The fourth exercise for year 2005 (2005-4) of the NASCOLA/ECAT External
Quality Assessment Program for Hemostasis and Thrombosis has be
shipped this week via Federal Express.
Page two of this correspondence contains all instructions necessary for
sample handling. Please review carefully before proceeding with storage
and reconstitution.
On the web site (www.nascola.org) you will find, in addition to this
document, the Methods and Instrument List for 2005 Survey
Program (from which you will choose codes for test methods and
instrumentation) and How to Report Survey Results via the Internet
(This Web Site). All laboratories will report results via our web site. You
will not receive any paper report forms. Laboratory codes (3 digits) and
password (8 digits) were distributed to all members for the first survey of
2005 and these have not changed. The deadline for internet reporting
is December 12, 2005.
As a reminder, should assays appear as on-line choices that do not match
those that you ordered/received, please remember that NASCOLA does
not subscribe to all modules provided by ECAT. Only those samples
denoted on page two are ones to which we subscribe.
Should you have any issues regarding this survey, please immediately
contact Dr Marlar or Melissa Goodsell in his laboratory. On behalf of the
NASCOLA Central Laboratory (Oklahoma City VA), we wish you our best
and hope you enjoy the Exercise!
EXERCISE 2005-4
CLOSING DATE December 12, 2005
WARNING
The plasma samples have been tested by an FDA approved method for the presence of HIV antigen as well as for
hepatitis B surface antigen and have been found to be negative. However, as with all preparations of human
origin, suitable precautions should be taken in their handling and disposal.
STORAGE AND STABILITY
Unreconstituted lyophilised plasma should be stored at 2-8°C or lower.
Reconstituted plasma should preferably be used within 1 hour after reconstitution. Plasma should be stored at 2-
8°C after reconstitution. For immunological methods the reconstituted plasma can be stored for 1 month at –
20°C.
RECONSTITUTION
For proper reconstitution the vial must reach room temperature before adding the water. Dissolve the contents of
each vial in sterile, distilled, room temperature water. For the exact volume of water to be used: see table below.
Leave the vial for 5 minutes. Swirl the vial gently to mix and leave for a further 15 minutes for complete reconstitution. Before
use mix the vial again gently.
Sample Code Volume (ml) Test
05.46 0.5 Antithrombin
05.47 0.75 Protein C activity and antigen
05.48 1.0 Protein S activity, total and free antigen
05.49 1.0 Activated Protein C Resistance Ratio
05.51 (2 vials) 1.0 (per vial) Lupus Anticoagulant
05.54 (2 vials) 1.0 (per vial) Coagulation Factors VIII, IX, XI and XII
05.55 (2 vials) 1.0 (per vial) Coagulation Factors VIII, IX, XI and XII
05.56 (2 vials) 1.0 (per vial) Coagulation Factors II, V, VII and X
05.57 (2 vials) 1.0 (per vial) Coagulation Factors II, V, VII and X
05.60 (2 vials) 1.0 (per vial) Von Willebrand Factor
The samples you receive depend on the test(s) on which you are registered
Special information for users of Latex Immuno Assays
Users of Latex Immuno assays should centrifuge the samples after reconstitution for at least 10 minutes
at 10.000 x g (or higher speed).
LISTS WITH CODES FOR METHODS AND EQUIPMENT
The lists with the codes for the methods and equipment used were provided separately in February.
This list will not be provided anymore with each survey. If you need an additional copy, please e-
mail to P.Meijer@ecat.nl
