STRESS AND NO 5I_01_P (poster section B1, poster board #239, 25 th <strong>of</strong> August) WITHANIA SOMNIFERA INDUCES THE STRESS INDUCED MODULATION OF NEUROBEHAVIORAL PATTERNS VIA NITRIC OXIDE SYNTHASE Zaved Ahmed Khan 1 , Iftikhar Aslam Tayubi 2 1,2School <strong>of</strong> Biotechnology , Chemical and Biomedical Engineering,Vellore Institute <strong>of</strong> Technology University, Vellore TN-632014, India Withania somnifera (ashwagandha, Indian ginseng) is an immunostimulant herbal medicine used to improve overall health and prevent diseases, particularly in the elderly. We had found to be an effective stress modulator herbal medicine. However, the mechanisms underlying its anxiolytics effect is poorly understood. To elucidate the mechanism <strong>of</strong> Withania somnifera, we investigated the effect <strong>of</strong> a methanolic extract from the root and stem <strong>of</strong> Withania somnifera (WS) on nitric oxide (NO) production in wistar rats.We found that WS (1–256 µg/ml) produced a significant and concentration-dependent increase in NO production, an effect which was abolished by N-nitro-L-arginine methyl ester (L-NAME, 3–300 µM), a nonselective inhibitor <strong>of</strong> NO synthase (NOS).As it has been already shown about the involvement <strong>of</strong> nitric oxide (NO) in stressinduced neurobehavioral changes in rats .Moreover, elevated plus maze (EPM) and open field test (OFT) analysis showed that WS increased, in a concentration-dependent fashion modulates stress induced neurobehavioral patterns. Activity <strong>of</strong> NOS was checked using NOS detection system(Sigma FCANOS ) in brain sample. These results demonstrate that WS may induce the synthesis <strong>of</strong> neuronal NOS expression likely by acting at transcriptional level. nNOS level is checked by western blot analysis. The increased NO production by brain could account, at least in part, <strong>for</strong> the anxiolytics properties <strong>of</strong> Withania somnifera. 5I_02_P (poster section B1, poster board #240, 25 th <strong>of</strong> August) ROOT DEVELOPMENT UNDER OSMOTIC STRESS AND IN THE PRESENCE OF EXOGENOUS AUXIN IN PISUM SATIVUM L.: THE ROLE OF NITRIC OXIDE Zs. Erdei Department <strong>of</strong> Plant Physiology, University <strong>of</strong> Szeged, P.O.B. 654., 6701 Szeged, Hungary e-mail: kolzsu@bio.u-szeged.hu; erdei@bio.u-szeged.hu During this work the effects <strong>of</strong> exogenous auxin (indolebutyric acid, IBA) and osmotic stress on root morphology and nitric oxide (NO) generation in roots were compared in pea plants. Five-day old plants were treated with IBA at a concentration range <strong>of</strong> 10 -9 - 10 -3 M and osmotic stress was induced by 50-400 mOsm polyethylene glycol (PEG 6000) <strong>for</strong> another 5 days. NO generation was examined by in situ and in vivo fluorescence method. Both root characteristics and NO intensities were followed as the function <strong>of</strong> time. The increasing concentrations <strong>of</strong> IBA as well as PEG resulted in shortening <strong>of</strong> primary roots (PRs), enhancement <strong>of</strong> lateral root (LR) number and significant increase <strong>of</strong> NO generation. By virtual sectioning <strong>of</strong> PRs, it was determined that the accumulation <strong>of</strong> NO occurred at the basis <strong>of</strong> LRs during both treatments. Timedependence investigations revealed that in the case <strong>of</strong> IBA treatments, the LR number increased in parallel with an intensified NO generation, while elongation <strong>of</strong> PRs was not followed by changes in NO levels.
