Western Blotting
The western blot is an analytical technique used to detect
specific proteins in a given sample of tissue homogenate or
extract. It uses gel electrophoresis to separate native or
denatured proteins by the length of the polypeptide
(denaturing conditions) or by the 3-D structure of the protein
(native/ non-denaturing conditions).
In Western blotting, water is used to prepare the transfer
buffer, buffers used to prepare the blocking solution,
primary and secondary antibodies, and the wash
solutions. It is critical to use water that is free of
contaminants that are biologically active and could
interfere. Apyrogenic type I ultrapure water is essential.
