Evaluation of various ELISAs provided by nal von minden, for the serological diagnosis of infectious agents in human serum Introduction Enzyme-linked Immunosorbent Assay (ELISA) is a standard method for the serological detection of pathogen specific antibodies. The diagnostic sensitivity (proportion of actual positive samples which are correctly identified) and diagnostic specificity (proportion of negative samples which are correctly identified) are important parameters during the validation process of assays intended for routine laboratory diagnostic use. In this study, the efficiency of three different ELISA kits was evaluated by comparing the diagnostic sensitivities and specificities with those of benchmark assays. All tests were performed at the Department of Virology, Institute of Medical Microbiology and Hygiene, University of Regensburg, Germany (RIMMH). Study design and results The study comprised testing of the following ELISA kits and their corresponding parameters; the focus was on the detection of specific antibodies against pathogens which can be relevant during pregnancy: Chlamydia pneumoniae (IgG, IgA), Chlamydia trachomatis (IgG, IgA), CMV (IgG, IgM), EBV EA (IgG), EBV EBNA-1 (IgG), EBV VCA (IgG, IgM), HSV-1/2 (IgG, IgM), Measles (IgG, IgM), Mumps (IgG, IgM), Rubella (IgG, IgM), Toxoplasma gondi (IgG, IgM), Treponema pallidum (IgG, IgM), VZV (IgG, IgM). The study was performed in two steps. First, patients‘ frozen sera were tested by three different immunoassays provided by nal von minden in parallel with the corresponding comparator assays. In a second step, discrepant results (5–15 samples per parameter) obtained by the best performing ELISAs were repeated and tested against the comparator. The table below shows the final results including assay rating compared with the comparator. The tests are easy to use and handle, in accordance with the description provided within the package inserts. Moreover, the packaging of the ELISA kits is suitable for storage and is made from recycled and reusable material. In conclusion, the mostly high agreement of diagnostic sensitivity and specificity for all parameters tested indicates that the diagnostic performance of the provided ELISA kits is comparable to the comparator assays used in this study. page 33
