Annual Report
July 1, 2008 – June 30, 2009
Michael Kesinger, Chief
Bureau of Citrus Budwood Registration
The Bureau of Citrus Budwood Registration oversees budwood
certification in Florida’s commercial citrus nurseries. The protection of
Florida’s propagating material is fundamental to ensuring growers
receive disease-free trees to plant. Citrus growers have enough
challenges without having to worry if the nursery stock they are planting
is healthy. Planting pathogen-infected trees is economically
unsustainable and, at the very least, farm profits would be greatly
diminished. Disease-infected trees often are not noticed until after
growers have absorbed several years of caretaking expenses on top of
tree and planting costs. Budwood registration includes grafttransmissible
pathogen testing of all propagating sources in Florida.
Allowing only disease-tested material into the nurseries increases the
likelihood of disease-free plants leaving the nursery. The Bureau of
Citrus Budwood Registration’s foundation facility provides nurserymen
with tested scion material for establishment of their own clean stock. The
bureau’s laboratory tests foundation and nursery propagating source
routinely for the endemic graft-transmissible diseases of citrus.
This year’s high point was the completion of the Chiefland budwood
foundation facility. The rapid start-up of this project was a result of
having clean, protected trees available to move from a central Florida
greenhouse to the new greenhouse - without having to start over with
newly budded trees. The fact that those pathogen-tested trees had been
under protected greenhouse cover their entire lives, accelerated the
transition. Therefore, budwood was made available almost from day one.
The Chiefland greenhouses are in their second year of operation, and are
starting to produce significant quantities of budwood. One hundred
thirty-two thousand budeyes were cut from the Chiefland greenhouses
this fiscal year, bringing the total budwood cut in the first 1½ years to
174,210 eyes.
Nine hundred seventy-one tree spaces are planted at Chiefland, leaving
room for only 164 additional trees. The planted trees represent 321
different clonal selections. The Chiefland foundation planting is 33.9%
sweet orange, representing 52 different sweet orange varieties. Mandarins
make up 27.0% of the trees, representing 73 varieties. Grapefruit is
represented with 22 varieties and consists of 9.3% of the planting. The
greenhouse trees are inspected every day. Also, the perimeter of the
structure is walked daily, being closely scanned for any possible areas
that could allow insect incursion.
The lowest temperature recorded outside the greenhouse during the
winter of 2009 was 17 degrees. Inside the greenhouses the temperature
never dipped below 50 degrees. Approximately 2,800 gallons of LP gas
1959 1 st foundation grove started at I-4 and 27
were used during the winter to maintain greenhouse temperatures, and at no
time were more than 50% of the heaters operating.
A new office/headhouse building was added to the bureau’s Chiefland
foundation facility in 2008-2009. The new office includes a walk-in cooler
for the most favorable storage of budwood before shipment. A large work-
room allows for trimming budwood in an air-conditioned environment,
further optimizing the condition of the budwood. A conference room
provides a place for small groups to meet and receive information on the
budwood facility without having to enter the greenhouses. Because of the
importance of minimizing any risk of pathogen contamination of the
foundation clean stock, the greenhouse areas are off limits to visitors.
Although a window has been placed in the side of the greenhouse for visitors
to view the operation.
The Immokalee IFAS cooperative foundation greenhouse facility, which has
not been used for budwood since the 2006-07 fiscal year, has been partially
top-worked to seed source varieties. Eleven different seed source varieties
have been top-worked inside of greenhouse #2.
Since the advent of Florida’s budwood program, more than 163 million
registered nursery trees have been produced. Nursery capacity and
production has been increasing the past three years, after a four year period of
decline. Production levels have nearly returned to what was produced in the
2003-2004 season. Although the number of citrus nurseries is lower than we
had five years ago, the average production per nursery has increased. The
average nursery made 85 thousand propagations this fiscal year. Forty-six
percent of Florida’s nursery trees are produced in Polk County, and this is
where the majority of the citrus nursery inspectors are stationed. Forty-four
commercial citrus nurseries reported making nearly 3.8 million propagations
in 15 counties.
The bureau registered 8,665 scion trees this fiscal year. This is up 594 trees
from last year. This represents 5,515 trees (64%) that are budwood sources
and 3,150 trees (36%) that are seed source trees. Budwood sources are
required to be grown in protected greenhouses that have to be approved by
the Division of Plant Industry, while seed source trees can be field grown.
Bureau personnel witnessed the planting of 933 new scion trees this fiscal
year, of which 415 are seed source. The average age of a budwood scion tree
is 3.2 years old. The transformation of the nursery industry from field to
greenhouse in 2006 resulted in newly established protected budwood trees.
While initially reducing the availability of budwood, the longer term result is
cleaner pathogen-free propagation material. Scion trees produced 51% of the
nursery propagating material, with
the average scion tree producing 878
budeyes in 2008-2009. Increase
trees accounted for 45% of nursery
propagation material. Qualifying
increase trees, those originating
from foundation trees, can be
converted to scion trees by being
witnessed and pathogen tested.
Annual Report 2009 (FY 2008-2009) Bureau of Citrus Budwood Registration 2

This fiscal year, all program seed source and budwood scion trees were tested
for citrus greening with no greening-positive trees being found. Seed source
trees were all additionally tested for citrus leaf blotch virus, with only 2 trees
testing positive. Budwood scion trees were also tested for citrus tristeza virus
and citrus viroids. Collecting the samples from all 8,665 scion trees takes over
3 months. The chopping of the samples is done immediately upon arrival in the
lab. Testing begins with all trees being run for citrus greening, before any other
tests are conducted. Citrus tristeza virus testing begins once all the HLB tests
have been completed. Additional pathogen testing then begins, once the tristeza
tests are completed. All these tests are qPCR based, and the initial extraction is
suitable for all subsequent tests. This is a very efficient and cost- effective
means of testing source trees, as only
one collection per year is required.
In past years, multiple trips to nursery
sites were required for sample
collection. One trip is all that is now
needed, due to the ability to detect all
pathogens from the one extraction. A
total of 33,118 pathogen tests were run
at the bureau’s Winter Haven laboratory. The majority of these analyses were
on scion trees, and foundation trees. The Florida Citrus Arboretum had 2
additional HLB positive trees this year, bringing the total of positives at this
location to 4. No program budwood source trees were identified as being
greening infected. The old foundation grove at Dundee continues to struggle
with citrus canker infection. Approximately 644 grapefruit trees were removed
from the grove to reduce the risk of spreading canker inoculum to the sweet
oranges. The original seed source block containing many Poncirus trifoliate
hybrids, has been particularly hard hit by canker infection, with 65 percent of
the trees being removed.
Hamlins and Valencias, the perennial favorite varieties produced, have
consistently been the top two citrus varieties grown. Midsweet continues in
third place, and is the top midseason variety. With Florida’s citrus industry
predominately producing an orange juice product, it is no surprise that these
three sweet varieties are the most widely planted. Sweet oranges comprise 89
percent of the total nursery production. Grapefruit represent 4.7 % of nursery
propagations, and mandarin 4.1%. Ray Ruby was the most widely used
grapefruit variety, and is also the #1 variety being topworked. Ruby Red is
the second top grapefruit selection propagated, followed by Rio Red.
One hundred fifty-three different
selections were propagated this fiscal
year. Although the citrus industry
utilizes only a minority of varieties in
commercial plantings, the dooryard
(homeowner) market employs a vast
array of citrus types. Swingle
citrumelo remains the top used
rootstock, a position it has held now for 21 years. Nursery trees on Swingle
account for 40 % of all propagations. Kuharske citrange and Carrizo citrange
were the 2 nd and 3 rd most utilized rootstocks, accounting for 24% and 18% of
Historical look at the numbers of tests run for routine citrus pathogen indexing. A larger number of indicator plants were used as the number of indicators
per test varied from 3 to 10.
1 Due to citrus canker concerns and the proximity of clean stock to testing areas, testing of outside sources of budwood was suspended. This reflects in
lower numbers of tests performed 2004-2007. Testing will resume once clean stock is moved out of Winter Haven.
2 HLB extractions only processed by the bureau lab in 2006-2007, PCR test run by Mike Irey.
3 The Budwood Bureau began running the complete test in the 2 nd quarter 2007-2008.
E = ELISA; LB = Citrus Leaf Blotch virus; x = Cachexia Viroid.
CTV testing of budwood sources transitioned from Elisa based to qPCR in 2009.

Pathogen Testing
Testing for Citrus greening (HLB) and Citrus tristeza virus (CTV) are done annually as both of these diseases
have insect vectors. This test ensures that if any insects were able to breach the greenhouse approved structures
and transmit either of these diseases it would be detected. There is one annual collection of samples and it starts
in late August and continues until all collections are complete with December as the goal for completion. As
HLB is our most important pathogen, this time of year for collection was chosen as it is the optimal time of year
to scout for HLB and also for its detection. As we do not have a dedicated collection crew, Bureau laboratory,
grove and inspection personnel from the Winter Haven office collect the majority of the samples. Staff at the
Chiefland Foundation help with the collections from the northern nurseries in their vicinity. This fiscal year our
laboratory extracted 10,265 samples. Utilizing these extractions 7,128 scion trees and 926 Chiefland trees were
tested for HLB using a real time PCR (qPCR test). From the same extract used for the HLB test, CTV qPCR
tests were run also. CTV qPCR tests have replaced the ELISA tests that required a separate spring collection.
These qPCR tests are more sensitive than the MCA-13 ELISA test, use the same principle and can distinguish
MCA-13 severe strains from mild strains of CTV.
Citrus leaf blotch virus (CLBV) is an important virus as we have found it in some of the source trees and it can
be seed transmitted. We developed a qPCR test that we used this past year to test all seed source trees as well as
scion and foundation trees. A low level of positive trees were detected and removed. Real-time PCR testing for
three diseases using the same extract from one collection has allowed sensitive detection and maximized the
time and effort used for the collections from over 10,000 trees.
Shoot-Tip Grafting and Florida Parent Tree Introduction
The application of the efficient qPCR testing technology to incoming parent candidate trees and their shoot-tip
grafts (STGs) has helped to decrease the length of time for testing. Tests on incoming parent candidates give us
an early indication of what disease(s) need(s) to be eliminated. Because of the potential of Citrus greening in
incoming parent tree introductions, they are automatically shoot-tip grafted to remove this risk. QPCR tests on
STGs allow early determination of disease status while slower biological tests are in progress. After initial
qPCR testing results are obtained, a horticulturally superior specimen is selected, grown off and increased for
eventual release.
We recognize the importance of having all possible Florida Germplasm selections as disease free as possible
and kept safe in our new screen house facility at Chiefland. This fiscal year an emphasis was placed on the
Parent Tree Indexing and Shoot-tip grafting programs to expedite progress on all selections and also to work to
remove mild isolates of CTV from clones at Chiefland where necessary.
Sixty-two trees were in the Parent Tree Indexing Program at year’s end. Twenty-five were entered this year and
32 parents and/or STGs were released this year. Of the released clones, five were Breeder’s varieties, eight were
already available at Chiefland, but had the Mild CTV removed, nine were STGed to remove severe CTV, three
viroids, and one Citrus leaf blotch virus.
In 2008-2009, we had our greatest production of shoot-tip grafts, doubling that of the previous year. 1,846
shoot-tip grafts were set up, of these, 135 were grafted into rootstocks in the greenhouse. At the end of the fiscal
year a total of 43 cultivars were in the process of being shoot-tip grafted. Fifty-six cultivars have been
successfully STGed and are undergoing various stages of testing. The process of STGing a selection used to
take a minimum of five years. Now for selections newly entered into the program, the entire process takes under
two years if there are no complications.
Annual Report 2009 (FY 2008-2009) Bureau of Citrus Budwood Registration 34

Our Vision
The Bureau of Citrus Budwood Registration will provide high yielding, pathogen free, quality budlines that will
positively impact the productivity and prosperity of our citrus industry.
Our Mission
The Bureau of Citrus Budwood Registration administers a program to assist growers and nurserymen in
producing citrus nursery trees that are believed to be horticulturally true to varietal type, productive, and free
from certain recognizable bud-transmissible diseases detrimental to fruit production and tree longevity.
Benefits of a Budwood Program
The Florida budwood program has greatly impacted Florida citrus, as the use of registered budwood selections
or “budlines” is responsible for impressive increases in yield and fruit quality in the industry. The budwood
program introduced many nucellar lines that are still the basis of many of our main varieties. The ability to test
for graft-transmitted pathogens and exclude unclean propagating material has not only increased production and
tree longevity but also the ability of our groves to withstand other environmental stresses.
• Diseases are kept out of the propagation system
o Using clean nursery stock means growers receive disease free trees coming out of the nursery
• Testing for citrus viroids is more critical than ever as the industry uses predominantly citrange
and citrummelo type rootstocks that are more susceptible to viroids
o The budwood program is credited with having viroid free trees for the industry to use when the rootstocks
were changed from rough lemon and sour orange to Carrizo and Swingle
Other countries without budwood programs had major viroid problems when changing rootstocks
away from sour orange
• Citrus tristeza virus strains are worse today than a few years ago
o Stem pitting tristeza virus strains have been found in several commercial grove locations in Florida
• Psorosis virus is becoming hard to find in Florida after 50 years of budwood registration
o Prior to the budwood program psorosis was widespread in Florida
Yields in psorosis infected trees were reduced 10 to 35 percent and made the trees easier to
succumb to cold and other environmental stresses
• Protection of our industry from exotic citrus diseases requires controlling citrus propagating
material
o Citrus greening is the latest disease to move from the exotic disease list to become an endemic disease
o Citrus variegated chlorosis and citrus sudden death are examples of pathogens whose main means of
control is budwood certification
Grove trees started from clean stock have the potential to be profitable even in the presence of
disease, whereas nursery trees started systemically infected will never become profitable grove
trees
• Yields are increased with registered budwood clonal selections
o Nucellar Valencia clones alone increased yields by 25% over old line selections
o Examples of registered clonal selections in every variety show remarkable yield gains
• True to type varieties and rootstocks are propagated
o Inspected trees with off-types are removed from budwood supply
• Budwood programs are recognized world wide as the most effective means of excluding and
containing citrus graft-transmissible pathogens
