Corning HYPERStack vessels can be connected to tubing via tube welding
or by using the pre-assembled, multi-purpose connects (MPCs).
Seeding the HYPERStack Vessel
1 2 3
Tube weld serum and media bags, mix thoroughly.
1: Clamp off ~300 mL of media in the top of the bag. Fill syringe with cell inoculum.
Inject into bottom portion of media bag.
Attach inoculated media bag to HYPERStack vessel’s 3/8" I.D. liquid handling tubing.
2: Close liquid handling tubing clamp.
3: Close chase tubing clamp.
Filling the HYPERStack Vessel
1 2 3 4 5
1: Place HYPERStack vessel into fill position at a 10° angle and position media bag at
same level as the HYPERStack vessel.
2: Open liquid handling tubing clamp.
3: Open media bag tubing clamp. Fluid will enter the HYPERStack vessel.
4: Raise media bag to increase flow into the HYPERStack vessel, emptying the inoculated
portion of the media bag.
5: Remove clamp on top of media bag; finish filling HYPERStack vessel to the fill line.
To reduce rate of flow and prevent overfilling, lower media bag during final fill.
Isolate Liquid into HYPERStack Layers
1 2 3
4
✕
5 6 7
1: Place HYPERStack vessel into “isolation” position and lower media bag below
the height of the HYPERStack vessel.
2: Hold the chase filter upright and open the chase tube clamp. Allow excess media
to empty back into the media bag.
3: Close the media bag tubing clamp.
4: Open the liquid handling tubing clamp.
5: Return HYPERStack vessel to starting position.
6: Close chase tube clamp.
7: Close liquid handling tube clamp.
Place HYPERStack vessel into incubator. (Option: Empty media bag can be
removed or remain attached to the HYPERStack vessel.)
Keep the vent filter upright to prevent wetting out the filter.
Empty Spent Media from HYPERStack Vessel
1 2 3 4
5 6 7 8
1: Remove HYPERStack vessel from incubator; keep vent tubing upright.
2: Place HYPERStack vessel in “fill” position at a 10° angle.
Lower media bag below height of HYPERStack vessel.
3: Open the media bag tubing clamp.
4: Open the vessel’s liquid handling tubing clamp. Fluid will enter empty media bag.
5: Place HYPERStack vessel in final empty position and let all media drain.
6: Lift tubing to chase remaining fluid into the media bag.
7: Close liquid handling tubing clamp.
8: Close media bag tubing clamp. Remove spent media bag.
Keep the vent filter upright to prevent wetting out the filter.
Cell Harvest from HYPERStack Vessel
1 2 3 4 5 6
1: Connect cell dissociation and quench bags together to form harvest bag assembly.
2: Attach to HYPERStack vessel.
3: Raise harvest bag assembly above height of HYPERStack vessel and place it into
“fill” position at a 10° angle.
4: Open cell dissociation solution tube clamp.
5: Open harvest bag assembly tube clamp. Fluid will enter the HYPERStack vessel.
6: Close liquid handling tube clamp.
(continued on back)
Cell Harvest from HYPERStack Vessel (continued)
1 2 3 4 5 6
1: Close cell dissociation tube clamp.
2: Return HYPERStack vessel to the starting position; rock the vessel to distribute cell
dissociation solution.
3: After incubating the HYPERStack vessel for the required time for cell release, place
HYPERStack vessel in “fill” position at a 10° angle.
4: Open quench bag tube clamp.
5: Open liquid handling tube clamp. Quench solution will enter the HYPERStack vessel.
6: Close liquid handling tube clamp. Distribute the quench solution to completely
deactivate the cell dissociation solution.
1 2 3 4 5
1: Place harvest bag assembly lower than HYPERStack vessel.
2: Place the vessel in the empty position.
3: Open liquid handling tube clamp.
4: Transfer cell solution into empty quench bag.
5: Disconnect harvest bag assembly from HYPERStack vessel. Cell suspension is
ready for processing.
