Technical TipFebruary 2013<str<strong>on</strong>g>Use</str<strong>on</strong>g> <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>ROX</str<strong>on</strong>g><str<strong>on</strong>g>as</str<strong>on</strong>g> <str<strong>on</strong>g>reporter</str<strong>on</strong>g> <str<strong>on</strong>g>dye</str<strong>on</strong>g> <strong>on</strong> <strong>the</strong>LightCycler ® <strong>96</strong> <strong>System</strong>The LightCycler ® <strong>96</strong> <strong>System</strong> supports seven <str<strong>on</strong>g>dye</str<strong>on</strong>g>s for multicolorapplicati<strong>on</strong>s using Hydrolysis Probes:J Channel 1 (470/514): FAMJ Channel 2 (533/572): VIC, Hex, Yellow555J Channel 3 (577/620): Red 610, Tex<str<strong>on</strong>g>as</str<strong>on</strong>g> RedJ Channel 4 (645/697): Cy5<str<strong>on</strong>g>ROX</str<strong>on</strong>g> <str<strong>on</strong>g>dye</str<strong>on</strong>g> h<str<strong>on</strong>g>as</str<strong>on</strong>g> a similar excitati<strong>on</strong> and emissi<strong>on</strong> spectrum <str<strong>on</strong>g>as</str<strong>on</strong>g> Red610 and Tex<str<strong>on</strong>g>as</str<strong>on</strong>g> Red, and <strong>the</strong>refore can be used in place <str<strong>on</strong>g>of</str<strong>on</strong>g> Red610 or Tex<str<strong>on</strong>g>as</str<strong>on</strong>g> Red in LightCycler ® <strong>96</strong> multicolor applicati<strong>on</strong>s. Thefollowing observati<strong>on</strong>s are made:1. <str<strong>on</strong>g>ROX</str<strong>on</strong>g> is detected with sufficient sensitivity in Channel 3(577/620).2. The generic color compensati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>the</strong> LightCycler ® <strong>96</strong>s<str<strong>on</strong>g>of</str<strong>on</strong>g>tware compensates <str<strong>on</strong>g>ROX</str<strong>on</strong>g> quantitatively in multicolorapplicati<strong>on</strong>s, showing no visible remaining crosstalk.When using <str<strong>on</strong>g>ROX</str<strong>on</strong>g> in <strong>the</strong> LightCycler ® <strong>96</strong> <strong>System</strong>, select Red610 in <strong>the</strong> Detecti<strong>on</strong> Format dialog box. In all instances <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>the</strong>s<str<strong>on</strong>g>of</str<strong>on</strong>g>tware (sample editor, raw data, analysis modules), <str<strong>on</strong>g>ROX</str<strong>on</strong>g> data(e.g., amplificati<strong>on</strong> curves, heat map values, result table values)are <strong>the</strong>n displayed in <strong>the</strong> instance marked <str<strong>on</strong>g>as</str<strong>on</strong>g> Red 610.The enclosed data show a typical 4-color (4-plex) <str<strong>on</strong>g>as</str<strong>on</strong>g>say with <strong>the</strong>following <str<strong>on</strong>g>dye</str<strong>on</strong>g> combinati<strong>on</strong>:FAM + VIC + <str<strong>on</strong>g>ROX</str<strong>on</strong>g> + Cy5The data show various target combinati<strong>on</strong>s including <str<strong>on</strong>g>ROX</str<strong>on</strong>g>amplificati<strong>on</strong> <strong>on</strong>ly (see Figure 1), <str<strong>on</strong>g>ROX</str<strong>on</strong>g> amplificati<strong>on</strong> withall dual-color combinati<strong>on</strong>s (see Figures 2 – 4), all possibletriple color combinati<strong>on</strong>s (see Figures 5 – 8), and a 4-colorcombinati<strong>on</strong> (see Figure 9).
