Historically, the high surface expression of CD25, in association with the intracellular expression of the transcription factor Foxp3, has been considered one of the main phenotypic features of T-REGs. Recent data showed an unexpected expansion of Foxp3+ CD4+ T cells lacking of surface CD25 in the peripheral blood of patients with systemic lupus erithematosus (SLE). Thus, other surface molecules are needed to further characterize T-REGs. GITR is a well established marker of murine T-REGs, but little is known about its role in humans (1-3). Object: Phenotypic and functional characterization of T-REG cells in healthy donors (HD) and SLE patients. We aimed to assess the possible role of GITR as a marker of human peripheral blood T-REGs Methods: The study included 31 SLE patients and 30 sex- and age-matched HD. CD4+ T cells were negatively selected by magnetic cell sorting (CD4+ T Cell Isolation KIT II, Miltenyi). Positive selection strategies were also performed in order to isolate CD25high and GITRhigh subsets. T-REGs phenotype was investigated by both flow cytometry and qRT-PCR. Primary and secondary antibodies, provided by BD-Pharmingen and Biolegend, as well as TaqMan probes (Applied Biosystems) were used. Proliferation assays were performed on CFSE-labeled anti-CD3-triggered cells. Results: Results obtained by flow cytometry in HD, CD4+ T cells showed a percentage of CD25highGITR- and CD25high- GITRhigh subsets of 15±3.7% and 0.7±0.2%, respectively. A small CD25-GITRhigh cell population (0.7±0.3% of CD4+ cells) was also detected. Phenotypic analysis at mRNA level and proliferation assays demonstrated that the 3 cell subsets were suppressive T cells.
