Derivative spectroscopymay be used to enhance fine detail, reduce the effectsof a sloping background and accurately locate a maskedband for measurement.Derivatives may be displayed alone or superimposedon the fundamental, with or without an offset.Band QuantificationFor accurate quantitative work it is often required todetermine the area or height of an absorption bandcorrected for background absorption.A very easy to use programme is provided for thispurpose. The analytical result is displayed on thescreen, with construction lines, enabling the quality ofthe chosen parameters to be assessed.Band quantification may be stored as a methodsecurity protected.Multi-Cell KineticsThe Super Aurius Series offers multi-cell Kinetics aswell as single cell Kinetics as available on the AuriusSeries. FAST DATA TRANSFER TO PCPERFORMANCE VALIDATION : GLPPerformance ValidationUNIVERSAL ACCESSORY RANGEInstrument performance may be validated using acomprehensive software module available on allinstruments and which may be released by E-SEF.Tests are provided for laboratories subject toregulatory control or following G.L.P. These include arange of Pharmacopoeia tests (EP/BP/USP), a selectionof which are shown here.Absorbance ValidationThe Pharmacopoeia (EP/BP) test for absorbanceaccuracy is one of the tests provided. The testmeasures the values of peaks and valleys in theabsorbance spectrum of a 60mg/L solution ofPotassium Dichromate in a 10mm pathlength cuvette.The results of such a test run are shown here.Optical BandwidthThe very accurate method for validating opticalbandwidth uses the narrow deuterium emission line at656.1nm. The width, at half height, of the measuredband due to the emission line measures the actualbandwidth. The band is drawn and the half widthautomatically documented.uriusStraylightStraylight may be measured using the Pharmacopoeia(EP/BP) test shown here. This test uses a 1.2% W/Vsolution of Potassium Chloride in water, measuredagainst water in the reference beam. An absorbance inexcess of 2A must be measured at a wavelength of200nm.The ASTM tests at 220nm and 340nm are more widelyused and are provided.Validation Using FiltersAbsorbance and Wavelength validation are mostconveniently carried out using filter sets calibrated by<strong>Cecil</strong> Instruments on its reference spectrophotometeragainst standards certified by the NPL.
