Plasma/Serum HIV RT-PCR Kit
Plasma/Serum HIV RT-PCR Detection Kit
Norgen’s Plasma/Serum HIV RT-PCR Detection Kit constituents
a complete, ready-to-use system for the isolation and
detection of Human immunodeficiency virus using endpoint
RT-PCR. The kit first allows for the isolation of total
RNA, including viral RNA, from the plasma/serum samples
using a convenient spin-column. The viral RNA is isolated
free from PCR inhibitors, and can then be used as the template
in a RT-PCR reaction for HIV detection using the provided
HIV Master Mix. The HIV Master Mix contains reagents
and enzymes for the specific amplification of a 297 bp region
of HIV. In addition, Norgen’s Plasma/Serum HIV RT-PCR
Detection Kit contains a second heterologous amplification
system and controls to identify possible false negatives
and/or inadequate isolation. The kit is designed to allow for
the testing of 12 samples and is ideal for use in surveillance
of drug resistant pathogens, epidemiologial studies, field
surveillance of pathogens and surveys.
Human immunodeficiency virus (HIV) is a lentivirus (a member of the retrovirus family) that causes acquired
immunodeficiency syndrome (AIDS), a condition in humans in which the immune system begins to
fail, leading to life-threatening opportunistic infections. Infection with HIV occurs by the transfer of blood,
semen, vaginal fluid, pre-ejaculate, or breast milk. Within these bodily fluids, HIV is present as both free
virus particles and virus within infected immune cells. The four major routes of transmission are unsafe sex,
contaminated needles, breast milk, and transmission from an infected mother to her baby at birth
(vertical transmission). Screening of blood products for HIV has largely eliminated transmission through
blood transfusions or infected blood products in the developed world. HIV infection in humans is considered
pandemic by the World Health Organization (WHO).
Plasma/Serum HIV RT-PCR Detection Kit Benefits
A complete optimized kit for
isolation and detection
The Plasma/Serum HIV RT-PCR Detection Kit is a complete kit that
contains: 1) all the required components to allow for optimized isolation
of viral RNA, and 2) HIV Master Mix and controls to allow for the
optimized amplification and detection of the viral RNA.
High Sensitivity and Specificity The limit of detection is less than 1000 VP/mL of plasma/serum
Isolate high quality, inhibitorfree
RNA
Removal of highly concentrated salts, metabolic wastes and proteins
provides high quality, concentrated RNA.
Rapid Procedure Isolate, amplify and detect HIV in under 3 hours.
Includes controls to identify
PCR inhibition and problems
with RNA isolation
Ideal for use in a number of
different applications
The HIV 2X RT-PCR Master Mix contains an independent PCR control
to identify false negatives. An HIV Isolation Control (IsoC) is also supplied
to allow the user to control the DNA isolation procedure.
Ideal for use in surveillance of drug resistant pathogens, epidemiologial
studies, field surveillance of pathogens and surveys.
Figure 1. Sensitivity of Detection using the Plasma/Serum HIV RT-PCR Detection Kit. A representative 1X
TAE, 1.7% agarose gel showing the amplification of HIV at different concentrations (Target). The size of the
HIV target amplicon corresponds to the 297bp band represented by the provided DNA Marker (M). The
size of the HIV Isolation Control (IsoC) corresponds to the 500bp band represented by the provided DNA
Marker (M). The HIV 2X RT-PCR Master Mix contains a HIV PCR Control (PCRC). The HIV PCRC controls for
PCR inhibition. The size of the HIV PCRC corresponds to the 150bp band represented by the provided DNA
Marker (M). Lanes A-H represents samples spiked with different HIV concentrations isolated from 2 mL
plamsa samples (interpreted as positive results). The HIV spiked in plasma samples is an in vitro transcribed
HIV RNA fragments.
Storage Conditions
The Positive Control (PosC) and Isolation Control
(IsoC,) should be stored at -70°C. If needed, make
aliquots of the controls according to the volume
used in the protocol prior to freezing. The 2X RT-PCR
Mastermix should be stored at -20°C. Make appropriate
aliquots if needed. All other kit components
may be stored at room temperature. The 2X RT-
PCR Mastermix, Postive Control and Isolation Control
should not undergo repeated freeze-thaw (a
maximum freeze-thaw of three times). Do not use
kit or reagents past their expiration dates
