INTRODUCTION<br />
This <strong>DAB</strong> (diaminobenzidine) substrate will produce a brown colored deposit upon<br />
reaction with HRP (horseradish peroxidase), and is intended to be used in<br />
immunohistochemistry protocols. This <strong>DAB</strong> product has improved stability, ease of<br />
formulation, and greater sensitivity than previous Zymed <strong>DAB</strong> products.<br />
REAGENTS PROVIDED<br />
A. One bottle (110 ml) <strong>DAB</strong> Substrate Buffer (Ready-To-Use)<br />
B. One dropper bottle (3.5 ml) <strong>DAB</strong> Chromogen (40X)<br />
PROTOCOL<br />
1. Add 1 drop (~26.5 µl) of <strong>DAB</strong> Chromogen (Reagent B) to 1 mL of <strong>DAB</strong><br />
Substrate Buffer (Reagent A). Mix well.<br />
2. Add to tissue specimen and develop for 5 min.<br />
3. Rinse for 2 minutes with running tap water.<br />
4. Proceed with normal counterstaining and mounting protocols.<br />
RECOMMENDED GUIDELINES FOR USE<br />
1. Signal development may be monitored under a microscope.<br />
2. Keep solution away from light and use within 2 hours.<br />

INTRODUCCIÓN<br />
Este sustrato producirá un depósito de color marrón al reaccionar con la peroxidasa.<br />
REACTIVOS SUMINISTRADOS<br />
A. 1 frasco (110 ml) de <strong>DAB</strong> Substrate Buffer (listo para usar)<br />
B. 1 frasco cuentagotas (3.5 ml) de <strong>DAB</strong> Chromogen (concentración 40x)<br />
PROTOCOLO<br />
1. Añadir una gota (~26.5 µL) de <strong>DAB</strong> Chromogen (Reactivo B) a 1 ml de <strong>DAB</strong><br />
Substrate Buffer (Reactivo A). Mezclar bien.<br />
2. Incubar la mezcla de reactivos durante unos 5 minutos a temperatura<br />
ambiente.<br />
3. Enjuagar por 2 minutos con el agua del grifo.<br />
4. A partir de este punto, el investigador debe proceder siguiendo sus propios<br />
protocolos de laboratorio para la contratinción y el montaje.<br />
DIRECTRICES DE USO RECOMENDADAS<br />
1. La evolución del color puede observarse con el microscopio.<br />
2. Mantener la solución apartada de la luz. No dejar pasar más de 2 horas<br />
antes de su utilización.<br />
ALMACENAMIENTO<br />
Guardar a una temperatura de entre 2ºC y 8ºC.<br />

INTRODUCTION<br />
Ce substrat produira un dépôt de couleur marron à la réaction du peroxydase.<br />
REACTIFS FOURNIS<br />
A. Une bouteille (110 ml) de <strong>DAB</strong> Substrate Buffer (prêt à l'utilisation)<br />
B. Une bouteille compte-gouttes (3.5 ml) de <strong>DAB</strong> Chromogen (40X)<br />
PROTOCOLE<br />
1. Ajouter 1 goutte (~26.5 µl) du <strong>DAB</strong> Chromogen (Réactif B) ΰ 1 ml de <strong>DAB</strong><br />
Substrate Buffer (Réactif A). Bien mιlanger.<br />
2. Incuber le mélange réactif pour ~5 minutes à température ambiante.<br />
3. Rincer pendant 2 minutes de l'eau du robinet.<br />
4. À partir d’ici, l’investigateur doit procéder avec ses propres protocoles de<br />
laboratoire pour la contre-coloration et la fixation.<br />
DIRECTIVES D'UTILISATION RECOMMANDEES<br />
1. Le développement des couleurs peut être régulé au microscope.<br />
2. Garder la solution loin de la lumière et utiliser la dans les 2 heures qui<br />
suivent.<br />

INTRODUZIONE<br />
Questo substrato produrrà un deposito di colore marrone in reazione con perossidasi.<br />
REAGENTI FORNITI<br />
A. Una boccetta (110 ml) di <strong>DAB</strong> Substrate Buffer (pronto per l’uso)<br />
B. Una boccetta contagocce (3.5 ml) di <strong>DAB</strong> Chromogen (40X)<br />
PROTOCOLLO<br />
1. Aggiungere 1 goccia (~26.5 µl) di <strong>DAB</strong> Chromogen (Reagente B) ad 1 ml di<br />
<strong>DAB</strong> Substrate Buffer (Reagente A). Mescolare bene.<br />
2. Incubare la mistura di reagenti per ~5 minuti a temperatura ambiente.<br />
3. Risciacquare con 2 minuti di acque di rubinetto.<br />
4. Da questo punto in poi, l’investigatore dovrebbe attenersi ai protocolli vigenti<br />
presso il proprio laboratorio per l’esecuzione delle procedure di<br />
controcolorazione e fissaggio.<br />
RACCOMANDAZIONI PER L’USO<br />
1. Lo sviluppo del colore può essere monitorato al microscopio.<br />
2. Conservare la soluzione al riparo dalla luce ed utilizzare entro 2 ore.<br />

EINFÜHRUNG<br />
Dieses Substrat wird mit der Reaktion auf Peroxidase eine rot gefärbte Ablagerung<br />

A. Eine Tropfflasche (110 ml) <strong>DAB</strong> Substrate Buffer (gebrauchsfertiger)<br />
B. Eine Tropfflasche (3.5 ml) <strong>DAB</strong> Chromogen (40X)<br />
PROTOKOLL<br />
1. 1 Tropfen (~26.5 µl) <strong>DAB</strong> Chromogen (Reagens B) auf 1 ml <strong>DAB</strong> Sustrate<br />
Buffer (Reagens A). Gut mischen.<br />
2. Die Reagensmischung für 5 Minuten bei Zimmertemperatur inkubieren.<br />
3. 2 Minuten in Leitungswaßer.<br />
4. Ab diesem Punkt sollte der Prüfer gemäß den eigenen festgelegten<br />
Laborprotokollen für Gegenfärbung und Mounting fortfahren.<br />
EMPFOHLENE BENUTZUNGSRICHTLINIEN<br />
1. Farbentwicklung kann unter einem Mikroskop überwacht werden.<br />
2. Die Lösung fern von Licht halten und sie innerhalb 2 Stunden benutzen.<br />
