PELONews Spring/Summer 2023 Organ/Cell based products Hepatocytes Primary human hepatocytes are the main cells in the liver, characterized by their cubical shape and abundant smooth endoplasmic reticulum. They exhibit eosinophilic cytoplasm, basophilic stippling, and round nuclei with dispersed chromatin. Hepatocytes form plates separated by sinusoids, which have fenestrated endothelial linings and are supported by reticulin. Kupffer cells and stellate cells are present in the sinusoids, performing phagocytosis and storing vitamin A, respectively. These hepatocytes “gold standard” for in vitro hepatic assays such as Drug Metabolism and Pharmacokinetics (DMPK).
