partec true Volumetric Absolute counting and the cyFlow ® space flow cuvette<br />
ensure that cells and particles are analysed and counted with the highest<br />
possible precision and accuracy.<br />
the cyFlow ® space analyses concentrations of any particle<br />
or cell subpopulation using true volumetric absolute counting<br />
(tVAc). this advanced technology is solely based on the<br />
fundamental definition of absolute counting i.e.: the particle<br />
concentration (c) is equal to the counted number (n) of cells<br />
in a given volume (v), c = n/v. In the cyFlow ® space, the volume<br />
is precisely measured directly by mechanical means, rather<br />
than indirectly with expensive and sometimes problematic<br />
beads, thus eliminating any errors related to varying bead<br />
concentrations or bead aggregations. the cyFlow ® space<br />
allows the analysis of a fixed volume as defined by the distance<br />
between two platinum electrodes. the volume can<br />
also be selected free of choice, based on digital sample<br />

aerosols, contamination of sorted samples, mechanical<br />

technology. the cyFlow ® sorter module includes a flow<br />
chamber with a diamond piezo element and an electronic<br />
device for piezoelectric activation. the piezo crystal rapidly<br />
and pre cisely deflects the sorted cells into a second channel<br />
of the flow cell. the cyFlow ® sorter is an optional add-on to<br />
the partec cyFlow ® space and serves as an optimized module<br />
for simplified and precise cell and particle sorting.
Non-destructive Particle and Cell Sorting based on Diamond Piezo Technology<br />
during piezoelectric activation (pictures 1 and 2 below),<br />
a gentle pressure wave is generated ( 3 ) and, as a conse-<br />
quence, a small volume of sheath fluid is deflected together<br />
with the cell or particle of interest ( 4 ). the piezo activation<br />
of the sorting process is repeated each time there is a cell<br />
to be sorted. the sorting process takes place very smoothly<br />
and without mechanical stress typical of droplet sorters.<br />
thus, even fragile cells or large particles are sorted by the<br />
cyFlow ® sorter without any distortion and other forces<br />
acting on the cells during deflection. A special feature of the<br />
cyFlow ® sorter is the correct sorting of All the cells in a<br />
region. the high yield is a result of the near-zero dead-time<br />
data acquisition system. no event is lost because the entire<br />
partec hardware and software acquisition works in real-time<br />
without any “interrupts” which often occur in other available<br />
instruments. therefore, no “wrong” cell can be sorted by<br />
mistake, thus directly achieving the highest sorting purity.<br />
the partec sorting system is extremely stable and produces<br />
no vibrations that might interfere with particle analysis,<br />

as can occur with droplet sorters, because the whole system<br />
is closed, non-elastic, and without air. the sort delay time<br />
is easily regulated based on the ccd camera screen, so no<br />
microscopic check is needed. the partec sorter works with low<br />
pressure differences in the capillary system. the morphology<br />
and vitality of cells (e.g. fragile particles like plant protoplasts)<br />
are not damaged by sorting. long sorting runs—e.g. for several<br />
hundred milliliters of sample volume—are possible. the sorting<br />
device can be sterilized safely. Any environmental contamination<br />
is completely avoided under normal conditions.<br />
the cyFlow ® sorter has the advantage of an extremely precise<br />
and stable system control of the time delay between measurement<br />
and piezo activation. Another unique feature of the<br />
cyFlow ® space is the built-in ccd camera which additionally<br />
allows an easy check of the whole analysis process directly<br />
at the screen of the instrument, this is also maintained while<br />
sorting (see the top left photograph on page 10: ccd camera<br />
showing the sorting process, sorted cells are deflected to<br />
the lower right).<br />
