7. RESEARCH:
The main research fields of Prof. Orlandi:
1) cardiovascular pathology, investigating experimental model of
atherosclerotic lesions and restenosis in vivo and restenotic and
plaque cells in vitro, with reference to atherogenetic
mechanisms, including risk factors as hypercholesterolemia and
ageing. In the same fields, main subjects of research are the
arterial cellular and extracellular modifications with ageing;
experimental atherosclerosis and restenosis, proliferative and
phenotypic modifications of arterial smooth muscle cells after
vascular injury and the characterization of plaque cell
population;
2) arterial smooth muscle cells biological properties, with particular
reference to the expression of proteins and genes regulating cell
survival or death, in particular clusterin, NK-KB and apoptoticrelated
proteins, such as Bax and cytochrome c; the possible
pharmacological modulation of smooth muscle cell apoptosis in
aimed to conteract restenosis in experimental models in vivo and
in vitro;
3) the study of cardiac tumors, with particular reference to the
application of biomolecular methods, in order to clarify the
mechanisms influencing their clinical behaviour and addressing
new hypothesis concerning the origin of cardiac tumours still
classified as of “unknown origin”, including myxoma and
unclassified sarcoma; the application of more moder genetic

and biomolecular approaches, in conjuction with classical
methods of investigation, will help to discover the origin of these
tumors, in particular malignant sarcoma and linfoma of the
heart, and better address the therapeutic approach to these
tumours still with a afavourable prognosis;
4) the study histomorphometrical study of human and experimental
tumors, with particular reference to the cellular expression of
cytoplasmic retinol and retinoic receptors. The investigation of
the variable expression of this receptor may address new
therapeutic prespectives aimed to moduly intracytoplasmic
retinol derivative bioavailability;
5) a project concerning the characterization of experimental
models of human tumorigeneisis, with particular reference to the
effect of new potential anti-angiogenetic drugs;
6) The study of melanocytic and non-melanocytic skin tumors, with
particular reference to mechanisms regulating proliferation and
apoptosis of tumour cells and its pharmacological modulation;
7) Role of retinoids in the phenotypic modulation of lung fibroblasts
and myofibroblasts during tumor progression and usual interstitial
pneumonia, with particular reference to proliferative and
apoptic response of different fibroblast phenotypes.
