Save time and hassle to buy separate items.Reagent Preparation<strong>Ultra</strong> <strong>BCA</strong> <strong>Protein</strong> assay requires 150 l of working reagent per well. Determine the total samples and 8 standards includingblank. Mix reagent A and B in 50:1 ratio (10 ml of A and 0.2 ml of B). Prepare just before the assay. Working reagent can bestored at room temp for several hours.<strong>Assay</strong> Procedure: Allow all reagents to reach room temperature. Arrange and label required number of strips.Prepare the <strong>BCA</strong> working reagents by mixing Reagents A and B (50:1). Dilute samples if necessary.Step 1. Use “pre-coated standard strip plate” and protein samples in duplicate into a fresh ELISA strip plate (wells).Dispense 150 l of <strong>BCA</strong> working reagent into all wells. Mix gently, cover the plate and incubate at 37 o C for 60 min. It isalso possible to incubate at room temp 25-28 o C (2 hrs).Step 2. Measure absorbance at A562 nm using an ELISA reader (540-590 nm filters may also be used, the absorbance valuesmay change slightly but the assay remains unaffected).
