Airway gene expression in smokers: an early diagnostic biomarker for lung cancer
The major goals of this project are to validate a gene-expression diagnostic biomarker for
lung cancer from histologically normal airway epithelial cells and integrate the biomarker into
predictive models that can be applied in a clinical setting; we also seek to develop a
biomarker in histologically normal airway epithelium capable of distinguishing subtypes of
lung cancer.
Role: Co-Investigator; direct gene-expression data analysis for biomarker validation and
refinement.
A non-invasive gene-expression biomarker of airway response to tobacco exposure
The major goals of this project are to use genome-wide expression profiling of upper airway
epithelial cells obtained non-invasively to develop host response biomarkers that reflect the
degree, intensity, and extent of recovery from tobacco-smoke exposure. We will also
develop biomarkers that assess the perturbation of physiologic pathways in response to
tobacco. These genomic biomarkers will be correlated with lung function and systemic
markers of oxidant stress and inflammation.
Role: Co-Investigator; direct gene-expression data analysis and biomarker discovery, testing
The Framingham Heart Study
One aspect of the long-running Framingham Heart Study (FHS) involves finding genetic
determinants of common medically important traits, and considerable recent effort has been
directed to finding SNP variants associated with these traits through Genome-Wide
Association Studies (GWAS). The informatic challenges presented by these studies are
formidable, but overlap considerably with the informatic challenges inherent in our Genome-
Wide Gene Expression Studies of lung disease. We are therefore collaborating with the
FHS investigators to build bioinformatic tools to store, manage, and integrate the results of
these studies; leveraging our expertise in this area.
Linking airway genomics to the pathogenesis and clinical heterogeneity of COPD
The major goals of this project are to apply whole-genome exon-level and microRNA
expression platforms and a variety of novel computational methodologies to identify
molecular subtypes of COPD that may contribute to the extreme clinical diversity of this
disease. We will also work to understand how these profiles change during disease
progression, and uncover genetic polymorphisms that influence both molecular and clinical
COPD-related traits. The hope is that this work will provide insights into the molecular
pathogenesis of COPD and specific COPD-related traits, including degree of airflow
obstruction, emphysema, small airways disease, and the rate of disease progression.
Molecular Profiles for Lung Cancer Pathogenesis and Detection in US Veterans
The overall hypothesis of this proposal is that smoking-induced chronic injury, inflammation,
and dysregulated epithelial repair result in a field of injury that supports lung carcinogenesis
and that the associated molecular, biochemical and cellular events will inform the networks
and pathways involved in lung carcinogenesis. We anticipate that this research will lead to
the discovery of biomarkers for early lung cancer detection and development of novel
therapeutic targets. These studies will be performed predominantly in veterans and will
provide novel screening methods and therapies for veterans, which will be applicable to the
broader population.
Detection of Early Lung Cancer Among Military Personnel (DECAMP)
A challenge inherent in the use of repeat imaging for lung cancer screening is that small
indeterminate nodules are found frequently, of which those that are related to a malignant
process represent only a small fraction. The goal of this project is to develop more
eeffective tools to more precisely identify individuals that will benefit from screening and to
rapidly differentiate the large number of benign radiographic abnormalities from those that
represent lung cancer following their initial detection.
