Carboxylic Acid to Amine Conjugation<br />

EDC and Sulfo-NHS are widely used to modify a carboxylic<br />

acid functional group to become reactive with a primary<br />

amine. EDC and Sulfo-NHS convert the carboxylic acid to<br />

an NHS-Ester, which readily reacts with primary amines<br />

on proteins, peptides, antibodies or even small molecules.<br />

Common uses include forming protein conjugates and<br />

immobilizing proteins or antibodies onto a solid support<br />

such as glass, plastic or agarose bead. The final conjugate<br />

has a stable and covalent amide bond holding the two<br />

moieties together.<br />

Amine to Sulfhydryl Conjugation<br />

Sulfo-SMCC is commonly used as a crosslinker to conjugate<br />

enzymes to antibodies or to conjugate antigens to carrier<br />

proteins. Sulfo-SMCC first labels primary amines with a<br />

cysteine-reactive functional group called a maleimide. The<br />

Maleimide targets reduced sulfhydryls, such as those found<br />

in the side chains of the amino acid cysteine, in the second<br />

step of the reaction.<br />

Amine to Amine Conjugation<br />

Because amine chemistry requires no amino acid modification<br />

and proceeds readily in physiological conditions, many<br />

researchers simply use crosslinkers that link to primary aminecontaining<br />

biomolecules. This is commonly used to immobilize<br />

antibodies to beads coated with Proteins A, G or A/G. Two<br />

commonly used reagents include BS 3 and DSS, which differ<br />

only by the increased solubility of BS 3 in aqueous buffers.<br />
