Liquid sample bags<br />
Whirl-Pak ® . Made of LDPE. Sterile. Thickness 57 µm.<br />
Ideal for storing liquid and solid samples.<br />
Procedure:<br />
1. Whirl Paks ® can be opened easily by simply tearing the plastic along the<br />
perforation. The bag interior is therefore protected against contamination,<br />
as long as the bag is not used.<br />
2. Practical pull-open loops enable easy opening of bag,<br />
without contact to the interior. The bag remains open when filling.<br />
3. The wide-neck opening of the bag makes filling easier.<br />
Hold the bag as indicated and fill approx. 2/3 full.<br />
4. The bags are closed by pulling on the outer loops.<br />
5. Hold the bag at a distance and spin it at least 3 times.<br />
6. The bags are sealed tightly by bending the loops.<br />

Whirl-Pak ® . Made of LDPE, sponge and bag. Sterile. Speci-Sponge ® for<br />
sample taking from <strong>lab</strong>oratory benches, furniture, equipment and all other<br />
areas which must be tested for listeria, salmonella, E.coli and other<br />
pathogens. Sponge and bags are sterile. Sponge is free from bacteria and<br />
not growth-inhibiting. After "wiping" the sample, return the sponge to the<br />
bag. Add medium and if required incubate directly in the bag.<br />

SOB<br />
Super Optimal Broth (SOB) is a complex, nutritious nutrient medium which is used prior<br />
to and after transformation of E. coli bacteria. By adding 20 mM glucose it is enriched to<br />
SOC (Super Optimal Broth with Catabolite repression) which is optimal for coddling up of<br />
freshly transformed bacteria. Through this, SOC increases transformation efficiency.<br />
In order to avoid caramelisation, glucose may only be added after autoclaving.<br />

Roti ® -Transform is a simple, effective and fast method of producing<br />
competent E. coli in three steps. The competent cells may be used<br />
directly for transformation or may be frozen for later transformations.<br />
Transformation efficiency is 5 x 10 5 to 1 x 10 6 colonies per µg DNA.<br />
Long-term Storage of the Kit at -20 °C.<br />
Storage for use:<br />
Reagent 1: Short-term storage may be done at 4 °C. For long-term storage<br />
of over 14 days we recommend a storage temperature of -20 °C.<br />
Reagent 2: Storage at 4 °C.<br />
Roti ® -Transform consists of 45 ml Roti ® -Transform 1 (Art. No. P041) and<br />
5 ml Roti ® -Transform 2 (Art. No. P042). One kit is sufficient for producing<br />
competent E. coli for 500 transformations.<br />

Replicator stamp<br />
Replicator stamp for transfer of bacterial or yeast colonies from one culture<br />
medium plate to another. Stamp with fixationring in yellow or silver coloured.<br />
12 autoclavable transfer tissues are included. Transfer tissues may also be<br />
bought separately in colours blue, red, and beige (for brightly coloured<br />
colonies).<br />
In order to duplicate the colony plates, fix one of the sterile tissues on the<br />
stamp with the aluminium ring (see figure). Then press the top side of the<br />
original colony plate gently onto the stamp. For cloning press a fresh plate<br />
onto the inoculated tissue surface.<br />
Applicable for petri dishes of 90 to 100 mm diameter. The replicator stamp<br />
is made of PVC. Height: 72 mm, diameter: 81 mm. Fixation rings are made<br />
from coated aluminium. Transfer tissues are made from an autoclavable<br />
cotton-polyester mix. The stamps can be disinfected with ethanol<br />

The History of Antibiotics<br />
In the history of mankind there have always been highly developed cultures possessing a remarkable knowledge of things. For example, surgeons in the<br />
ancient world and in the Middle Ages placed mouldy rags on wounds to prevent infection. First observations of antibiotic effects in modern times were made<br />
by the French chemist, Louis Pasteur, in the 19th century. Pasteur discovered that certain bacteria could kill anthrax germs. The German bacteriologist,<br />
Rudolf von Emmerich, isolated the antibiotic reagent pyocyan (cholera and diptheria) in around 1900 and in 1910 experiments carried out by the German<br />
doctor and chemist, Paul Ehrlich, led to the development of salvarsan (syphillis and tropical diseases). Pencillin was discovered by chance in the mould<br />
fungus cultures of Penicillium notatum in 1928 by Alexander Fleming when he observed that P. notatum inhibited the growth of vicinal bacterial colonies in<br />
<strong>lab</strong>oratory cultures. Streptomycin followed in 1943. The first antibiotics were isolates from bacterial cultures. The preparations used today are, however,<br />
almost exclusively of chemical origin thus enabling specialisation and optimisation.<br />

GELRITE ®<br />
Highly-purified, natural heteropolysaccharide which forms stable,<br />
agar-like gels when exposed to soluble salts. Compared to agar however,<br />
only about half the amount is required. Linear polysaccharide comprising<br />
of O-acetylate glycuronic acid, glucose and rhamnose units. When using<br />
very high salt concentrations like e.g. seawater, gelling capacity may be<br />

Agar types<br />
The designation Agar Kobe I derives from the geographical origin of the agar, which is<br />
the Japanese city of Kobe on the main island of Honshu. In former times, when agar was<br />
still traded in strips, threads or squares, agar was graded into three main quality groups:<br />

- top quality, pure white<br />
- slightly inferior quality, slightly yellowish to light beige<br />
- inferior quality, yellow-beige<br />
These terms and the associated differences in quality are now obsolete - for some time<br />
now, agar of consistent quality has been avai<strong>lab</strong>le from all parts of Asia. However,<br />
for reasons unknown, the designation Kobe I is used today to describe ground agar<br />
of very high quality which is used as a microbiological standard agar.<br />
In addition, the designation bacteriological agar has become an established definition<br />
for ultra-pure agar, which represents a preliminary stage of purified agarose and is<br />
manufactured for special microbiological applications.<br />
Danish agar, also known as Furcelleran, is a saccharide-enriched carrageenan<br />
extracted from the red algae Furcellaria fastgiata.
