system configurations, which reduces or eliminates a range of problems associated<br />

all become things of the past. ÄKTApurifier, together with the new ÄKTAdesign <br />

Intuitive system design<br />

from damaging events such as overpressure. Select a prepacked column from the<br />

extensive column library and ÄKTApurifier automatically sets the optimal running<br />

parameters for that column. New columns can be added to the library and will be<br />

run in the same secure way.<br />

Simplified system control with UNICORN<br />

software<br />

The system’s UNICORN software automatically provides method Wizards for<br />

simple method creation and controls and manages system hardware, saving you<br />

time and giving you better control of each purification run. Results are presented in<br />

Advanced pumps ensure reliable results<br />

The main challenges of a chromatography system pump are to achieve accurate<br />

gradient formation over a broad flow rate range and avoid introducing air into the<br />

system. ÄKTApurifier 10 and UPC 10 systems are equipped with Pump P-901, which<br />

enables a wide flow rate range for both laboratory- and pilot-scale applications.<br />

ÄKTApurifier 100 and UPC 100 systems use Pump P-903, which operates at the<br />

lower flow rate and the higher pressure required for running high-resolution<br />

analyses at laboratory scale.<br />

Visualize more protein<br />

ÄKTApurifier 10 and 100 core systems are equipped with Monitor UV-900,<br />

which allows multiple UV-wavelength monitoring of up to three wavelengths<br />

simultaneously. Now you can visualize more protein when adequate detection<br />

cannot be achieved at 280 nm. The novel xenon flash lamp of Monitor UV-900<br />

ensures long lamp lifetime and that no heating of the sample in the flow cell occurs.<br />

ÄKTApurifier UPC 10 and 100 core systems each offer convenient UV-wavelength<br />

monitoring, pH, and conductivity measurement at the same time using Monitor<br />

UPC-900. This monitor gives greater flexibility and provides an economical<br />

alternative to Monitor UV-900.<br />

ÄKTAdesign automation kits<br />

—spend less time at the lab bench<br />

ÄKTApurifier systems eliminate much of the tedious and time-consuming<br />

manual steps involved in sample application by bringing a greater degree of<br />

automation to the system’s setup. Optional ÄKTAdesign automation kits and<br />

hardware are available to add functionality to your ÄKTApurifier core systems<br />

and make sure you get the most from your protein purification.<br />

Examples of additional functionality that can be applied to ÄKTApurifier core systems using<br />

ÄKTAdesign automation kits.<br />

Keep your system and buffers free from air<br />

Air in an expensive gel filtration column can be devastating, ruining the sample run.<br />

Air usually enters the system when the sample is completely applied to a column<br />

and the sample reservoir is empty. Replacing or replenishing depleted stocks<br />

of buffer is time-consuming and makes overnight, unmanned purification runs<br />

difficult. In a worst-case scenario, air in the system resulting from depleted buffer<br />

stocks can lead to irreversible column damage.<br />

Air sensor Kit contains all you need to exclude air from ÄKTApurifier during<br />

purification. Air sensor Kit, which is controlled by UNICORN software, detects air<br />

in the system, and pauses the purification run. The kit therefore prevents column<br />

damage caused by air and secures that the sample is completely injected.<br />

Buffer selection Kit manages buffer reagents effectively, allowing buffer screening<br />

and minimizing the risk of air entering the system. The kit enables automatic<br />

selection among up to eight different buffers to allow optimized purification. Buffer<br />

selection Kit eliminates manual changing of buffers and moving inlet tubing,<br />

ensuring that no air enters the system.


Speed up buffer preparation with BufferPrep<br />

The accuracy of pH is crucial in many separations. BufferPrep Kit, which is optional<br />

for ÄKTApurifier systems, eliminates time-consuming manual buffer preparation<br />

and titration usually needed for pH changes in chromatography. For any pH and<br />

salt concentration entered, BufferPrep automatically calculates the composition of<br />

the buffer online. The accuracy of BufferPrep greatly increases the reproducibility<br />

of results from day to day by eliminating and correcting errors that arise when<br />

mixing buffers manually. BufferPrep enables automatic buffer titration and<br />

eliminates errors occurring during manual preparation.<br />

The BufferPrep function can also be used for pH scouting in rapid method<br />

optimization, all under the control of UNICORN software.<br />

Reliable sample application<br />

Sample application presents a number of significant challenges in protein<br />

purification. Loading samples is time-consuming and managing setups for<br />

different sample volumes can be difficult. Viscous samples provide an additional<br />

risk of system overpressurization and inadequate sample loading can lead to air<br />

inadvertently entering the system.<br />

Increased automation using the optional Sample application Kit can eliminate<br />

many of the problems associated with sample loading. Sample application Kit<br />

gives you the versatility of loading small, medium, or large sample volumes directly<br />

to the column or via capillary or Superloop. When speed is an issue, the pump<br />

supplied in the kit can be modified for automatic, multiple sample injection.<br />

Convenient media and column screening<br />

Finding the best column for your gel filtration, optimal pH for your ion exchange<br />

purification, or right binding and elution buffer concentration for your histidinetagged<br />

protein purification is laborious, especially when performed manually. Or<br />

perhaps you are unsure as to what column to use?<br />

Column screening Kit provides the necessary motor valves to allow automated<br />

method optimization, multistep purification, and easier column selection. Up to<br />

eight columns can be connected to the system in one go. Column performance can<br />

be easily compared while changing conditions in a run. Preset protocols prevent<br />

system overpressurization and prevent column breakage during use.<br />

Connecting two columns to one of the motor valves included in the kit enables<br />

automated column selection for optimizing both column and media performance,<br />

as well as column switching for two-step purification protocols.<br />

Reversing flow speeds up affinity<br />

chromatography purification<br />

If the full capacity of affinity columns is not used, dilution may occur during<br />

dilution of the target protein, which increases elution time and broadens peaks. To<br />

combat this, elution is preferentially performed in the reverse direction to sample<br />

application.<br />

Reverse flow Kit contains motor valves for automated reverse flow through a<br />

column. Strongly bound target proteins elute more effectively, speeding up your<br />

purification. Resulting peaks are sharper and more concentrated target protein is<br />

recovered. The kit increases the life expectancy of your columns since reversing the<br />

flow efficiently rinses impurity away from the top of the column.<br />

Collect any volume of purified product<br />

Large sample volumes inevitably lead to large elution volumes. If your protein of<br />

interest elutes in a large volume, collecting in small sample tubes becomes timeconsuming<br />

and prone to error.<br />

Fractionation Kit enables collection of large volumes of purified sample. Up to<br />

eight vessels of any size can be used to collect fractions. Collection in large vessels<br />

rather than smaller vessels minimizes the risk of losing valuable target protein,<br />

improving protein recovery and giving you greater piece-of-mind.


Simplify FPLC purification even<br />

further with ÄKTA accessories<br />

In addition to the ÄKTAdesign automation kits, a wide range of<br />

hardware accessories is available to simplify protein purification<br />

with ÄKTApurifier systems.<br />

Flexible fraction collection<br />

Automated fraction collection speeds up fraction collection as well as minimizes<br />

peak dilution and cross-contamination.<br />

Fraction Collector Frac-950 is specifically designed for collection in 96-well<br />

microplates, as well as 12-, 18-, and 30-mm diameter tubes. Fraction Collector<br />

Frac-920 allows collection of up to 175 fractions in 12-mm diameter tubes, up to 95<br />

fractions in 10–18 mm tubes, as well as 40 fractions in 30-mm diameter tubes. Both<br />

fraction collectors are available as options for ÄKTApurifier core systems.<br />

Monitor pH/C-900<br />

Monitor pH/C-900 expands ÄKTApurifier 10 and 100 systems to allow measurement<br />

of conductivity, pH, and temperature. Since temperature can influence both pH<br />

and conductivity, the monitor compensates for temperature variations of these<br />

readings. Self-diagnostics ensure continual reliability and reproducibility. Both pH<br />

and conductivity/temperature flow cells feature low cell volume and high linearity<br />

for accurate measurements.<br />

Faster sample injection using autosamplers<br />

Autosamplers give you more time to work on other projects during the day, and<br />

increase throughput by enabling overnight purification runs.<br />

Autosampler A-900 is available with Peltier cooling, which effectively protects<br />

heat-labile proteins during sampling. The autosampler is used with 7 or 12 mm<br />

outer diameter vials. Autosampler A-905 is designed for sample injection from<br />

microplates, as well as microvials, with Peltier cooling as standard.<br />

Add additional instruments easily<br />

AD-900 Converter is an analog/digital converter for connecting external<br />

instruments to UNICORN control software. The AD-900 Converter allows easier<br />

monitoring of signals received as voltage inputs from external sources such as<br />

fluorescence detectors, radioactivity monitors, or diode arrays.<br />
