Sartorius offers a wide selection of filter types, filter holders <strong>and</strong> ready-to-use<br />

filtration units. There are certainly reasons <strong>for</strong> you to use these products if you<br />

are engaged in analysis, research, quality control or production. But which are<br />

The choice naturally depends on your particular application.<br />

Air filter holders / units<br />

The Midisart unit (bottom right) <strong>and</strong> the Sartofluor Capsule (middle left) are mainly used <strong>for</strong> sterile venting. Both are<br />

ready <strong>for</strong> use, <strong>and</strong> repeatedly autoclavable <strong>for</strong> re-use. The stainless steel housing (top centre) is used <strong>for</strong> the same<br />

purpose, in combination with Sartofluor Mini Cartridges (see page 81).<br />

The stainless steel holder <strong>for</strong> 47 mm membranes (middle right) is mainly used <strong>for</strong> in-line air/gas filtration, as are the<br />

smaller 25 mm holder <strong>and</strong> the Mini Cartridge housings which are not shown here (see pages 52 <strong>and</strong> 53).<br />

2000 Sterile Venting Units,<br />

light in weight <strong>and</strong> easy to connect<br />

Re-usable complete filtration units with<br />

naturally hydrophobic PTFE membrane<br />

<strong>for</strong> reliable sterile venting of small<br />

fermenters <strong>and</strong> of containers <strong>for</strong> culture<br />

media.<br />

Midisart 2000 units have been<br />

designed <strong>for</strong> maximum h<strong>and</strong>ling ease<br />

<strong>and</strong> reliability. Tapered hose barbs<br />

ensure a simple, secure hold <strong>for</strong> 6 to<br />

12 mm i.d. tubing. The light weight,<br />

just 20 g, will not weigh down <strong>and</strong><br />

kink tubing. The membrane is rein<strong>for</strong>ced<br />

with polypropylene gauze <strong>for</strong> stability<br />

to pressures of up to 3 bar. The 20 cm 2<br />

large filter area allows high air flow<br />

rates at low differential pressures.<br />

Each unit is printed with lot number<br />

<strong>and</strong> individual piece number on the<br />

housing <strong>for</strong> total security <strong>and</strong> traceability.<br />

Capsule filters<br />

Sartofluor ®<br />

Capsules with PTFE membrane<br />

<strong>for</strong> maximum security in sterile venting<br />

Sartofluor Capsules are ready-toconnect,<br />

pre-tested, complete filter<br />

units. The PTFE membrane is pleated<br />

to obtain the largest possible usable<br />

filtration area in the small, translucent<br />

polypropylene housing. The two vents<br />

in the upstream side of the housing<br />

ensure good steam passage <strong>and</strong><br />

corresponding certainty of sterilization<br />

of the Capsules by autoclaving.<br />

The extreme hydrophobicity of the<br />

PTFE membrane ensures maximum<br />

reliability, even when filtering moist<br />

air. The high air flow rates of the<br />

membranes <strong>and</strong> the large filtration<br />

area allow effective sterile venting at<br />

low differential pressures.<br />

Centrisart-C4 microconcentrators are<br />

volume ultrafiltration or microfiltration.<br />

Five types are available. Major<br />

applications are the concentration<br />

<strong>and</strong> purification of proteins <strong>and</strong><br />

DNA, buffer exchange, desalting, the<br />

removal of low molecular weight<br />

substances such as primers, non-incorporated<br />

nucleotides <strong>and</strong> excess<br />

linkers (5, 10, 20 or 100 kD units),<br />

as well as the removal of agarose<br />

particles from eluted DNA <strong>and</strong> the<br />

retention of cells <strong>and</strong> bacteria<br />

(0.2 µm unit)<br />

Identification of the units is quick <strong>and</strong><br />

easy. The MWCO (or pore size) <strong>and</strong><br />

an abbreviation <strong>for</strong> the filter material<br />

are printed on the outer (filtrate) tube,<br />

<strong>and</strong> an etched area can be used <strong>for</strong><br />

individual labelling.<br />

The larger Centrisart-C30 units are<br />

used <strong>for</strong> the same applications when<br />

larger sample volumes, up to 3 ml,<br />

are to be processed.<br />

Centrisart I consists of an outer centrifuge<br />

tube, in which the sample is<br />

placed, <strong>and</strong> a free-sliding inner tube,<br />

closed on the underside with an<br />

ultrafilter. During centrifugation, the<br />

inner tube is pulled down against the<br />

sample, so that liquid is <strong>for</strong>ced<br />

through the ultrafilter, while particles<br />

<strong>and</strong> retained proteins are simultaneously<br />

pulled away from it. This is so<br />

effective in preventing blockage that<br />

even whole blood samples can be<br />

deproteinated.<br />

EasyFlow ®<br />

single use units with pleated<br />

A ready-to-connect EasyFlow unit<br />

<strong>and</strong> a peristaltic pump represent the<br />

simplest way to ultrafilter laboratory<br />

scale batches. Applications include<br />

protein concentration, diafiltration,<br />

virus retention <strong>and</strong> pyrogen removal.<br />

In addition to the ultrafilter units, an<br />

0.2 µm membrane filter unit is also<br />

available <strong>for</strong> cell harvesting.<br />

EasyFlow units are supplied sterile<br />

<strong>and</strong> pyrogen free. They are intended<br />

<strong>for</strong> single use, but can be cleaned<br />

<strong>and</strong> sanitized to some extent. The<br />

pleated ultrafilter enables a large<br />

filter area to be incorporated in a<br />

Collodion Bags<br />

Collodion Bags are used <strong>for</strong> gentle,<br />

small volume protein concentration,<br />

immersed in water in a glass holder<br />

connected to vacuum. The time taken<br />

depends upon the degree of concentration<br />

required <strong>and</strong> the starting<br />

volume. 5 ml of spinal fluid are<br />

concentrated down to about 20 µl in<br />

1.5 to 2 hours. The smaller Micro<br />

Collodion Bags of 1 ml capacity,<br />

used in a pressure system, allow far<br />

quicker protein concentration. 1 ml<br />

of spinal fluid can be concentrated to<br />

4 µl within about 4 minutes.<br />

The use of Sartorius Collodion Bags<br />

in the isolation of high molecular<br />

weight DNA from mammalian cells is<br />

described in ”Molecular Cloning. A<br />

Laboratory Manual“, Sambrook,<br />

Fritsch <strong>and</strong> Maniatis, Cold Spring<br />

Harbor Laboratory Press, 1989. The<br />

procedure is more gentle, <strong>and</strong> gives<br />

higher DNA yields, than<br />

Sartocon Slice cassettes are pictured<br />

the right. The same construction <strong>and</strong><br />

materials are used in each of these<br />

cassette types, <strong>and</strong> the flow path<br />

lengths are identical, so that scale-up<br />

or scale-down is perfectly linear<br />

throughout the range of applications,<br />

Ultrasart ® X<br />

The spiral flow cell which avoids<br />

the drawbacks of stirred cells<br />

In ultrafiltration, liquid movement<br />

across the filter surface is necessary<br />

to prevent a filter-blocking layer of retained<br />

material from building-up on it.<br />

This is achieved in stirred cells by the<br />

rapid rotation of a stirrer bar. Ultrasart<br />

X uses the more gentle <strong>and</strong> effective<br />

approach, recirculation across the filter<br />

in spiral flow mode.<br />

The extra top part next to an assembled<br />

Ultrasart X (above) is inverted to<br />

show the spiral recirculation channel.<br />

As shown below, the recirculation inlet<br />

<strong>and</strong> outlet are both on the top part, allowing<br />

simple connection to a peristaltic<br />

pump.<br />

Whereas adding more sample to a<br />

stirred cell entails stopping the run<br />

<strong>and</strong> opening the cell, Ultrasart X offers<br />

requirements are a peristaltic pump<br />

To obtain maximum yields with an<br />

ultrafiltration system, the pump,<br />

container, ultrafiltration unit <strong>and</strong><br />

tubing system must be designed to<br />

match each other exactly <strong>and</strong> to<br />

keep the fluid pathways as short as<br />

possible. This has been excellently<br />

achieved in Perisys 70, a complete<br />

system which requires little bench<br />

space (only 185 x 235 mm), is easy<br />

to assemble <strong>and</strong> use <strong>and</strong> ensures<br />

highest recoveries.<br />

The dead volume of the tubing is less<br />

than 0.5 ml. The 1 liter glass container<br />

has 100 ml graduations <strong>and</strong> can<br />

be autoclaved together with the<br />

tubing <strong>for</strong> certain sterilization <strong>and</strong><br />

avoidance of cross-contamination.<br />

The peristaltic pump housing is made<br />

of stainless steel, the pumphead of<br />

POM. The pump supplies flow rates<br />

of 0.4 to 620 ml/min <strong>and</strong> accepts<br />

tubing of 3.2 to 6.4 mm internal<br />

Collodion Bags<br />

Supplied with three polycarbonate<br />

supports of 1 ml capacity, each <strong>for</strong> a<br />

single Micro Collodion Bag. After<br />

insertion of a bag, a support is<br />

simply snapped into one of the three<br />

quick-connectors on the manifold. The<br />

connector is only open when a<br />

support is in position. Connection of<br />

the quick-connect nipple pressure inlet<br />

of the manifold to a pressure source<br />

requires a pressure hose with G1/4<br />

threaded nut (order number 16929),<br />

or with G3/8 threaded nut (order<br />

number 16931), according to the<br />

male thread of the pressure source<br />

outlet. The maximum operating<br />

can be screwed directly into the ports<br />
