Thermocyclers
Thermo Scientific Piko Thermal Cyclers
deliver high performance in a compact package. Although merely half the size of other
instruments, the Piko® Thermal Cycler meets the highest criteria in performance and is able
to complete a PCR protocol in less than 15 minutes. This is achieved using novel technical
advances that allow significant reduction in PCR run times and overall size of the instrument. The
Piko Thermal Cycler is an ideal solution for both conventional and fast PCR applications. Due to
its small size and low power consumption, the Piko Thermal Cycler is also conveniently portable
for field use.
Thermo Scientific Piko Thermal Cyclers offer you
• Best-in-class thermal performance Consistent results from well-to-well due to extremely
short settling times and high temperature uniformity across the block.
• Time savings PCR in as little as 15 minutes enabled by fast ramp rates and quick settling
times.
• Reagent savings Repeatable yields from as little as 5 μL when using UTW vessels and heat
sealers.
• Space savings The smallest footprint available - fits into a tiny bench space.
• Energy saving Short protocols and low wattage requires only ¼ the power consumption of
typical thermal cyclers.
• Repeatable sealing to prevent sample evaporation Automatic heated lid provides consistent
and tight sealing from run-to-run.
Two formats: 24- and 96-well blocks
Piko Thermal Cyclers are available in two different block configurations: 24-well and 96-well. 24well
cyclers accept all standard low profile single tubes, and 8-tube strips with flat caps as well
as 24-well Piko PCR Plates. The 96-well instrument utilizes 96-well Piko PCR Plates, which are
only ¼ the size of a conventional microplate. Despite the small size, all Piko PCR Plates maintain
industry standard well spacing and sample capacity. The 24-well and 96-well Piko PCR Plates
are compatible with standard Thermo Scientific multichannel pipettes, Multidrop® reagent
dispensers and the Versette automated liquid handling system.
High yields and uniform results in less than 15 minutes
The best-in-class thermal performance of Piko Thermal Cyclers means significant reduction
of incubation times in most PCR protocols. This is due to the fast ramp rates, near-perfect
temperature uniformity and incredibly short block settling time. It only takes 1 second for all the
samples to reach temperature equilibrium.
A 400 bp amplicon from lambda DNA was amplified with an ultra-fast PCR protocol on a 24-well Piko
Thermal Cycler using UTW reaction vessels and Phusion Flash High-Fidelity PCR Master Mix. 5 μl aliquots
of each 15 μl reaction were run on an agarose gel. The following PCR protocol was used: 25 cycles of [98°C
for 0 sec, 72°C for 0 sec], 72°C for 10 sec. Total cycling time was about 10 min.
VIAFLO 96 Unique
Multichannel microplate pipetting is a growing task because
of the need to increase productivity and improve
the quality of the data obtained by increased numbers
of replicates. In addition to the large investments that
are traditionally required to obtain such instrumentation,
many customers are also concerned about the complexities
associated with using an automated system.
INTEGRA’s VIAFLO 96 introduces a totally new
concept in 96 well pipetting, which is affordable and
simple to use. It renders 96 channel pipetting as easy
as manual single channel pipetting.
VIAFLO 96 performs all tasks easily in multiwell format,
just as a standard pipette would in a single tube.
Applications may include:
 Replication or reformatting of plates
Transfer of reagents and compounds from plate to
plate or within plates
Gentle and convenient dissolution or re-suspension
of solid compounds
Fast execution of serial dilutions by columns
 Increment dispensing to multiple plates
VIAFLO 96 is used like a hand-held pipette. The
movement and positioning of the 96 channel pipette
is supported by a servo assisted steering mechanism
which allows fast, precise and stress free multichannel
pipetting.
Just by moving the assisted pipetting head between
two plate positions, as you would move a hand-held
pipette, you are able to simply perform 96 channel
pipetting!
Therefore, no special skills or training are required to
work with this small bench top unit. Immediately
benefit from a whole new level of usability, ergonomic
comfort and productivity.
The schematic structure of Phusion® High-Fidelity DNA Polymerase. The double-strand DNA-binding
domain (purple) is fused to a novel Pyrococcus-like proofreading enzyme (green) forming a unique highperformance
polymerase - Phusion DNA Polymerase.
The unique Phusion® DNA Polymerase offers performance that no other enzyme can match.
The Phusion DNA Polymerase generates templates with an accuracy and speed unattainable
with a single enzyme, even on your most difficult templates. The unique structure and
characteristics of Phusion DNA Polymerase make it a superior choice for cloning, and sets
a new standard for PCR performance: its error rate is 50-fold lower than that of Thermus
aquaticus, and 6-fold lower than that of Pyrococcus furiosus - thus being the most accurate of
available thermostable polymerases. Phusion DNA Polymerase possesses processivity 10-fold
greater than Pyrococcus furiosus.
Accuracy - The highest fidelity of any available thermostable polymerases
Speed - Increased processivity allows shorter reaction times (extension 15-30 s/kb)
Robustness - Fewer reaction failures and minimal optimization
High yields - Increase product yields with minimal enzyme amounts
Specificity - Hot start modification reduces non-specific amplification and primer degradation
illustra RT-PCR is commonly used to amplify cDNA by combining first-strand cDNA synthesis
of an RNA template with PCR amplification. Typically the reaction is performed in two steps.
A first-strand cDNA reaction is performed, the resulting cDNA is then transferred to another
tube containing Taq DNA polymerase and PCR buffer where the reaction mixture is subjected
to multiple cycles of denaturation, annealing and elongation, resulting in the exponential
amplification of the target cDNA. Ready-To-Go RT-PCR Beads simplify this process to a
single-tube, single-reaction procedure. ’
Ready-To-Go RT-PCR Beads have been optimized for RT-PCR. Each room-temperature-stable
bead contains M-MuLV Reverse Transcriptase, RNase Inhibitor, Buffer, Nucleotides and
Taq DNA polymerase. The only additional reagents required are water, template RNA, and
primers.
The Ready-To-Go Bead format significantly reduces the number of pipetting steps, thereby
increasing reproducibility of the RT-PCR technique and minimizing risk of contamination and
RNA degradation. Ready-To-Go RT-PCR Beads are provided in either thin walled 0.5 ml or
A fast and reliable system for generating mutations
The Phusion® Site-Directed Mutagenesis Kit is a versatile and efficient tool for introducing
point mutations, insertions or deletions in any type of plasmid DNA. With this kit, the entire
plasmid is amplified using phosphorylated primers that introduce the desired changes. The
amplified, linear PCR product, containing the desired mutation, is circularized in a 5-minute
ligation reaction with Quick T4 DNA Ligase. The resulting plasmid can be then transformed
into any competent E. coli cells.
• Robust and reliable exponential amplification method
• No requirements such as special vectors, restriction sites or methylation status for the
target plasmid
• No need to destroy the starting template in a separate step
• Due to the high fidelity of Phusion® Hot Start II DNA Polymerase unwanted secondary
mutations are minimized
• Amplification of large plasmids up to 10 kb
• The hot start modification in the polymerase prevents the amplification of non-specific
products and unwanted degradation of primers prior to first cycle of PCR
2x Reddymix PCR Master Mix
ThermoPrime Taq DNA Polymerase is an ultra-pure recombinant thermostable
DNA polymerase obtained by high level expression of the Taq gene
in Escherichia coli. With enhanced thermal stability, non-specific binding
of template and primers is minimized leading to a decrease in the amount
of non-specific product. Being a recombinant enzyme, it is characterized by
significantly higher purity than those enzymes that are native. This higher
level of purity gives superior lot-to-lot reproducibility. ThermoPrime has 5’
to 3’ Taq DNA polymerase activity as well as 5’ to 3’ strand displacement (exonuclease)
activity. The convenient Master Mix format contains all the components required for PCR (with
the exception of template and primers), reducing protocol steps and minimizing the risk of
pipetting errors and sample contamination
White PCR Products for qPCR
The use of Real-Time PCR (qPCR) is becoming increasingly widespread. In particular, the direct
quantification of the DNA formed can be achieved by the use of fluorescence measurement.
The new line of white PCR products by BRAND offers significantly better results in this
application than transparent 8-tube strips or plates. The different products in this line are
uniformly colored with TiO2 (titanium dioxide), so that in combination with the smooth
surfaces, the optimum reflection of the fluorescence signal is provided.
PCR cooler from Eppendorf
By changing color from violet to pink or from
dark blue to light blue, the PCR Cooler shows
when upper temperature value of 7°C has
been exceeded. Decisive for the cooling of
samples is thereby the color value in the
depressions of the PCR Cooler. The individual
temperature of single tubes can be determined
from the coloring of the respective receptacle
bore.
• Cools sensitive samples reliably for more
than an hour at 0°C (with 2 hour precooling at -20°)
• Temperature safety through color change when temperature is exceeded: > 7 °C : pink or
Quantitation and quality control
After extraction of RNA and DNA from samples such as tissues or cells, it is
important to have an idea of the quantity, quality and purity of the extracted
material. It can even be a good idea to input the same amount of RNA of each
sample into each cDNA synthesis reaction to minimize variation between
samples. UV-spectroscopic methods, such as the NanoDrop, use the absorbance
at 260nm to quantify the nucleic acids. Note though that absorbance at 260nm does not
discriminate between RNA and DNA and also does not discriminate between intact nucleic
acids and free nucleotides. One can also use the ratio of absorbances at 260/280 to assess
the amount of contaminating protein and/or phenol. UV-spectroscopic methods are widely
used because of their simplicity and that no additional reagents are required. The NanoDrop
also only utilizes 1-2µl of the sample. Fluorometric methods, such as the Qubit, can also
be used and different fluorescent dyes will discriminate between DNA and RNA. Also
fluorometric methods tend to be more sensitive.

The Fast Digest® enzymes are an advanced line of restriction enzymes for
rapid DNA digestion. One big advantage of these enzymes are that they
are 100% active in the universal Fast Digest® and the Fast Digest® Green
buffers and that they are able to digest DNA in 5-15 minutes. This enable any
combination of restriction enzymes to work simultaneously in one reaction
tube, eliminating the need of perfoming sequnential digestions.
Another improvment for your experiments are that the Fast Digest® Green
buffer allows for direct loading of the reaction mixture on a gel, no need
for extra loading buffer. The Fast Digest® Green buffer contains a density
reagent and two tracking dyes. The blue dye migrates with 3-5 kb DNA
fragments in 1% agarose gel and has an excitation peak at 424 nm while the
yellow dye migrates faster than 10 bp DNA fragments in 1% agarose gel and
has a excitation peak at 615 nm.
