Culture serum Q & A Latest news How should I condition my culture to new sera & media? For best results, the process of adapting cells to a novel culturing milieu should be sequential. Direct migration to a new serum or media can be detrimental to cell viability and should be avoided. The most common procedure is to dilute the culture in a low percentage of new media at the time of passage, increasing gradually in proportion upon each subculture (split). What are the benefits of using animal-free media? Animal-free media contain no animal-derived components and have defined components which eliminate any inconsistencies associated with serum batch variations. Hence, it is not necessary to conduct time consuming lot-testing of serum supplements in order to acquire reliable and reproducable results. Media free of animal components are commonly used in the production of human and animal biopharmaceuticals, diagnostic reagents and bioagricultural products. It is also beneficial for cell culture applications aimed at high-throughput production of recombinant proteins, monoclonal antibodies and viral vectors, as reduced protein content improves both purification yield and efficiency. Do I need heat inactivated sera for general cell cultures? The heat inactivation procedure involves warming thawed FBS to between 45°C to 62°C for 15 to 60 minutes, and was once regularly performed to destroy components of the complement cascade and neutralise mycoplasma in the sera. However, recent findings suggest that this practice is often not essential for many cell culture applications and may instead negatively affect growth rates as vital temperature-sensitive nutrients are also destroyed in the heating process. Vigorous sterile filtration of HyClone FBS through three consecutive 0.1µm pore-size rated filters ensures only minuscule level of complement and mycoplasma persists in the serum.
