Abstract: Astaxanthin is a natural color carotenoid found in salm<strong>on</strong>, salm<strong>on</strong><br />

eggs, krill, and crab. Therefore, <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> has been c<strong>on</strong>tained in the<br />

<strong>human</strong> diet for a l<strong>on</strong>g time. Astaxanthin <str<strong>on</strong>g>from</str<strong>on</strong>g> krill has been used for<br />

cosmetics to suppress post-UVB hyperpigmentati<strong>on</strong> in <strong>human</strong> <strong>skin</strong> and food<br />

color additives. Recently, <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> <str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g> is<br />

available using new fermentati<strong>on</strong> technology <str<strong>on</strong>g>of</str<strong>on</strong>g> H. <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g> and it is used for<br />

dietary supplements, food color additives and cosmetics.<br />

<str<strong>on</strong>g>Effects</str<strong>on</strong>g> <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> <str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g> <strong>on</strong> <strong>human</strong> subjects<br />

were tested. No serious adverse effects were observed by patch testing and<br />

sequencing applied test <strong>on</strong> <strong>human</strong> <strong>skin</strong>. In a pilot study, the <strong>skin</strong> repeated<br />

applicati<strong>on</strong> test <str<strong>on</strong>g>of</str<strong>on</strong>g> cream c<strong>on</strong>taining <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> <strong>on</strong> <strong>human</strong> <strong>skin</strong> showed the<br />

visual wrinkle reducti<strong>on</strong>. The present paper described about patch testing,<br />

<strong>skin</strong> repeated applicati<strong>on</strong> test, and a pilot study evaluating the wrinkle<br />

reducti<strong>on</strong> effect <strong>on</strong> <strong>human</strong> <strong>skin</strong>.<br />

1. Introducti<strong>on</strong><br />

Astaxanthin bel<strong>on</strong>gs to the carotenoid family that includes β-carotene etc.<br />

It is a red color widely and naturally distributed in foods such as fishes e.g.<br />

salm<strong>on</strong>, sea bream etc. and Crustaceans such as shrimps, crabs etc. which<br />

have been well c<strong>on</strong>sumed. In the past, <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> <str<strong>on</strong>g>from</str<strong>on</strong>g> krill was <strong>on</strong>ly put<br />

into practical use. However, <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> <str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g>, a<br />

green algae, is able to accumulate sufficient quantities and ec<strong>on</strong>omically<br />

after establishment <str<strong>on</strong>g>of</str<strong>on</strong>g> its industrial cultivating technology and the compound<br />

expects to increase its number <str<strong>on</strong>g>of</str<strong>on</strong>g> potential applicati<strong>on</strong>s.<br />

Astaxanthin <str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g> has been listed in the Standards<br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> Cosmetic Combinati<strong>on</strong> Ingredients by Category 2) and both are put into<br />

practical use.<br />

Recently, str<strong>on</strong>g anti-oxidative acti<strong>on</strong> 3)4) <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> was reported and<br />

diverse range <str<strong>on</strong>g>of</str<strong>on</strong>g> its physiological functi<strong>on</strong>s and bioactivities have been<br />

studied intensely. For instance, many papers <strong>on</strong> its acti<strong>on</strong>s, such as<br />

anti-inflammatory acti<strong>on</strong> 5) , anti-arteriosclerotic acti<strong>on</strong> 6)7) , acti<strong>on</strong> against<br />

diabetes 8)9) , daily rhythm regulating acti<strong>on</strong> 10) , immuno-activating acti<strong>on</strong> 11) ,<br />

anti-stress acti<strong>on</strong> 12) and inhibiti<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> urinary bladder carcinogenesis 13) etc.,<br />

have been reported. On the other hand, its acti<strong>on</strong>s <strong>on</strong> <strong>skin</strong>, such as inhibiti<strong>on</strong><br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> pigmentati<strong>on</strong> 14) , inhibiti<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> melanin-generati<strong>on</strong> and <str<strong>on</strong>g>of</str<strong>on</strong>g> light-induced<br />

aging 15) have been reported.<br />

At this time, we prepared a cream preparati<strong>on</strong> combined with <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g><br />

<str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g>, which may be expected its cosmetic effect for<br />

the <strong>skin</strong> and performed a patch test and a <strong>skin</strong> repeated applicati<strong>on</strong> test in<br />

<strong>human</strong> using the preparati<strong>on</strong> in order to assess safety <strong>on</strong> the <strong>skin</strong>. At the<br />

same time, we examined its effect <strong>on</strong> <strong>skin</strong> moisture retenti<strong>on</strong> as well as <strong>on</strong><br />

<strong>skin</strong> c<strong>on</strong>diti<strong>on</strong>. We will report the results as follows:<br />

2. Patch Test in Human Subjects<br />

We performed a patch test using <strong>human</strong> subjects in order to assess safety<br />

<strong>on</strong> the <strong>skin</strong>, using a preparati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> <str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g><br />

<str<strong>on</strong>g>pluvialis</str<strong>on</strong>g> and a cream preparati<strong>on</strong> combined with <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> <str<strong>on</strong>g>from</str<strong>on</strong>g><br />

<str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g> .<br />

2-1.Test materials<br />

Sample No.1: Astaxanthin <str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g><br />

AstaReal Oil 50 F R was used as Sample No.1: Dried powders, obtained<br />

<str<strong>on</strong>g>from</str<strong>on</strong>g> the disrupted cell-walls <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g> supplied by<br />

Microgaia Inc. (Maui Island <str<strong>on</strong>g>of</str<strong>on</strong>g> Hawaii State, USA) were extracted with<br />

acet<strong>on</strong>e, and then tri (capryl / capric acid) glycerol was added to the<br />

extracts to make 5% (as free <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g>) <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> preparati<strong>on</strong>.<br />

The <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> <str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g> <str<strong>on</strong>g>of</str<strong>on</strong>g> Sample No.1 and the<br />

cream base <str<strong>on</strong>g>of</str<strong>on</strong>g> Sample No. 2 were combined to form 0.7 mg/g <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

<str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> cream.<br />

2-2. Subjects<br />

The study was performed using 45 subjects <str<strong>on</strong>g>of</str<strong>on</strong>g> healthy adults who<br />

c<strong>on</strong>sented to be enrolled in the study. Table 1 shows an age structure.<br />

2-3. Method<br />

Samples were applied <strong>on</strong> the medial site <str<strong>on</strong>g>of</str<strong>on</strong>g> the upper arm subjects and a<br />

plaster (Fin-Chamber, Taisho Pharmaceutical Co., Ltd.) was placed over the<br />

treated area. The plaster was removed after 24 hours and then the<br />

<strong>skin</strong>-irritability observed after 30 minutes (24 hours) and also a further 24<br />

hours after the removal (total 48 hours).<br />

2-4. Judgement<br />

Judgment was c<strong>on</strong>ducted in accordance with “Standard <str<strong>on</strong>g>of</str<strong>on</strong>g> Japanese Patch<br />

Testing Research Group” and evaluati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> each Sample was made by<br />

calculati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>skin</strong>-irritability index in accordance with Evaluati<strong>on</strong> Criteria<br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> Skin-Irritability Index 16) .<br />

2-5. Results<br />

1) Astaxanthin <str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g> (Sample 1): 42 subjects<br />

showed no reacti<strong>on</strong> (-) to the applied AstaReal® oil after 24 and 48 hours.<br />

Of the three subjects <strong>on</strong>ly 2 (Male 24 yrs. and Male 25 yrs.) showed slight<br />

reacti<strong>on</strong>s (±) after 24 hours, and <strong>on</strong>e subject (Female 25 yrs.) reacted (±)<br />

<strong>on</strong>ly after 48 hours. The male subjects (Male 24 yrs., Male 25 yrs.)<br />

experienced dermatitis that was induced by the adhesive plaster in<br />

c<strong>on</strong>tact with the <strong>skin</strong> and suspecting its expansi<strong>on</strong> to the tested sites.<br />

Overall Skin-Irritability Index showed 3.3.<br />

2) Cream base (Sample No.2): 42 subjects experienced no <strong>skin</strong> reacti<strong>on</strong>s to<br />

the cream base c<strong>on</strong>trol. Only 2 (Male 24 yrs., Male 25 yrs.) <str<strong>on</strong>g>of</str<strong>on</strong>g> 45 subjects<br />

showed slight signs <str<strong>on</strong>g>of</str<strong>on</strong>g> irritati<strong>on</strong> (±) <strong>on</strong>ly after 24 hours, and <strong>on</strong>e subject<br />

(Female 25 yrs.) showed ± <strong>on</strong>ly after 48 hours. Of the two male subjects<br />

(Male 24 yrs., Male 25 yrs.), dermatitis was again induced by adhesive<br />

plaster, suspecting its expansi<strong>on</strong> to the tested sites. Skin-Irritability<br />

Overall Skin Irritability Index showed 3.3.<br />

3) Cream base + <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> (Sample No.3): 43 <str<strong>on</strong>g>of</str<strong>on</strong>g> 45 subjects showed no<br />

reacti<strong>on</strong> (-) to the <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> c<strong>on</strong>taining cream. Only 2 (Male 24 yrs.,<br />

Male 25 yrs.) <str<strong>on</strong>g>of</str<strong>on</strong>g> 45 subjects showed signs <str<strong>on</strong>g>of</str<strong>on</strong>g> irritati<strong>on</strong> (±) both after 24<br />

and 48 hours. The both had remarkable dermatitis induced by adhesive<br />

plaster and suspected expansi<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> the dermatitis to the tested sites.<br />

Overall Skin-Irritability Index showed 2.2.<br />

2-6. C<strong>on</strong>clusi<strong>on</strong><br />

All the three Samples were judged safe and c<strong>on</strong>cluded no problem in<br />

safety c<strong>on</strong>cerning primary irritant inducti<strong>on</strong>.<br />

3. Skin Repeated Applicati<strong>on</strong> Test in Human Subjects<br />

Since the safety for <strong>skin</strong>-irritability was assessed in Patch Test using the<br />

cream preparati<strong>on</strong> combined with <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> <str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g>,<br />

we performed a <strong>skin</strong> repeated applicati<strong>on</strong> test using the cream preparati<strong>on</strong> in<br />

order to assess safety as well as to evaluate its effects <strong>on</strong> <strong>human</strong> <strong>skin</strong>.<br />

3-1. Test material<br />

The cream preparati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> Sample No.3 (see paragraph 2-1) was used.<br />

3-2. Subjects<br />

The study was performed using 11 subjects <str<strong>on</strong>g>of</str<strong>on</strong>g> healthy female adults who<br />

c<strong>on</strong>sented to be enrolled in the study. Table 2 shows an age structure.<br />

3-3. Method and study period<br />

After washing face and applying cosmetic loti<strong>on</strong> <strong>on</strong> the face every morning<br />

and evening, the subjects applying a small bean sized volume (approx. 0.2 g)<br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> sample <strong>on</strong> their face and spread it thinly <str<strong>on</strong>g>from</str<strong>on</strong>g> under the eyes to the cheeks.<br />

They were permitted to use other cosmetics likely as usual. Study period was<br />

3 weeks-with daily applicati<strong>on</strong>s.<br />

3-4 Test Parameters<br />

Visual examinati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> the following measurements were performed before<br />

and after the 3-week applicati<strong>on</strong> respectively. They were performed in a room<br />

kept approx. 21℃- R.T. and 55 % - R.H., after 15 minutes <str<strong>on</strong>g>of</str<strong>on</strong>g> face washing.<br />

3-4-1. Visual Inspecti<strong>on</strong>s for Safety.<br />

Irritability to <strong>skin</strong> and safety were evaluated observing subjects’ face-<br />

<strong>skin</strong> c<strong>on</strong>diti<strong>on</strong> before and after the applicati<strong>on</strong>.<br />

3-4-2. Skin moisture c<strong>on</strong>tent<br />

Skin moisture c<strong>on</strong>tent <str<strong>on</strong>g>of</str<strong>on</strong>g> the outer site, approx. 1cm <str<strong>on</strong>g>from</str<strong>on</strong>g> the left outer<br />

canthus was measured using Moisture Checker MY707S, Scalla Inc.<br />

3-4-3. Questi<strong>on</strong>naires<br />

Subjective symptoms <strong>on</strong> <strong>skin</strong> c<strong>on</strong>diti<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> enrolled subjects in this study<br />

were checked by questi<strong>on</strong>naires, and safety and effectiveness <str<strong>on</strong>g>of</str<strong>on</strong>g> the cream<br />

preparati<strong>on</strong> were evaluated.<br />

3-5. Results<br />

3-5-1. Safety assessment<br />

There were no subjects that showed deteriorati<strong>on</strong> in <strong>skin</strong> c<strong>on</strong>diti<strong>on</strong> caused<br />

by the test samples.<br />

3-5-2. Clinical efficacy<br />

8 subjects out <str<strong>on</strong>g>of</str<strong>on</strong>g> 11 were observed improvement <str<strong>on</strong>g>of</str<strong>on</strong>g> subjective symptoms in<br />

<strong>skin</strong>-dryness. One subject with senile pigment macule and 2 subjects with<br />

chloasma (sun-burn spots) were observed light improvement in <strong>skin</strong>-colour.<br />

3-5-3. Skin moisture c<strong>on</strong>tent<br />

Fig.1 shows changes <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>skin</strong> moisture c<strong>on</strong>tent <str<strong>on</strong>g>of</str<strong>on</strong>g> the outer site, approx. 15<br />

cm <str<strong>on</strong>g>from</str<strong>on</strong>g> the left outer canthus before and after the applicati<strong>on</strong> respectively. A<br />

difference, + 3.32% between the value before applicati<strong>on</strong> (mean value <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

moisture c<strong>on</strong>tent: 43.059%) and the value after applicati<strong>on</strong> (mean value <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

moisture c<strong>on</strong>tent: 46.381%) showed an elevati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> moisture c<strong>on</strong>tent<br />

Fig.4 shows the improving effects by questi<strong>on</strong>naires after 3 week-repeated<br />

applicati<strong>on</strong>s. Almost <str<strong>on</strong>g>of</str<strong>on</strong>g> all subjects obtained improvement <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>skin</strong> c<strong>on</strong>diti<strong>on</strong><br />

except <strong>on</strong>e subject (Female 29 yrs.) who had a feeling <str<strong>on</strong>g>of</str<strong>on</strong>g> coarse <strong>skin</strong>.<br />

week-applicati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> the cream preparati<strong>on</strong> combined with <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> and<br />

the same result in safety assessment was also obtained in this Skin Repeated<br />

Applicati<strong>on</strong> Test as in the preceded Patch Test. Furthermore, the <strong>skin</strong><br />

moisture retenti<strong>on</strong> increased significantly and improvement <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>skin</strong> c<strong>on</strong>diti<strong>on</strong><br />

was dem<strong>on</strong>strated in this study.<br />

4. Beauty Effect<br />

From the results showing increase <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>skin</strong> moisture retentivity in Skin<br />

Repeated Applicati<strong>on</strong> Test, we have expected a beauty effect <str<strong>on</strong>g>of</str<strong>on</strong>g> the cosmetic<br />

preparati<strong>on</strong> combined with <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g>. Then, we performed a preliminary<br />

study <strong>on</strong> beauty effect <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> using 3 subjects.<br />

4-1. Test material<br />

The cream preparati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> Sample No. 3 (See Paragraph 2-1) was used.<br />

4-2. Subjects<br />

The study was performed using 3 subjects <str<strong>on</strong>g>of</str<strong>on</strong>g> healthy female adults who<br />

c<strong>on</strong>sented to be enrolled in the study. Table 3 indicates an age structure and<br />

<strong>skin</strong>-types.<br />

4-3. Method and study period<br />

Study period: 2 weeks<br />

Method: After washing face, apply a test loti<strong>on</strong> <strong>on</strong> the whole face, and then<br />

paint the test cream <str<strong>on</strong>g>of</str<strong>on</strong>g> Sample No. 3 spreading <str<strong>on</strong>g>from</str<strong>on</strong>g> under the<br />

eyes to the cheeks. Other cosmetics are permitted to use likely as<br />

usual.<br />

Observati<strong>on</strong> was performed after 15 minutes under keeping rest with<br />

4-6. Results<br />

4-6-1. Skin c<strong>on</strong>diti<strong>on</strong> evaluated by subject’s self-assessment<br />

Fig.5 shows <strong>skin</strong> c<strong>on</strong>diti<strong>on</strong> in each item evaluated by subject’s<br />

self-assessment (See Attachment 1).<br />

4-6-2. Inspecti<strong>on</strong> / Palpati<strong>on</strong><br />

Fig.6 shows the results obtained <str<strong>on</strong>g>from</str<strong>on</strong>g> <strong>skin</strong>-inspecti<strong>on</strong> and palpati<strong>on</strong> by<br />

beauty specialists. Moderate improvement <str<strong>on</strong>g>of</str<strong>on</strong>g> “Fine wrinkles” and<br />

“Flabbiness under eyes” which are typically caused by <strong>skin</strong>-dryness was<br />

observed in M2 (Dried <strong>skin</strong>) and M3 (Mixed type). The <strong>skin</strong> c<strong>on</strong>diti<strong>on</strong>s<br />

expressed as “Moistness”, Smoothness” and “Elasticity” obtained <str<strong>on</strong>g>from</str<strong>on</strong>g><br />

<strong>skin</strong>-palpati<strong>on</strong> showed improving tendency in all cases (See Attachment II).<br />

4-6-3. Skin moisture c<strong>on</strong>tent / sebum (<strong>skin</strong> oil) c<strong>on</strong>tent<br />

Fig.7 shows changes <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>skin</strong>-moisture c<strong>on</strong>tent and sebum (<strong>skin</strong> oil) c<strong>on</strong>tent<br />

after 2 week-applicati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> the cosmetic preparati<strong>on</strong> combined with<br />

<str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g>. Elevati<strong>on</strong>s <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>skin</strong> moisture c<strong>on</strong>tent were observed in the<br />

subjects, M2 (Dried <strong>skin</strong>) and M3 (Mixed type). It was c<strong>on</strong>sidered that the<br />

sample had moisturizing effects. Increased sebum c<strong>on</strong>tent was observed in<br />

M1 (Normal <strong>skin</strong>) and M3 (Mixed type), it was c<strong>on</strong>sidered such increase<br />

might be affected by the cream base and not c<strong>on</strong>taining <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> (See<br />

Attachment III).<br />

4-6-4.Wrinkle-coefficient outer canthi / Skin grain-coefficient <str<strong>on</strong>g>of</str<strong>on</strong>g> cheeks<br />

Fig.8 shows changes <str<strong>on</strong>g>of</str<strong>on</strong>g> wrinkle-coefficient and <strong>skin</strong> grain-coefficient after<br />

2 week-applicati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> the cosmetic preparati<strong>on</strong> combined with <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g>.<br />

Wrinkle-coefficient (deep wrinkles), S2 <str<strong>on</strong>g>of</str<strong>on</strong>g> left and right outer canthi, and <strong>skin</strong><br />

grain-coefficient (Grain uniformity), K4 <str<strong>on</strong>g>of</str<strong>on</strong>g> left and right cheeks were<br />

measured. The smaller values indicate the smaller or the less in wrinkles and<br />

the finer in <strong>skin</strong> grain-uniformity. Decreases <str<strong>on</strong>g>of</str<strong>on</strong>g> both wrinkle-coefficient and<br />

<strong>skin</strong> grain-coefficient were observed in M2 (Dried <strong>skin</strong>). In this subject,<br />

wrinkles <str<strong>on</strong>g>of</str<strong>on</strong>g> the outer canthi were improved and <strong>skin</strong> grains <str<strong>on</strong>g>of</str<strong>on</strong>g> the cheeks<br />

were uniform. On the c<strong>on</strong>trary, any changes were not observed in other 2<br />

subjects.<br />

Fig.9 shows magnified photographs <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>skin</strong> in M1 where improvement <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

fine wrinkled <strong>skin</strong> c<strong>on</strong>diti<strong>on</strong> was observed. Furthermore, mitigati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> fine<br />

wrinkles under eyes and outer canthi caused by dryness was observed in all 3<br />

cases.<br />

4-7. C<strong>on</strong>clusi<strong>on</strong><br />

After 2 week-applicati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> the <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> cream-preparati<strong>on</strong>, increased<br />

<strong>skin</strong> moisture retenti<strong>on</strong> was observed and improvements <str<strong>on</strong>g>of</str<strong>on</strong>g> fine wrinkles <strong>on</strong><br />

outer canthi and <strong>skin</strong> grain uniformity <str<strong>on</strong>g>of</str<strong>on</strong>g> cheeks were c<strong>on</strong>firmed.<br />

5. Discussi<strong>on</strong><br />

We studied the effects <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> <str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g> <str<strong>on</strong>g>pluvialis</str<strong>on</strong>g> <strong>on</strong><br />

<strong>human</strong> <strong>skin</strong> and first c<strong>on</strong>firmed its safety for the <strong>skin</strong> in Patch Testing and<br />

Skin Repeated Applicati<strong>on</strong> Test and at the same time, the increase <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>skin</strong><br />

moisture retenti<strong>on</strong> was observed in the latter study using a cream<br />

preparati<strong>on</strong>. Sec<strong>on</strong>dly, we then carried out the preliminary study for beauty<br />

effect using 3 subjects with normal <strong>skin</strong>, dried <strong>skin</strong> or mixed <strong>skin</strong> type<br />

respectively and c<strong>on</strong>firmed the improvement in such parameters as<br />

moistness, smoothness and elasticity <str<strong>on</strong>g>of</str<strong>on</strong>g> the <strong>skin</strong> obtained by <strong>skin</strong>-inspecti<strong>on</strong><br />

and <strong>skin</strong>-palpati<strong>on</strong>. Furthermore we c<strong>on</strong>firmed the increase <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>skin</strong> moisture<br />

c<strong>on</strong>tent (<strong>skin</strong> moisture retenti<strong>on</strong>) and the improving tendency <str<strong>on</strong>g>of</str<strong>on</strong>g> the fine<br />

wrinkles outer canthi and <strong>skin</strong> grain <str<strong>on</strong>g>of</str<strong>on</strong>g> cheeks.<br />

Since it was clarified in these studies that <str<strong>on</strong>g>astaxanthin</str<strong>on</strong>g> <str<strong>on</strong>g>from</str<strong>on</strong>g> <str<strong>on</strong>g>Haematococcus</str<strong>on</strong>g><br />

<str<strong>on</strong>g>pluvialis</str<strong>on</strong>g> is a promising material combined with cosmetics, we will perform<br />

further investigati<strong>on</strong>s in order to rec<strong>on</strong>firm its beauty effect and to elucidate<br />

a mechanism <str<strong>on</strong>g>of</str<strong>on</strong>g> its acti<strong>on</strong>.<br />
