RNomics – Analysis of novel in silico derived non-protein<br />

coding RNAs in the human genome<br />

Since the increased complexity of vertebrates over invertebrates (e.g. human versus<br />

C.elegans with 10 14 and 10 4 cells, respectively) cannot be explained by a mere 1.5 fold<br />

increase in protein-coding genes, it becomes inevitable that genomic sequences resulting<br />

in non-protein coding RNA molecules (npcRNAs) might play a much more substantial role<br />

in gene regulation networks, than expected. Increasing evidence shows that most of the<br />

complex genetic phenomena in higher organisms, such as gene silencing, imprinting,<br />

targeting, splicing, RNA editing and methylation are connected to RNA signalling and the<br />

involvement of different classes of npcRNAs is the rule rather than the exception. The<br />

rapidly growing list of functional npcRNAs shows great diversity and suggests that by<br />

now we have only seen a small fraction of all functional npcRNAs.<br />

Within an Exploratory Project of the German National Genome Research Network<br />

(NGFN), we have set out to investigate the possibility to identify novel npcRNAs encoded<br />

in the human genome by a computational approach followed by the evaluation through a<br />

set of experimental methods including RT-PCR, real-time PCR and northern<br />

hybridisation. At a later stage, the novel npcRNAs will be assessed for expression levels<br />

in different tissues and screened for potential interaction partners in active RNP<br />

complexes through gel retardation assays, phage-displayed cDNA expression product<br />

library screening, mass spectroscopy and transfected cell arrays.<br />
