<strong>Corning</strong> <strong>HYPERStack</strong> vessels can be connected to tubing via tube welding<br />

or by using the pre-assembled, multi-purpose connects (MPCs).<br />

Seeding the <strong>HYPERStack</strong> Vessel<br />

Tube weld serum and media bags, mix thoroughly.<br />

1: Clamp off ~300 mL of media in the top of the bag. Fill syringe with cell inoculum.<br />

Inject into bottom portion of media bag.<br />

Attach inoculated media bag to <strong>HYPERStack</strong> vessel’s 3/8" I.D. liquid handling tubing.<br />

2: Close liquid handling tubing clamp.<br />

3: Close chase tubing clamp.


Filling the <strong>HYPERStack</strong> Vessel<br />

1: Place <strong>HYPERStack</strong> vessel into fill position at a 10° angle and position media bag at<br />

same level as the <strong>HYPERStack</strong> vessel.<br />

2: Open liquid handling tubing clamp.<br />

3: Open media bag tubing clamp. Fluid will enter the <strong>HYPERStack</strong> vessel.<br />

4: Raise media bag to increase flow into the <strong>HYPERStack</strong> vessel, emptying the inoculated<br />

portion of the media bag.<br />

5: Remove clamp on top of media bag; finish filling <strong>HYPERStack</strong> vessel to the fill line.<br />

To reduce rate of flow and prevent overfilling, lower media bag during final fill.<br />

Isolate Liquid into <strong>HYPERStack</strong> Layers<br />

1: Place <strong>HYPERStack</strong> vessel into “isolation” position and lower media bag below<br />

the height of the <strong>HYPERStack</strong> vessel.<br />

2: Hold the chase filter upright and open the chase tube clamp. Allow excess media<br />

to empty back into the media bag.<br />

3: Close the media bag tubing clamp.<br />

4: Open the liquid handling tubing clamp.<br />

5: Return <strong>HYPERStack</strong> vessel to starting position.<br />

6: Close chase tube clamp.<br />

7: Close liquid handling tube clamp.<br />

Place <strong>HYPERStack</strong> vessel into incubator. (Option: Empty media bag can be<br />

removed or remain attached to the <strong>HYPERStack</strong> vessel.)<br />

Keep the vent filter upright to prevent wetting out the filter.


Empty Spent Media from <strong>HYPERStack</strong> Vessel<br />

1: Remove <strong>HYPERStack</strong> vessel from incubator; keep vent tubing upright.<br />

2: Place <strong>HYPERStack</strong> vessel in “fill” position at a 10° angle.<br />

Lower media bag below height of <strong>HYPERStack</strong> vessel.<br />

3: Open the media bag tubing clamp.<br />

4: Open the vessel’s liquid handling tubing clamp. Fluid will enter empty media bag.<br />

5: Place <strong>HYPERStack</strong> vessel in final empty position and let all media drain.<br />

6: Lift tubing to chase remaining fluid into the media bag.<br />

7: Close liquid handling tubing clamp.<br />

8: Close media bag tubing clamp. Remove spent media bag.<br />

Keep the vent filter upright to prevent wetting out the filter.<br />

1: Connect cell dissociation and quench bags together to form harvest bag assembly.<br />

2: Attach to <strong>HYPERStack</strong> vessel.<br />

3: Raise harvest bag assembly above height of <strong>HYPERStack</strong> vessel and place it into<br />

“fill” position at a 10° angle.<br />

4: Open cell dissociation solution tube clamp.<br />

5: Open harvest bag assembly tube clamp. Fluid will enter the <strong>HYPERStack</strong> vessel.<br />

6: Close liquid handling tube clamp.<br />

1: Close cell dissociation tube clamp.<br />

2: Return <strong>HYPERStack</strong> vessel to the starting position; rock the vessel to distribute cell<br />

dissociation solution.<br />

3: After incubating the <strong>HYPERStack</strong> vessel for the required time for cell release, place<br />

<strong>HYPERStack</strong> vessel in “fill” position at a 10° angle.<br />

4: Open quench bag tube clamp.<br />

5: Open liquid handling tube clamp. Quench solution will enter the <strong>HYPERStack</strong> vessel.<br />

6: Close liquid handling tube clamp. Distribute the quench solution to completely<br />

deactivate the cell dissociation solution.<br />

1: Place harvest bag assembly lower than <strong>HYPERStack</strong> vessel.<br />

2: Place the vessel in the empty position.<br />

3: Open liquid handling tube clamp.<br />

4: Transfer cell solution into empty quench bag.<br />

5: Disconnect harvest bag assembly from <strong>HYPERStack</strong> vessel. Cell suspension is<br />

ready for processing.<br />
