Peroxynitrite (O=N—O—O - ) is generated by nitric oxide’s reaction with superoxide anion in<br />
cell and organelles. It is a powerful oxidizing and nitrating agent, and his presence was correlated with<br />
the pathogenesis of many degenerative conditions such as Alzeimer’s, Parkinson’s, cancer or AIDS.<br />
Currently, peroxynityrite is detected by chemiluminescence, fluorescence, LC-MS or with<br />
electrochemically modified carbon microfibers<br />
This talk outlines efforts to fabricate and test metallo-porphirin-modified carbon fiber<br />
microelectrodes (CFEs) for peroxynitrite’s quantification. The analyte peroxynitrite was synthesized in<br />
house fresh every time, by the two-phase displacement reaction. The investigations included: field<br />
emission scanning electron microscopy (FESEM) characterization, cyclic voltammetry and time based<br />
amperometry. The sensitivity, detection limit, response time and selectivity are characterized.<br />
Moreover, the electroactive polymer poly(3,4-ethylenedioxythiophene) or PEDOT was<br />
employed to improve both the detection limit and sensitivity. Indeed, this talk will show that the<br />
sensitivity for the hemin-PEDOT modified CFE was 13 pA/nM. That is 130 times higher than bare<br />
CFE, or 50 times higher than hemin-modified CFE. Also, the detection limit for the hemin-PEDOT<br />
CFE was 200 nM for a 3/1 response/noise ratio.<br />
