2.2 Experimental<br />
2.2.1 Materials<br />
Octadecyltrichlorosilane (98%) and chlorotrimethylsilane (97%) were purchased from<br />
Aldrich and used as received. Silanated PEG2000 is synthesized according to literature<br />
procedures.[3, 4] Poly(ethylene glycol) methyl ether (m-PEG, average Mn ca. 2000) and<br />
dibutyltin dilaurate were purchased from Aldrich. 3-Isocyanatopropyltriethoxysilane (IPTS) was<br />
purchased from Gelest. 6-Propionyl-2-dimethylaminonaphthalene (Prodan) was purchased from<br />
<strong>Molecular</strong> Probes.<br />
2.2.2 Instrumentation<br />
A 400 MHz NMR spectrometer (Unity 400, Varian) was used to collect the 1 H-NMR<br />
spectrum of the silanated PEG 2000. IR spectrum (512 scans at 4 cm -1 resolution) of the<br />
silanated PEG 2000 was collected using a Nexus 670 FT-IR E.S.P. (Thermo Nicolet). The<br />
molecular weight and polydispersity of the silanated PEG 2000 were tested using Waters 2410<br />
GPC (RI detector, three Waters Styragel columns HT2 + HT3 + HT4, polystyrene as standard<br />
and THF as eluant at a flow rate of 1.0 mL/min). A Gaertner Ellipsometer Model L116C<br />
(Gaertner Scientific Corp.) was used to measure the thickness of the SAMs. Contact angle data<br />
was collected using a Ramé-Hart Goniometer (Model 100-00). A spincoater (Speedline<br />
Technologies, Specialty Coating Systems, Inc., Model P6204) was used to deposit Prodan<br />
solution onto the SAM modified surfaces. A Leica confocal laser scanning microscope (Model<br />
TS SP2) equipped with Ti:Sapphire tunable wavelength laser (Tsunami, Spectra-Physics) was<br />
used to collect fluorescence recovery after photobleaching (FRAP) images, and samples were<br />
