Principles ofOperationConstant Cell Disruption Systems are based on the use of highpressure to force a sample through a small fixed orifice at highspeed under controlled, contained and repeatable conditions.1. As the high pressurepiston descends, sample is introducedinto the high pressurecylinder.2. The high pressure piston thenaccelerates the sample through the fixedorifice at high velocity. The controlledacceleration causes the cells to rupture.3. When the jet containing disrupted cells hits the target,it is spread radially across the cooled heat exchangesurface. Product then flows from the disruptionchamber for collection. The hydraulics are rechargedand the cycle continues.Reproducible ResultsConstant Cell Disruption Systems give reproducible and superior resultsbecause control of the disruption pressure is achieved by the hydrauliccontrol system. The disruption pressurecycle is shownadjacent. This has beencreated by analysingand recording thehydraulic pressure inthe operating cylinder, which is related to thedisruption pressure bythe ratio of the pistonareas.Pressure (bar x 100)302010DisruptionstrokeReturnstrokeRecharging periodfor hydraulics22 4 6 8 10 12 14Time (second)Graph showing a typical controlled pressure cycle in the cell disrupter40302010Pressure (psi x 1000)Contained ProcessThe disruption head, high pressure cylinder and cooling system have beendesigned with containment and safety in mind so that the volume of highpressure product is kept to minimum at all times. The integrity of thecontainment system has been tested by the Biosafety Testing Station at theCentre for Applied Microbiology Research, UK. “ ...a high level ofbio-containment.” “No evidence of emission from the equipment in any test.”
