POTENIAL SIGNIFICANCE OF LEAD LEAD-BINDING BINDING PROTEINS IN HUMAN TISSUES FOR RISK ASSESSMENTS • Individuals vary in their susceptibility to Pb poisoning • A portion of this variability at lower levels of Pb exposure appears t to be b due d in i part tt to genetic ti polymorphisms l hi in i major j lead -binding binding proteins in blood (eg. ALAD) which is also a sensitive biomarker of lead biological activity in the heme biosynthetic pathway. • Cellular mechanisms of Pb toxicity are not fully understood and may be modulated by a number of molecular mechanisms (eg. Pb - binding proteins in target tissues such as kidney and brain) • No apparent threshold for neurobehavioral toxicity 14
