrane blocking
Integrated Dead-Stop-Volume No risk (to concentrate to dryness)
Low-binding materials High recovery
No Re-spin necessary Easy handling, fast
Membrane selection Highest recovery with extremely
low-binding Hydrosart ® membrane in
Vivaspin 2 and Vivaspin 15R
Broad volume range from 0.6 – 100 ml High volume flexibility
Wide MWCO range from 2 kDa – 0.2 µm High MWCO flexibility
Special Devices
Vivascience offers an extended range of
membranes to cover the different ultrafiltration
requirements which will typically
yield recoveries of the concentrated
sample in excess of 90% when the starting
sample contains over 0.1 mg/ml
of the solute of interest.
For highest recovery, select a membrane
MWCO which is at least half to one third
of the molecular weight of the solute to
be retained.
Highest recovery with extremely
low binding Hydrosart ® membrane
– The membrane of choice for antibody
concentration
– MWCO range from 2 kDa – 30 kDa
discs in the
market
Ultrafiltration Membrane Discs
For all standard stirred cells
Generon’s ultrafiltration membranes are a series of anisotropic
membranes specially selected following research into the
performances of existing membrane discs in the market.
The membrane has been proprietary-treated to ensure low non-specific
protein binding at higher speed.
The membrane discs are available in a complete range of diameters for
stirred cells and for other filtration products which require ultrafiltration membranes.
The hydrophilic regenerated cellulose membrane discs offer superior
chemical resistance and are preferred for highest protein
recovery when handling dilute protein solutions.
The polyethersulphone membrane discs have a more open structure which provides fast separation
flux. Use the polyethersulphone membranes for fast protein separation over a broad pH range.
Alvetex-3D Advantage
Alvetex 3-D Cell Culture
alvetex ® is a highly porous
polystyrene scaffold engineered
into a 200μm thick membrane
which provides a 3D space into
which cells can invade, proliferate
and grow. The vertical axis of the
third dimension essentially enables
additional layers of cells to form
complex 3D interactions with
adjacent cells simulating the
structure of a tissue.
alvetex ® can be used for routine 3D cell culture, as an inert plastic.
alvetex ® is a 3D culture product which can be treated in the same manner
as traditional 2D cell culture plastic. The scaffold can be plasma-treated
without any detrimental effect to its structure.
alvetex ® can be sterilised using gamma radiation. The use of polystyrene
as a cell growth substrate is well accepted and recognised.
Cell Culture
CELLine
Two-Compartment Bioreactor for Efficient Cell Cultivation
Description
The CELLine bioreactor is a disposable,
two-compartment cultivation device suitable
for many cell culture applications, e. g. the
production of monoclonal antibodies on a
laboratory scale. Two different sizes of the
CELLine are available: CELLine CL350 and
CL 1000. Details are listed below.
Introduction
Efficient cell cultivation is dependent on
an optimal supply of oxygen and nutrients,
as well as an efficient removal of inhibiting
metabolic waste products. In static cell
culture the optimal balance of these factors
is not given. These limitations lead to a
reduced cell growth and are unfavorable for
achieving high levels of protein expression.
Features & Benefits
The two-compartment bioreactor CELLine
is designed to overcome these limitations
by dividing the bioreactor into a medium
compartment and a cell compartment. A
semi-permeable membrane (10 kDa MWCO)
between the compartments allows small
molecules to diffuse from one compartment
to the other. Higher molecular weight molecules
secreted by the proliferating cells are
retained within the cell compartment. This
results in a continuous flow of nutrients
into the cell compartment and a concurrent
removal of any inhibitory waste products.
The cell compartment and the medium compartment
are individually accessible which
enables the cells to be supplied with fresh
medium according to their individual needs.
In addition, the bottom of the cell compartment
is made of a gas permeable membrane
which ensures an optimal oxygen supply
to and carbon dioxide exchange from the
cell culture. All together, this bioreactor
construction allows optimal maintenance
of the cells at quasi in vivo cultivation
conditions. These conditions enable cells
to proliferate to very high densities within
the cell compartment.
Application
The CELLine is perfectly suited for a wide
range of applications involving suspension
cell culture, like monoclonal antibody
production or long-term continuous culture
maintenance. The unit is optimised for
cultivation of hybridoma, CHO (Chinese
hamster ovary) cells, NSO (mouse myeloma)
cells, SF (Spodoptera frugiperda, insect cells)
cells and many more.
High Cell Density Cultivation
With the two-compartment technology of
CELLine, deficiencies in nutrition or oxygenation
are problems of the past. Therefore,
cell densities inside the cell compartment are
usually between 10 7 to 10 8 cells per ml which
is about two magnitudes higher than with
conventional static cell culture techniques.
High product concentrations
Benefiting from the high cell densities,
product concentrations achieved in CELLine
are in the range of 1 to 5 mg per ml, making
the bioreactor most suitable for antibody
expression in hybridomas, protein expression
in transfected cell lines, or virus production.
Product concentrations in the cell compartment
of CELLine are in most cases 50–100
times above those found in static cell culture
disposables, such as flasks and roller bottles.
Long-term, continuous culture
maintentance
Separate access to the medium compartment
of CELLine allows for continuous delivery
and efficient oxygenation of the cells, thus
enabling the user to perform continuous
studies on a specific cell culture, or to benefit
from a consistent protein production over
several weeks.

Any Resin
1. Remove the clear TOP spin cap and load the required amount of resin. Add 10 ml
binding buffer, replace the clear TOP spin cap and spin at 200 g for 2 min to equlibrate
the resin matrix.
2. Carefully remove the spin column barrel and push the beige BOTTOM cap on to the end
of the tapered part of the spin column. Place the spin column into a fresh 50 ml
centrifuge tube.
3. Remove the clear TOP spin cap, load the sample, either filtered or unfiltered, into the
upper reservoir (containing the resin) and screw the yellow TOP cap tightly on to the
centrifuge tube.
4. Place the column on a standard tube roller and mix for a required time. Most
applications require only a 30 min incubation.
5. After batch incubation, replace the yellow TOP cap with the clear TOP spin cap.
Carefully remove the spin column barrel from the centrifuge tube and the beige
BOTTOM cap from the tapered end of the spin column. Place the spin column barrel
into a 50 ml centrifuge tube and spin at 200g for 2 min. Remove the filtrate.
6. Add 15 ml wash buffer. Wash off the unbound protein at 200 g for 2 min. Remove the
filtrate. Repeat this step.
7. Place the spin column barrel into a fresh tube. Add up to 10 ml elution buffer to the
upper reservoir and elute the purified protein at 200 g for 2 min.
Generon’s Proteus ‘1-step-batch’ spin columns
allow you to batch mix your filtered or
unclarified sample with the resin of your choice
and then wash and elute your purified and
concentrated protein in a small volume. These
columns allow you to process multiple samples
in parallel. The spin column barrels fit all 50 ml
conical centrifuge tubes and can be used in
standard bench centrifuges containing a swing
bucket rotor.
There is no resin carry-over to a subsequent downstream analyses, no messy transfer of
samples/resin slurries and negligible hold-up volume. As the device is contained, it is ideal for
handling biohazardous samples and samples from infectious sources.
Proteus NoEndo TM M (Mini), NoEndo TM S (Standard) and NoEndo TM HC
(High Capacity) Spin Column Kits: The Gold Standard
Residual endotoxin contamination in advanced biotherapy products is an expensive and
often difficult contaminant to control.
Many commercially-available protocols are unable to remove endotoxins effectively
and are based on non-affinity chromatography methods e.g. Ion exchange
chromatography, phase separation using Triton X-114 or require time consuming and
expensive affinity steps. These costly resins are often supplied as loose resin or packed
in slow gravity columns.
The Proteus NoEndo TM spin column kits offer a standardised method for high grade
clearance of endotoxin from recombinant proteins, antibodies and viral vectors. These
agents are increasingly being designed for therapeutic applications, hence moving them
forward efficiently through in vivo studies requires pure preparations of the samples.
LPS molecule
Next generation Proteus kits combine the quality separation you expect from gravity flow columns with the speed and
ease-of-use of spin columns. Both column formats reveal a high degree of innovation! The Standard and High Capacity
columns incorporate pre-packed resin cartridges utilizing our FlowGo TM technology. The Mini columns are empty
columns that incorporate our proprietary SelfSeal TM membrane technology. This ensures that there is no passage of
the sample through the membrane during the batch incubation at 4 0 C or at room temperature.
NoEndoS and NoEndoHC Columns:
The proprietary FlowGo TM technology regulates sample movement through the technologically-advanced affinity resin
cartridge, increasing both endotoxin removal and protein recovery. Uniquely, we offer flow rate control for endotoxin
removal in a centrifuge.
NoEndoM Columns:
The NoEndo TM Mini columns incorporate a SelfSeal TM membrane technology which retains the NoEndo TM resin and
sample in the batch incubation chamber. When the column spun in a low speed centrifuge, the pores of the
membrane dilate and the filtered eluate is collected in the bottom of the centrifuge tube.
We offer three versions of the Proteus kits: Mini, Standard and High Capacity
Proteus NoEndo TM Mini: for endotoxin loads less than 3,000 EU
Proteus NoEndo TM Standard: for endotoxin loads less than 30,000 EU
Typical protein recoveries close to 90% were obtained.
When to use Proteus NoEndo TM Mini, NoEndo TM Standard or NoEndo TM High Capacity spin columns?
The Proteus NoEndo TM columns have different endotoxin binding capacities. For samples with endotoxin loads less
than 3,000 EU, Proteus NoEndo TM M columns are ideal. For samples with endotoxin loads less than 30,000 EU, Proteus
NoEndo TM S can be used. For samples with endotoxin loads less than 1,000,000 EU, Proteus NoEndo TM HC columns are
required.
Technical Data and Operating Instructions:
The Peristaltic Pump Collar
(Optional Accessories)
Large volumes of dilute and particle-laden samples can be loaded on to Proteus spin columns using the peristaltic pump
collar. The peristaltic pump collar (Fig. 1) locks the Proteus sealing cap securely to the spin column barrel to ensure that
high sample loading flow rates can be used (Fig. 2).
Materials Included:
Peristaltic Pump Collar: Black Acetal
Peristaltic Pump Tubing: Nalgene 180 tubing (L/S 16, 4 ft). Tubing is configured for 0.8 ml/rev. For example
when taking into account the back pressure exerted by the resin column, 10-20 rpm
would provide a flow rate of approximately 5-8 ml/min.
Required equipment:
Peristaltic pump: Variable speed peristaltic pump capable of speeds less than 20 rpm e.g. Masterflex L/S
pump.
Peristaltic pump head: Pump head designed for L/S 16 tubing E.g. Masterflex L/S Easy-Load pump head for
precision tubing. Pump head can accommodate L/S 13, L/S 14, L/S 16, L/S 25, L/S 17,
and L/S 18 precision tubing.
Operation:
Fig. 1
Load the correct size tubing in to the peristaltic pump head. (For optimum tubing life, keep the tubing straight where it
exits and enters the pump)
Insert the white fittings on to the end of the peristaltic pump tubing. Connect one end of the tubing tightly to the barb
connector on the sealing cap. Place the other end of the tubing into the filtered sample reservoir.
Fill the column barrel with 18 ml filtered sample. Push the sealing cap fully into the top of the column barrel and then
slide the peristaltic pump collar on to the sealing cap. Turn the collar (a near ¼ turn) until one of its legs slides under the
hinge of the column barrel lid (Fig. 2). Ensure that the collar is positioned over the top rim of the column barrel.
Run the peristaltic pump at the desired flow rates noting that 10-20 rpm (using the Type 16 tubing) is equivalent to flow
rates of approximately 5-8 ml/min.
The Proteus spin column can be run in peristaltic mode only. All you need to purchase is the peristaltic pump collar,
sample loading cap and peristaltic pump tubing configured in cat no. PC-PPCSC (Fig. 2).

Centri•Pure P25 Gel Filtration Columns
are designed for rapid and efficient
removal of small molecules (salts, dyes,
ammonia, haptens, biotin, etc.) from
antibodies, enzymes and other proteins.
The gel matrix of Centri•Pure is
Zetadex25, a beaded composite
material developed by emp BIOTECH
and composed partially of polymerized
dextran. It exhibits high selectivity, high
resolution and chemical stability.
Molecules purified with Zetadex25 are
separated according to size. Smaller
molecules pass significantly slower
through the column than larger
molecules. Buffer and pH effects on resolution are minimal.
The size exclusion cutoff for Zetadex25 is 10 kD for proteins. Proteins larger than
10 kD in a sample volume of up to 2.5 mL can be purified with an elution volume of
2.7 to 3.5 mL.
NuSep nUView Tris-Glycine (TG) precast gels offer improved resolution and
increased mass range
NuSep’s nUView Tris-Glycine (TG) gels are based on
a reformulation of our popular iGel. nUView TG gels
offer improved resolution, broader mass range,
decreased run time and increased stability.
Separations using nUView TG gels can be carried out
under native or denaturing conditions using a broad
array of popular buffer systems. In this study we
compare the resolution and migration patterns of
nUView TG gels with the Bio-Rad Mini-PROTEAN®
TGX TM and Invitrogen NuPAGE precast gels.
Comparison of migration patterns
nUView TG gels come in a variety of well
configurations and acrylamide formulations (Figure 1).
One of the most popular formulations is the 4-20%
because it offers a broad molecular weight range and
separation characteristics suitable for a wide range of
proteins. We first compared the migration patterns
between the nUView Tris-Glycine, Bio-Rad Mini-
PROTEAN TGX and Invitrogen NuPage precast gels
using the Invitrogen Mark 12 protein standards (Figure
2). Excellent separation performance is achieved with
the nUView TG gels when compared to the TGX and
NuPAGE gels. At the lower MW region (4-2kDA)
nUView TG gels achieve higher resolution than both
the TGX and NuPAGE gels with both insulin chains
completely resolved. Furthermore separation with
nUView TG is achieved across the entire length of the
gel providing a broader MW range from 200kDA to
2kDa.
Figure 2. Migration pattern of protein standard between
nUView TG, Bio-Rad TGX and Invitrogen NuPAGE gels.
MW markers were: 200, 116, 97, 66, 55, 33, 31, 21, 14, 6, 3.5, 2.5
kDa. From left to right, nUView TG 4-20%, Bio-Rad TGX 4-20%,
Invitrogen NuPAGE 4-12%. Seperation over a broader MW range
is achieved with nUView TG gels.
Comparison of resolution
Gel selection is dependent not only on MW range but on
achievable resolution for the sample of interest. To evaluate
the resolution of nUView TG gels a variety of protein
samples were subjected to SDS page (Figure 4).
Figure 4. Resolution for complex protein samples on the
nuView TG gel. Lanes 1-6) Invitrogen Mark 12 protein MW
markers. 2-3) Gallus Gallus Egg white. 4-5) Pisum sativum (English
pea) protein extract. 7-8) Gallus Gallus Egg white dilute. 9-10)
Escherichia coli lysate. 11-12) Human plasma.
Resolution was then compared between the nUView TG,
Bio-Rad TGX, and Invitrogen NuPAGE using the SDS page
analysis of Pisum sativum extract. Comparison of the
banding pattern indicated an increased resolution for the
nUView TG gels, evident by the increase in protein band
separation and number (Figure 5).
Figure 5. Comparison of resolution between nUView TG,
Bio-Rad TGX and Invitrogen NuPAGE. Pisum sativum (English
pea) protein extract was separated and the number of protein
bands and spacing between bands was compared. Left) full gel
lane comparison. Right) The region between 55 and 31 kDa
indicating increased banding and higher resolution for the
nUView TG gel.

Simple 1-step Protocol
Remove the gel from the cassette and place
gel into container.
Cover gel with 25 ml QC stain and leave for
minimum 15 minutes or until all weak
protein bands are fully developed.
Store the gel in the QC stain overnight.
Then, either leave in QC stain or transfer to
DI water for gel storage. (N.B. A minimum 1
hour full stain is recommended before
storing the gel in water.)
Lysed bacterial cell pellet in PBS, 1% Triton X-100 and protease inhibitors (1:500). Samples sonicated at 20% amplitude for
7x20 sec on/off on ice. SDS-PAGE samples diluted 1:3 in PBS and loaded on to a 12% Nusep Tris-Gly gel supplied by
Generon and stained with our QC Stain. The overexpressed 50 kDa protein is visible.
GeBAflex-tube for small scale and rapid dialysis
Ability to dialyse any volume of sample between 10 µl and 20 ml
Available as GeBaflex-tube Mini for sample volumes between 10-250 μl, GeBaflex-tube Midi for
sample volumes between 50-800 μl, GeBaflex-tube Maxi for sample volumes between 100-3000 μl
and GeBaflex-tube Mega for sample volumes between 3 ml and 20 ml.
Simple design means that rapid dialysis is user-friendly with fewer steps and no mess. No syringes or
needles are required. Safe retrieval of sample.
Effective rapid dialysis
Complete and reproducible dialysis within 2 hours.
No air bubbles form between the sample and the dialysis membrane.
NaCl (% control)
The GeBAflex-tube system is a single-step
dialysis tube, free of RNase, DNase, and
available in 4 different volumes and
different MWCOs. Dialysis of a large
number of proteins can be achieved
simultaneously thereby avoiding
bottlenecks in applications as diverse as
de-salting and buffer exchange of protein
and peptide solutions to sample
conditioning in drug screening studies.
Dependence of [NaCl] in the GeBAflex-tube as a function of Time
Time (mins)
Multiple dialysis simultaneously
Multiple samples can be dialyses in parallel. Ideal for screening methodologies or drug candidates.
No pre-treatment necessary
Ready-to-use with no labour intensive preparation steps.
High Recovery
Easy sample loading and recovery and absence of hold up volume ensures high recoveries of dialysed
samples. The use of premium cellulose ester dialysis membrane ensures very low non-specific protein
binding. The membranes are free of heavy metals and sulphides.
GENERON Detergents for Protein Purifications, Reconstitutions and Crystallizations
GENERON has teamed up with Glycon, who have developed new procedures for the preparation of ultra pure
alkyl-β-glycoside-detergents. All α-isomers are completely removed by chromatography. All of our detergents are
crystallized to greater than 99.5% purity. The re-crystallization procedure removes all side products and
lipid alcohols eg dodecanol and the final detergents are presented in a lyophilized form. There is no α−isomer
present in the Glycon detergents.
The anomeric purity of the detergent is critical for the isolation and crystallization of membrane proteins
GENERON Detergents for Protein Purifications, Reconstitutions and Crystallizations
DETERGENT ANALYSES
All of our detergents are crystallised to be greater than 99.5% pure as measured by TLC, HPLC,
double melting point and by the following analytical procedures.
Measurement of purity (HPLC, TLC)
Each lot is first analysed by TLC and HPLC. All detergents are greater than 99.5% pure. TLC was
performed on silica gel (Merck 60 F (254), dichloromethane/methanol 7:1) and detection was effected
by spraying the TLC plate with a solution of ethanol-sulfuric acid (9:1) followed by heating. HPLC was
performed on a standard C18 column with an eluant of methanol/water. The ration of methanol to
water may vary from 65/35 to 85/15 depending on the hydrophobicity of the detergent. A 50% solution
of detergent in MeOH is injected into the HPLC system. [P. Fromme, H. T. Witt, Improved isolation and
crystallisation of Photosystem I for structural analysis; BBA, 1365 (1998) 175-184].
Alcohol and α-isomer contamination
β-n-Alkylglycoside detergents are prepared by glycosylation of the hydrophobic alcohol with activated
glucose or maltose acetates. Nearly all products are purified chromatographically and by
crystallisation. Alcohols and α-isomers are completely removed during the manufacturing process.
Trace amounts of this alcohol in the detergent lot can cause cloudiness in the detergent solution. The
β-anomeric purity of the detergent is advantageous for the crystallization of membrane proteins.
Measurement of purity (Double melting point)
The double melting properties of β-n-alkylglycosides have long been known (E. Fischer, B. Helferich;
Liebigs Ann. Chem. 383, 68 (1911). Alkylglycosides have liquid crystal properties and they can form
mesophases in the melt (a state of order between crystals and liquids). In the smectic A mesophase
the amphiphilic rod-like molecules form layered structures - a microscopic separation by a sequence
of hydrophilic and lipophilic layers. In the isotropic phase, the molecules are packed as a liquid in
a random way. An optical polarised microscope equipped with a hotstage and a central processor
is used to identify transition temperatures from the smectic A to the isotropic phase. Measurement of
transition temperatures (double melting temperatures) gives a very sensitive test for the contamination
of the crystalline detergent with hydrophobic alcohols, α-glycosides, solvents and others organic/
inorganic impurities.
Absorbance
Alkyl-glucosides and -maltosides have low absorbance throughout the UV region. MEGA detergents,
containing amido group, have high absorbance at 230 nm. The absorbance of the 1% w/v detergent
solution in water is measured in the UV region.
Fluorescence
A very low level of aromatic impurity can still result in a large fluorescence background. The
fluorescence of the detergent solution is compared to a standard 0.1% BSA solution. The excitation
wavelength is 280 nm and the emission is measured at 345 nm.
Conductance
Measurement of conductance gives a very sensitive test for the presence of ionic impurities. A solution
of non-ionic detergent should have conductance nearly the same as de-ionised water.
Measurement of pH
The pH of the detergent solution at appropriate concentration is measured. The pH value of the
solution should be neutral.
Solubility in water
Some of the impurities (e.g. hydrophobic alcohol) in detergent preparations are not soluble in water.
Therefore cloudiness of a detergent solution at a concentration where it is known to be soluble
indicates the presence of an insoluble impurity.
L-Selenomethionine (SeMet) and L-Selenocystine (SeCys) are
used to label proteins during cell culture and expression.
Incorporation of these amino acid analogues into target
proteins aids structure determination by X-ray
crystallographic methods, using the well-established
anomalous scattering approach (1, 2).
 Generate myeloma fusion prep.
Initial assay by ELISA of up to 960 colonies.
 Typically identify 20-30 'candidate' binders.
 Antibody screening methods including ELISA,
Western Blot or FACS. Select isotype-specific
antibodies.
3. Timescale: Complete in 3-5 weeks
Subclones with high specificity expanded and
cryopreserved.
Shipment of hybridoma cells and supernatant
fraction from each subcloned cell line.
Production of 5mg of pure mAb from the best
subclone.
4. Timescale: Complete in 3-6 weeks
 Milligramme to gramme quantities produced.
Standard 1 litre batch culture and purification
typically yields 25-40mg
Generon offers custom polyclonal
antibody services to run alongside its
monoclonal antibody production.
The polyclonal service is available in
rabbit, chicken and goat host species.
How to select the correct Host Species?
Quality Custom Polyclonal
Antibody Production
Rabbit Goat Chicken
Most common species Ideal for large volumes Non-mammalian
Less background in crude Long life span Non-invasive (egg yolks)
serum than goat
Good for peptide immunogens Titres tend to be lower in goats IgY purification available.
than rabbits. Not ideal for
modified peptides.
Not ideal for highly conserved Ideal for highly conserved antigens Often 5 to 10 times more antibody
mammalian antigens than rabbits
