7. RESEARCH:<br />

The main research fields of Prof. Orlandi:<br />

1) cardiovascular pathology, investigating experimental model of<br />

atherosclerotic lesions and restenosis in vivo and restenotic and<br />

plaque cells in vitro, with reference to atherogenetic<br />

mechanisms, including risk factors as hypercholesterolemia and<br />

ageing. In the same fields, main subjects of research are the<br />

arterial cellular and extracellular modifications with ageing;<br />

experimental atherosclerosis and restenosis, proliferative and<br />

phenotypic modifications of arterial smooth muscle cells after<br />

vascular injury and the characterization of plaque cell<br />

population;<br />

2) arterial smooth muscle cells biological properties, with particular<br />

reference to the expression of proteins and genes regulating cell<br />

survival or death, in particular clusterin, NK-KB and apoptoticrelated<br />

proteins, such as Bax and cytochrome c; the possible<br />

pharmacological modulation of smooth muscle cell apoptosis in<br />

aimed to conteract restenosis in experimental models in vivo and<br />

in vitro;<br />

3) the study of cardiac tumors, with particular reference to the<br />

application of biomolecular methods, in order to clarify the<br />

mechanisms influencing their clinical behaviour and addressing<br />

new hypothesis concerning the origin of cardiac tumours still<br />

classified as of “unknown origin”, including myxoma and<br />

unclassified sarcoma; the application of more moder genetic


and biomolecular approaches, in conjuction with classical<br />

methods of investigation, will help to discover the origin of these<br />

tumors, in particular malignant sarcoma and linfoma of the<br />

heart, and better address the therapeutic approach to these<br />

tumours still with a afavourable prognosis;<br />

4) the study histomorphometrical study of human and experimental<br />

tumors, with particular reference to the cellular expression of<br />

cytoplasmic retinol and retinoic receptors. The investigation of<br />

the variable expression of this receptor may address new<br />

therapeutic prespectives aimed to moduly intracytoplasmic<br />

retinol derivative bioavailability;<br />

5) a project concerning the characterization of experimental<br />

models of human tumorigeneisis, with particular reference to the<br />

effect of new potential anti-angiogenetic drugs;<br />

6) The study of melanocytic and non-melanocytic skin tumors, with<br />

particular reference to mechanisms regulating proliferation and<br />

apoptosis of tumour cells and its pharmacological modulation;<br />

7) Role of retinoids in the phenotypic modulation of lung fibroblasts<br />

and myofibroblasts during tumor progression and usual interstitial<br />

pneumonia, with particular reference to proliferative and<br />

apoptic response of different fibroblast phenotypes.<br />
