Plainfield Palooza….a rocking evening under the stars
The Power of the Palooza! Don‟t know what a
“Palooza‟ is? Keep reading, so you, too, can be
enlightened.
On Saturday Sept 25 th , our ALS Walk Team, The
Specktaculars put the FUN back in Fun-draiser at
the Plainfield Palooza Music Festival for ALS. Way
out in the cornfields between Davis and Woodland
stands the Plainfield Station, a historical bar and
grill. It has an amusing history, but that story can
be told another day. Inside are handi-capable
restrooms, a quirky bar, and yummy for the
tummy food. Behind the actual building, is a fantastic,
inviting outdoor concert venue, with lush
lawn seating, picnic tables and a great stage.
Of course, wherever there‟s a stage, you can be
sure that I, Cathy Speck, will be the first one to
roll up the ramp, grab the microphone and start
talking, singing or honking. Making a spectacle of myself. Me me me.
Page 3
Yes, I am a ham (coincidentally, my last name “Speck” is German, and means bacon or ham.) Ham I am.
And this trait benefits ALS Awareness and FUN-draising. Why do I insist on emphasizing fun? I imagine
some people might shrink away when they hear of a benefit for ALS. Many folks have no idea what ALS
is—American Laughter Society? And many folks who have experienced the ALS journey, never want to
see/hear/feel about it again.
So, to draw crowds to our events, we need to let people know that raising funds for this disease can be
FUN. We don‟t have to focus on the dark side-the suffering of PALS and CALS and the inevitable death.
Our loves and lives are to be celebrated
Since ALSSAC and ALS research need money, and we all need to have fun, what can we do?! Hey kids,
let‟s put on a show! And that we did. The Plainfield Palooza Musical Festival raised over $1,000 for the
Walk to Defeat ALS. But more importantly, the event created an even stronger bond between people and
communities who want to help find the cause and cure of ALS . This is part of the Power of the Palooza.
This “love glue” (that sounds nasty but please don‟t take it that way) created an unending circle of the
eager, generous ,loving, helpers and performers who made it happen. This powerful circle consisted of
creators, producers, stage hands, sound engineers, promotional folks, writers, go-fers, local businesses,
performers, and the unending list of passionate supporters.
The Specktaculars and their supporters offered their talent, skill and energy with no payment except the
reward of knowing they are helping a serious cause while bringing joy and laughter to lighten the load.
Listing every single person who helped would make
my fingers fall off. So, I‟ll put out energy of gratefulness
into the wind and hope all helpers will feel that
tickle of this thankful breeze.
The Power of the Palooza? “Palooza” is just a funnysounding
word that is a spin-off of an old concert tour
called the Lollapalooza. I think you know by now what
the “power is.” I call it unending love. You can add to
that power by supporting the performers (and The
Plainfield Station0 as the circle goes „round and
„round; the Davis Wakamatsu Taiko Drummers, Sandy
Coldiron and the Free Range Band, Sabrina McFall, the
Gary Lee Yoder Band, Zapato Viejo, Duval Speck and
the Essentials, and magician Trey Cromwell.

Ask the Doc: Q & A with Edward Kasarskis, MD, PhD
Ask the Doc
Q: What would an emergency department physician need to know about me if I
have to go to there?
Dr. Kasarskis: Sometimes the diagnosis of ALS is so overwhelming in and of itself that
people forget that they’re still susceptible to everyday health problems and the occasional
emergency, ranging from “pink eye” and bladder infections to cuts, sprains, broken bones
and back problems. And depending on the severity of the problem (which always seems
to occur on weekends or after your physician’s office hours), an emergency room visit is a
distinct possibility.
Emergency room physicians and staff will first need to know that you are a person with ALS, because that diagnosis can
affect not just how your symptoms are approached, but what medications and treatments will be prescribed. If you
should need anesthesia, it is critical that the staff understands you have ALS. If you have any significant allergies, those
should be prominently noted, too. I urge everyone with ALS to wear a MedicAlert bracelet or locket to make sure that
this essential information is quickly communicated.
The staff needs to know what other conditions or diagnoses you have, such as diabetes, heart disease, chronic obstructive
pulmonary disease (COPD), high blood pressure, thyroid conditions, and others. They’ll also need to know your
medications, and the exact dosages. Please prepare a list of both your diagnoses and your current medications – and
stow it in your wallet – so it will be easily accessible in an emergency. Ideally you’ll also have a list of your physicians
with their address and phone numbers.
If you have communication challenges, it is important that the ER staff be aware of that. If possible, try to have a spouse,
friend, or neighbor come with you to the Emergency Department to assist you with communication. The ALS Association
has wallet cards available to describe your communication challenges to someone quickly and easily; the cards are also
handy if the police pull you over so they don't misinterpret any communication problems you may have.
Emergency room physicians may, in serious circumstances, need to understand your wishes about medical intervention.
It’s important to discuss this with a family member BEFORE such an emergency arises, and designate someone to serve
as a surrogate for medical decision making. This means this individual would have the authority to express your wishes
to the physician in case you’re unable to express yourself or are unconscious.
Sometimes it’s hard to decide whether a health problem necessitates a trip to the emergency room. There are particular
problems that should get immediate attention, and they include breathing problems, allergic reactions, a fever, symptoms
of a blood clot in a leg or an arm (deep vein thrombosis), or a sudden acceleration of the symptoms you experience with
ALS.
If you live alone or are sometimes alone in your home, (or have a spouse who can’t drive) it’s smart to have a preidentified
friend or neighbor to call in case you need to be taken to the emergency department.
Your question shows you’re already thinking ahead and planning for the unexpected. With ALS, the Boy Scout motto
always applies: Be Prepared!
In loving memory of the PALS we lost in September
Genome Study Identifies Chromosome 9 Link to ALS
In an ALS Association-funded study conducted in Finland, researchers found that a major cause of familial ALS was located on
chromosome 9p21. Finland is a well suited location for a genome-wide association (GWA) study of ALS because the incidence of
the ALS is one of the highest in the world, and the genetic background of the Finnish population is relatively homogenous. Previous
GWA studies have failed to identify a single location that is significant and can be replicated in other studies probably because
of the heterogeneity of the disease.
“Although GWA study has been a disappointing approach to find new genes linked to sporadic ALS, it is encouraging
to see the approach used successfully in a more homogeneous population,” said ALS Association Chief Scientist
Dr. Lucie Bruijn, Ph.D.
In the study published in Lancet Neurology, 853 DNA samples were collected from 442 people with ALS and 521 control individuals.
Control samples were from a population-based study of elderly Finnish individuals. People known to carry the SOD1 gene
were included in the final analysis as positive controls to assess whether this genome-wide association study was able to detect an
association signal.
Two signal peaks were identified in the population that reached significance: one peak corresponding to the known autosomal recessive
D90A mutation of the SOD1 gene and the second corresponding to chromosome 9p21.2 a location previously linked to
autosomal dominant ALS, where the gene is passed from just one parent.
The study therefore describes a major cause of familial ALS in the Finland located on chromosome 9p21. Furthermore, the overlap
with the risk haplotype (a combination of DNA sequences) recently also reported for frontotemporal dementia provides further
evidence of a shared genetic cause for these two neurodegenerative diseases. Although the investigators are currently unable
to identify the specific genetic variation underlying this locus, it will be feasible to identify patients with chromosome 9p21-linked
ALS by sequencing for this risk haplotype.
The research team was led by Bryan Traynor, a neurogeneticist with the National Institute on Aging and with the Johns Hopkins
School of Medicine. "We are very excited about this result because for the first time, we have been able to explain a large proportion
of ALS cases in a population, Taynor said. “We will continue to pursue this chromosome 9 locus to understand how it
gives rise to disease.”
In addition to The ALS Association, funding for this project came from National Institutes of Health and National Institute on Aging,
Microsoft Research, Helsinki University Central Hospital, Finnish Academy, Finnish Medical Society Duodecim, and Kuopio
University.
Advocacy and Public Policy— Sign-Up for the National ALS Registry
What causes ALS? How can it be treated? How many Americans actually have the disease? All of us who have been
touched by ALS have asked those questions at some point. And we've also heard the answers: that there are no
clear answers; that there is no treatment. But that may soon change because the federal government is set to fully
implement the National ALS Registry this October!
Beginning this fall, every person with ALS in the country will be able to self-enroll in the Registry and provide scientists
and doctors with the information they need to answer these and countless other questions that have gone unanswered
since ALS was first discovered as a disease more than 130 years ago. Indeed, for the first time, every person
with ALS will be counted. And this new data about ALS may help us learn what causes the disease, how it can be
treated or even prevented from occurring in the first place.
But in order for us to realize the promise of the Registry, we will need your help. Once self enrollment begins, we
need every person with ALS to enroll in the Registry! And we will need everyone in the ALS community to spread
the word about the Registry to people across the country and encourage them to enroll. The ALS Association will
provide tools to help you on our website, including instructions on how to enroll, fact sheets and answers to frequently
asked questions. Please visit the site regularly this month for the latest news on the Registry and when selfenrollment
will begin. http://webba.alsa.org/site/PageServer?pagename=BA_ALS_Registry
The ALS Association Greater Sacramento Chapter and advocates like you led the fight to enact the ALS Registry Act
and secure Congressional funding to build the Registry. And together, we can continue to create the Roadmap that
will lead to a treatment and cure for Lou Gehrig's Disease. By the Agency for Toxic Sustances and Disease Registry
Defeating Amyotrophic Lateral Sclerosis (ALS) is one step closer. Indeed, progress in recognizing and treating the
disease has come about because of team effort. And because of the teamwork of ALS patients and their advocates,
the National ALS Registry will soon be a reality.
In 2008, Congress charged the federal Agency for Toxic Substances and Disease Registry (ATSDR) with developing a
registry to gather and organize information about people living with ALS. The National ALS Registry represents a
ground-breaking step in the fight against ALS. This registry will give researchers data about who has the disease and
where it occurs. They can use registry data to detect disease pattern changes over time and to investigate possible
common risk factors among ALS patients.
This information can help researchers: Estimate the number of new cases of ALS identified each year; Estimate the
number of people who have ALS at a specific point in time; Examine the connection between ALS and other motor
neuron disorders that can be confused with ALS, misdiagnosed as ALS, and in some cases progress to ALS, and improve
care for people with this disease.
To ensure privacy, only certain researchers from ATSDR have access to the database. Individual patient data will not
be distributed, but researchers outside ATSDR can request compiled data.
Currently, there is no accurate picture of who has ALS. But the more people living with ALS participate in this registry,
the more accurate the picture will become. Beginning in fall 2010, people living with ALS will be able to add their
information to the registry. To learn more about the National ALS Registry, visit www.cdc.gov/als.
The ATSDR, which is implementing the National ALS Registry, is a federal agency of the U.S. Department of Health
and Human Services and sister agency to the Centers for Disease Control and Prevention (CDC).

Misfolded Mutant SOD1 Directly Inhibits Conductance
in Mouse Model
New research uncovers what may be a primary neuron-damaging
insult that occurs in an inherited form of the devastating neurodegenerative
disorder, amyotrophic lateral sclerosis (ALS). The mutant
superoxide dismutase (SOD1) protein is one cause of familial
ALS. Although mechanisms underlying mutant SOD1 neurotoxicity
remain uncertain, this mutant protein is associated with the
cell’s mitochondria, tiny intracellular energy-producing structures.
Investigators, lead by Professor Don W. Cleveland at the University
of California San Diego, report in the current issue of the
scientific journal Neuron that abnormally folded mutant SOD1
interacts with voltage-dependent anion channel (VDAC1), which
is located in the outer membrane of the mitochondria.
This finding sheds light into possible molecular links between mutant
SOD1 mitochondrial dysfunction and spinal motor neuron
degeneration of inherited ALS. This channel controls important
metabolic and energetic functions between the mitochondria and
the rest of the cell. Interestingly, this same channel has been implicated
in Parkinson’s disease where studies have shown that a
protein linked to the disease called Parkin interacts with this
channel.
“Our evidence demonstrates that reduced VDAC1 function and
correspondingly reduced mitochondrial function are direct components
of intracellular damage from mutant SOD1,” says Professor
Cleveland. “The finding that VDAC1 is a target for mutant
SOD1 within the nervous system provides important insight into
the mechanism underlying premature degeneration and death of
motor neurons.”
These studies were performed in the mutant SOD1 rats using
antibodies that specifically recognize the abnormally folded mutant
form of SOD1. Investigators were able to demonstrate a
reduction in conductance through this channel. The channel conductance
was not affected in liver mitochondria, and previous
studies have shown that abnormally folded mutant protein does
not accumulate in liver mitochondria. This suggests that this interaction
only occurs with the abnormally folded mutant protein
in the affected tissues in ALS. Chronic mitochondrial dysfunction
can lead to oxidative damage and neuronal degeneration.
“This study builds on earlier findings that mitochondria are critical
players in the disease mechanism; however, the new findings highlight
a novel mechanism and a potential new target for the development
of therapies” commented ALS Association Chief Scientist
Lucie Bruijn, Ph.D.
