(D and E) Doma<strong>in</strong>s required <strong>in</strong> Wek to <strong>in</strong>teract with ei<strong>the</strong>r <strong>Toll</strong> or MyD88. Flag-tagged <strong>Toll</strong> DLRR (D) or V5-tagged DmMyD88 (E) was cotransfected<br />

with different HA-tagged Wek derivatives. Each <strong>in</strong>dividual doma<strong>in</strong> <strong>of</strong> Wek is sufficient to associate with <strong>the</strong> <strong>in</strong>tracellular doma<strong>in</strong> <strong>of</strong> <strong>Toll</strong>. However,<br />

WekM and WekC, but not WekN, associates with MyD88. Arrowheads: expected size <strong>of</strong> WekN. Open arrowheads: expected size <strong>of</strong> <strong>Toll</strong> DLRR . The<br />

additional <strong>in</strong>tense bands <strong>in</strong> (D) correspond to <strong>the</strong> light cha<strong>in</strong>s and heavy cha<strong>in</strong>s <strong>of</strong> anti-Flag antibodies, respectively.<br />

(F) Doma<strong>in</strong> required <strong>in</strong> MyD88 to <strong>in</strong>teract with Wek. S2 cells were cotransfected with HA-tagged Wek and different V5-tagged MyD88 derivatives.<br />

Upon removal <strong>of</strong> any regions <strong>of</strong> MyD88, <strong>the</strong> <strong>in</strong>teraction between Wek and MyD88 was greatly reduced. Open arrowheads: expected size <strong>of</strong> <strong>the</strong><br />
