Instructi<strong>on</strong>s for <str<strong>on</strong>g>setting</str<strong>on</strong>g> <str<strong>on</strong>g>up</str<strong>on</strong>g> <str<strong>on</strong>g>DIC</str<strong>on</strong>g> <strong>on</strong> <strong>the</strong> <strong>Zeiss</strong> <strong>Axioskop</strong>First:Make sure <strong>the</strong> 10x or 20x objective or higher is in placeMake sure <strong>the</strong> polarizing lens (8) is OUT of <strong>the</strong> light pathMake sure <str<strong>on</strong>g>DIC</str<strong>on</strong>g> c<strong>on</strong>denser (9) is in place1. Turn <strong>on</strong> <strong>the</strong> light source2. Put a sample <strong>on</strong> <strong>the</strong> stage and bring it into focus3. close down <strong>the</strong> field diaphragm all <strong>the</strong> way until you see a brightspot surrounded by darkness4. focus <strong>the</strong> edges of <strong>the</strong> bright spot by moving <strong>the</strong> c<strong>on</strong>denser <str<strong>on</strong>g>up</str<strong>on</strong>g> ordown slightly . NOTE: If you have to move <strong>the</strong> c<strong>on</strong>denser a lot, andyou are <strong>on</strong> <strong>the</strong> 10x objective or higher, make sure <strong>the</strong> fr<strong>on</strong>t lens(12)is in <strong>the</strong> light path, <strong>the</strong>n move it back <str<strong>on</strong>g>up</str<strong>on</strong>g> to <strong>the</strong> top and start again5. Once you can see a sharp octag<strong>on</strong>, center <strong>the</strong> bright spot with <strong>the</strong>two silver c<strong>on</strong>denser alignment knobs6. Open <strong>the</strong> field diaphragm (3) just enough that you can’t see <strong>the</strong>edges7. swing <strong>the</strong> polarizing lens into <strong>the</strong> light path8. Adjust brightness with light intensity c<strong>on</strong>trol (1)9. open c<strong>on</strong>denser about 2/3 of <strong>the</strong> way so that it is as bright aspossible while still having good c<strong>on</strong>trast10. c<strong>on</strong>trast can also be adjusted with small silver knob immediatelyabove objective (Nomarski prism)Note: if you change to a higher objective, you will need to close down<strong>the</strong> field diaphragm so that <strong>the</strong> light <strong>on</strong>ly just fills <strong>the</strong> field of view.
