Result: The protein is eluted early in the gradient. Resolution is
satisfactory.
Discussion: This can be a good choice of medium. The fact that the
protein is eluted early in the gradient indicates that the binding
capacity might be low. An increase in salt concentration to
compensate for this may lead to a decrease in selectivity since
some of the unbound proteins might be adsorbed together with
the protein of interest. Not much can be gained by changing the
pH of the start buffer since the resolution was considered
satisfactory.
Next step: If low capacity is a problem and a moderate increase in
salt concentration leads to loss in resolution, try a medium with a
different ligand or a medium with a higher degree of ligand
substitution.
