PRECAUTIONS AND WARNINGS OF LEUPROLIDE
ACETATE
Leuprorelin is recommended for use in the treatment of hormone-responsive cancers
such as breast cancer and prostate cancer. It can also be used for estrogendependent
conditions such as uterine fibroids or endometriosis.
Leuprolide acetate used for the precocious puberty in males as well as females, and
to prevent premature ovulation in the cycles of controlled ovarian stimulation for in
the IVF (vitro fertilization).
Leuprorelin acetate may be used in order to reduce the risk of the premature ovarian
failure in females receiving cyclophosphamide for chemotherapy.
In combination with goserelin and triptorelin, the drug can be used in order to delay
the puberty in transgender youth until they are old enough to start the hormone
replacement therapy.
Warning/Precautions:
Patients who are with the urinary tract obstruction or metastatic vertebral
lesions should be precisely observed during the initial few weeks of the
treatment.
Bone mineral density changes may occur during the hypoestrogenic state.
Bone mineral density loss could be reversible after the withdrawal of the
leuprorelin acetate.
Post-marketing signs of the convulsions have been seen in patients taking
leuprorelin acetate.
Early signs of spinal cord compression such as paraesthesia must alert the
healthcare provider to the need for the intensive monitoring and probable
treatment.
Patients who have a history of allergic reactions to the benzyl alcohol, which
is an ingredient of this drug, can be responsible for signs of hypersensitivity.
Blood glucose or glycosylated haemoglobin (HbA1c) should be monitored
periodically in patients taking GnRH agonists and should be managed with the
current practice for the treatment of the hyperglycaemia or diabetes.
Patients with this drug should be monitored for the symptoms and signs
suggestive of the development of the cardiovascular disease and should be
managed according to the current clinical practice.
The leuprorelin effect on the bone lesions should be monitored by the bone
scans while its effect on the prostatic lesions should be monitored by
ultrasonography or CT scan in addition to the digital rectal examination.
In the event of ovary hyperstimulation, the gonadotrophin administration
should be interrupted while continuing leuprorelin acetate treatment for a few
days, in order to prevent the elevation of the luteinizing hormone (LH).
Avoid the use of Leuprorelin in pregnancy due to its embryotoxic effects.
Leuprorelin acetate is not recommended for administered to a nursing mother
as it is unspecified whether leuprorelin acetate is excreted into the human
milk.
Genotoxicity studies have been reported with the leuprorelin using the
bacterial as well as mammalian systems.
During the initial 2 months of the treatment, leuprolide acetate injection may
cause an increase in some hormones.
Do not take this drug if you have a history of the seizures or a history of
epilepsy.
